könyv a molekuláris biológiáról (1966)
Bármilyen alakban létezzék is, mi annak princípiuma?
Diderot
A legegyszerűbb sejtek bakteriálisak
• Valamilyen primitív baktériumszerű sejt eredetét kívánjuk megmagyarázni
• Csakhogy ezek a sejtek is nagyon összeszetettek! • A genetikai kód egy „notóriusan nehéz probléma”
A Princípium első kiadása • Elképesztően jelentős
munka ismeretterjesztő álarcban
• Csak ÍGY lehetett publikálni
• Tartalmazta a „chemoton” koncepciót
• Dühödt reakciókat váltott ki
A könyv szerkezete
• A probléma • Az új adatok • Az új szemlélet
• Szakítás a molekuláris biológia egyeduralkodó szemléletével
• Az életkritériumok • Az élő minimálrendszer
A legfontosabb kiadás: Oxford University Press, 2003
• Sok magyar kiadás után
• Tartalmazza a Princípiumot és a Contra Cricket is.
• Értékelő jegyzetek és tanulmányok
The investigation of life criteria: absolute criteria
1. Inherent unity 2. Metabolism 3. Inherent stability 4. Information carrying subsystem 5. Processes regulated and controlled by a
programme VERBAL AND PHENOMENOLOGICAL
Quest for a biological minimal system
• Chemical supersystem • Should be conceptually as simple as
possible • Must not necessarily be realizable in its
simplest form • Comparison with other elementary units
(such as the elementary cell in crystallography)
• FORMAL AND EXACT

Anyagcsere
The chemoton shows „organizational closure”
• It is „autopoietic”, creating and maintaining its boundary conditions
• It is deeply routed in chemistry • Yet presents an abstract and idealized model • NO ENZYMES in the basic model
Chemical cycles are metabolic engines (1971)
• The cycle as a whole is a catalyst
• The Noble prize of Szent-Györgyi was awarded for catalysis by fumaric acid
• Krebs has recognized the whole cycle
• Enzymes are superimposed on the metabolic cycle
Is metabolism autocatalytic in contemporary organisms?
• This is at the root of metabolic organization • Is also important for heredity • An old question analyzed by modern
bioinformatics

INFRABIOLÓGIAI RENDSZEREK
COST Systems Chemistry
• CM0703 • The main objective of the Action is to
investigate autocatalytic reaction sytems within supramolecular, prebiotic, and other fields of chemistry and to develop methods for their integration into dynamic supersystems.
Egy infrabiológiai rendszer(1978)
• Anyagcsere és térbeli reprodukció • Nincs genetikai alrendszer: önreprodukáló
mikrogömb
Primitive ancestry of the reverse citric acid cycle?
• Was proposed by Günter Wächtershäuser (1990)
• Coupled to CO2 fixation and pyrite formation around deep-sea hydrothermal vents

Rosen’s catalytic closure and his strange terminology
• “Repair” replacement • “Replication” closure • A represents the set of metabolites
that are converted by metabolism into a second set B, catalysed by a set of enzymes f that are themselves products of metabolism.
• The catalysts needed for the replacement process is provided by a set of catalysts Ф that are produced from f.
• Closure is achieved by supposing that the efficient cause of Ф is a property of B (not B itself), represented as β.
Material causation (chemical transformation)
Efficient causation (catalysis)
What could this mean?
food
metabolites
housekeeping enzymes
genes and informational enzymes
Reflexively autocatalytic
Hmmm?
“Biblical scholarship”
Critical features
• Structural closure Varela, Gánti • Catalytic closure Rosen, Kauffman, Gánti,
Varela • Spontaneous appear. Kauffman? Varela? • Evolvability Gánti, Kauffman
(transformed)?
The main problem of the origin of life is metabolite channelling
• Enzymes speed up reactions relative to the unwanted reactions
• Spontaneous decay reactions abound • Maintenance, not only reproduction,
requires autocatalysis dx/ dt = k x – d x = 0
TP
All network models neglecting side reactions are seriously incomplete
• E.g. protein networks • In model assumptions, a reaction is either
good or neutral for the system – but the number of harmful transformations is in fact much higher
• Did life emerge from a chemical canyon?
Chemical evolution was a race between tar formation and life
formation
Chemical networks
Life Tar
What fraction of planets would end up with just tar?
Test-tube selection experiments yield novel ribozymes and show that
• Catalysis of C-C bonds was feasible • Even hydrophobic molecules can be
specifically recognized • The RNA world is likely to have been
metabolically complex
The assembly of RNA structures
• Combinatorial assembly of RNA structures
• Submitted to selection for function between chemotons
• 1979
The channelled assembly of ribozymes (1983)
• The presence of substrates gives guidance to ribozyme assembly
• Good enzymes and bad enzymes will affect the fitness of the chemotons
A hatás
