Melanogenesis is a complex biochemical process involved in the production of skin pigments that determine skin color.These pigments mission is to protect the skin from damage caused by the solar radiations.Melanogenesis depends on di�erent exogenous and endogenous factors which leads to an over production of melanin in localized areas provoking the appear-ance of unattractive blemishes. The major cause is exposure to UV radiations, but there are other factors involved such as hormonal and genetic factors, acne marks, race, pregnancy, menopause, hypo�siary disorders, ingestion of photosensitizing medication, de�ciency of vitamin D…etc.Melanocytes are dendritic cells located in the epidermal basement membrane, responsible for the melanin synthesis. These cells secrete melanosomes, which are melanin granules that are transferred through dendrites (cell prolongations) to the keratinocytes, causing the pigmentation of this.The melanogenesis process, also known as melanin synthesis, begins with the tyrosinase enzyme action that produces the tyrosine oxidation, transforming it into dopa (dihydroxyphenylalanine). Next step is also catalyzed by the tyrosinase that produces dopaquinone by dopa dehydrogenation. After di�erent chemical reac-tions dopaquinone will generate two di�erent types of melanin:

EUMELANINS:Pigmentations with black or brown colour. They create dark colouring.

PHEOMELANINS:Pigmentations with red or yellow colour. They create light colouring.

Due to the UV radiations stimulus, the skin melanocytes transfer melanosomes to the keratynocites located in germinative layers of epidermis, that become pigmented storing melanin. Melanosomes are degraded when ascending at di�erent epidermal layers to get the corneous stratum. The cell renewal process through desquamation means the natural channel to eliminate melanin.Melanin acts as a real protective barrier, forming a photoprotective cap over the nucleus of epithelial cells protecting skin cells from DNA damage caused by UV radiations.
