The Sysmex XE-5000 Automated Hematology Systemutilizes the power of fluorescent flow cytometry andhydrodynamic focusing technologies. Using a unique,state-of-the-art, diode laser bench, Sysmex fluorescentflow cytometry provides the sensitivity needed formeasuring and differentiating cell types in whole bloodand body fluid samples. Fluorescent technology andhydrodynamic focusing enable the XE-5000 toconsistently classify normal WBC, RBC and PLTpopulations from abnormal populations, therebydecreasing the number of manual interventions.

Enumeration of NRBC is critical toproviding an accurate WBC anddifferential in pathological samples.The XE-5000 fluorescent technologyprovides a NRBC in both % and # froma dedicated channel.

The Sysmex XE-5000 offers a 6-partdifferential, which includes an ImmatureGranulocyte Count (IG%, #). The IGcount offers reportable, quantitativeresults for immature granulocytes(metamyelocytes, myelocytes andpromyelocytes).

Fluorescent Reticulocyte Count

Known as the “Gold Standard” in reticulocyte testing, the fluorescentreticulocyte count is available on the XE-5000. Sysmex provides on-boardretic testing in a dedicated channel, improving your efficiency in reportingreticulocytes 24 hours per day.

With the use of fluorescent technology, the retic channel assures:

• Accurate reticulocyte % and #• Improved immature reticulocyte information for earlier diagnosis

and treatment by clinicians• Elimination of common interferences from Howell-Jolly bodies,

Pappenheimer bodies and immature reticulocytes to avoidmanual counts

Reticulocyte HemoglobinMeasurement (RET-He)

The Reticulocyte Hemoglobin (RET- He) is a parametermeasured in the reticulocyte channel and is used to measure theincorporation of iron into erythrocyte hemoglobin.

The RET-He parameter supports:

• Rapid, direct analysis of an earlier stage of RBC development forprompt clinical follow-up

• Assessment of anemia and is an established parameter used inKDOQI (Kidney Disease Outcomes Quality Initiative) guidelinesfor assessing the initial iron status of patients

Fluorescent Reticulocyte Count

Fluorescent Optical Platelet

The XE-5000 offers a fluorescentoptical platelet along with thetraditional impedance platelet count.These dual technologies maximizeinstrument output. Flagging associatedwith atypical or abnormal platelets,due to increased size or fragmentation,is minimized by use of the opticalplatelet count. The accuracy inreporting is supported by theavailability of both technologies.

Fluorescent Optical Platelet (PLT-O) isa complementary, reportable parameterto the impedance platelet count.

The PLT-O parameter provides:

• Improved accuracy on lowplatelet counts

• Accurate counts when interferencesare present, thus reducingmanual intervention

The Sysmex XE-5000 offers aquantitative hematopoietic progenitor(HPC) cell count as a screen for themeasurement of the optimal time forcell harvest. The Sysmex automatedHPC count is substantially equivalentto CFU and CD34+.

The HPC count from peripheral bloodand cord blood samples supports:
