Current area of research

Molecular characterization of efficient isolates of biocontrol agents and development of their biopesticides for the management fungal/nematode diseases and insect pests of agricultural crops

Molecular characterization and evaluation for efficacy of indigenous strains/isolates of various parasites of

phytonematodes (Aspergillus niger, Paecilomyces lilacinus, Pochonia chalmydosporia, Cylindrocarpon destructans,

Arthrobotrys oligospora, Dactylaria candida and Bacillus thuringiensis), mycoparasites of soil-borne fungal pathogens

(Trichoderma harzianum, T. hamatum, T. virens, T. viride, T. koningii, T. polysporum etc) and phosphate solubilising

microorganisms ( A. niger, A. awamori, Penicillium digitatum, Azotobacter chroococcum, Azospirillium lipoferum, Bacillus

subtilis, B. polymyxa, Beijerinkia indica, Pseudomonas fluorescens, P. striata, A. stutzeri etc.) under pot and field condition

have revealed that A. niger, P. chlamydosporia, T. harzianum or T. virens and P. flourescens can control root-knot, root-rot

and wilt diseases of vegetables and pulses. Biopesticides of these five microorganisms have been prepared (patents

granted in the USA and India). Trials of these biopesticides conducted in experimental and farmer’s fields in different

villages in the districts Aligarh, Muradabad, Budaun and Barielly under a DBT research project have shown that seed

treatment with the biopesticides @ 2 g/kg seed significantly controlled wilt, root-knot and wilt disease complex leading to

yield enhancement of 3-5 q, 2-3 q and 6-8 q/ha in pulses. A control of 42% in the damping off of legumes has been

recorded from trials of the biopesticides conducted in UAS, Bangalore under a DBT evaluation program. In addition, local

isolates of entomopathogenic nematodes (EPN) have been isolated and mass cultured to control insect pests and plant

parasitic nematodes. Preliminary studies have shown that seed treatment with Ag, Ti and Ni nanoparticles can suppress

the soil borne pathogens without causing any toxicity to the plants.

Effects of environmental contamination on crop productivity and disease development

Another major area of research is an evaluation of effects of environmental contamination and climatic change on crop

productivity and plant diseases. Two major projects have been sanctioned by DST in this research area. The researches

have revealed that SO2, O3, NH3, coal smoke, acidic rain and heavy metals at low concentrations did not injure plants, but

predisposed them to greater pathogenic damage. Sulfur dioxide at this concentration usually occurs in India within an

area of 2-6 km from coal-fired power plants; consequently the crops grown around power plants or other emission sources

may exhibit additional damage from pathogen(s). This observation has necessitated re-evaluation of existing air quality

standards for rural and agricultural areas. His ongoing DST project on climatic change has shown that 400-500 ppm CO2

can promote plant diseases.

Utilization of fly ash in crop production and disease management

Fly ash which is a black solid waste of coal fired thermal power plants, can supplement inorganic fertilizers, as it contains

exceedingly higher amounts of plant nutrients P, K, B, Si, Ca, Mg, Mn, Zn etc. Exploration of growth promoting effect of

fly ash on crop plants, and optimization and standardization of economic doses and mode of application have shown

that broadcast or row application of fly ash in soil at 3-4 q/ha can increase the yield of tomato, eggplant, okra, chickpea,

mung bean, pigeonpea and winter ornamental plants by 40-80%. A seed/root-dip treatment with phosphate solubilizing

microorganisms may further increase the availability of phosphorus to plants and subsequently greater yield enhancement.

If it is practiced commercially, the cost of farm-input may be reduced considerably, because fly ash is available at a very

low cost and its growth promoting effects can persist in soil for 1-3 years. Additionally, the money spent on pesticides

would also be saved/minimized, as fly ash application suppressed soil-borne pathogens.

Details of the biotechnology developed and its commercialization A novel process to produce biopesticides based on fungi and bacteria has been invented. This process involves development of stock culture of the biocontrol agents on a mixture of sawdust- soil- 5% molasses (15: 5: 1). One part of the stock culture is mixed with the 20 parts of carrier (flyash- soil and molasses in the ratio of 15: 3: 1 ratio). The CFU load and shelf life of the formulations were as follows:

Biopesticide CFU’s/g soil Shelf life

Biowilt-X (Trichoderma harzianum) 1010 CFUs 4-19 weeks Bionem-X (Pochonia chlamydosporia) 109 CFUs 4-15 weeks Biocomp-X (Pseudomonas fluorescens) 1013 CFUs 2-22 weeks Biocure-X (Bacillus subtilis) 1013 CFUs 2-21 weeks

The production figures of the above products and cost benefit ration of application of the biopesticides against fungal and nematode diseases on pulses and other crops is summarized as under.

1. A very low cost materials viz., sawdust, fly ash, soil and molasses which are available locally in India are

used to produce the commercial formulations (biopesticides) to control fungal and nematode diseases of crops. As a result final cost of the formulations is estimated and realized as Rs. 10/200 g which is much less than the cost of other biopesticides currently available in market (Rs. 80-90/200 g ).

2. The four biopesticides namely, Biowilt-X (Trichoderma harzianum), Bionem-X (Pochonia chlamydosporia)

Biocomp-X (Pseudomonas fluorescens) and Biocure-X (Bacillus subtilis) developed are superior than their

contemporaries available in the market with regard to effectiveness and cost. The formulations have a

CFU load in the range of 107-9 (biocontrol fungi) and 1012-13 (biocontrol bacteria), which is greater than other

biopesticides.

3. The bioagents applied through the commercial formulations established in the soil, and conferred a longer

crop protection.

4. Application of Biowilt-X and Bionem-X biopesticides @ 2g/kg seed satisfactorily managed fusarial wilt and

root-knot diseases of chickpea and pigeonpea, respectively, and gave a profit of Rs 6220-8140/ha against

wilt and Rs 3180-4180/ha against root-knot.

5. Treatment with Biocomp-X (Pseudomonas fluorescens) @ 2g/kg seed effectively controlled the fusarial wilt,

root-knot and wilt disease complex of chickpea and pigeonpea with a profit of Rs 3460-11980/ha depending

on the disease and crop as shown in the following table.

Biopesticide @ 2g/kg seed

Various national and international biopesticide manufacturing companies have shown interest in the technology that we have developed to prepare fungal and bacterial biopesticides because this is a very low cost technology and can drastically decrease the cost of biopesticides (Rs. 10/200g packet). Since patents have been filed in India and granted in USA, we shall now grant permit of using our technology to the following companies or some other which have been interested in our process and products.
