Edith Chepkorir In her thesis, Assessing the risk of transmission of Yellow Fever and Dengue viruses by Aedes (Stegomyia) mosquito populations in Northern Kenya, the promovenda employed cross-sectional field surveys and laboratory experiments to assess the risk of Flavivirus disease transmission. The researcher described the occurrence, diversity and blood feeding patterns of potential vectors of dengue and yellow fever, as well as the vector competence and genetic variability of Aedes aegypti populations. She further identified serological evidence for circulation of the Zika, dengue, and yellow fever virus in human populations in 2016 to 2017. Her study provides critical insights into entomologic risk factors and evidence of human exposure to these Flaviviruses in Kenya. The findings contribute to understanding of the potential for the re-emergence of these viruses, their circulation and the populations at risk, highlighting the need for improved disease surveillance incorporating a ‘One Health’ paradigm and measures to inform policy on arbovirus disease control in Kenya. Juan Lodiwicus Janse van Nieuwenhuizen In his thesis, Oxygen deprivation state as a marker for the severity of obstructive sleep apnea, the promovendus developed a new parameter to aid in the diagnoses of the severity of obstructive sleep apnea. Many patients are mis- or underdiagnosed as a result of some of the current methods of diagnosis and due to conventional parameters omitting important physiological variables. Using a full polysomnography to test subjects for a single night, the researcher was able to create a new parameter known as the oxygen deprivation state. This new parameter was compared with the major descriptors of sleep apnea and proved to correlate more strongly with most of them as compared to currently used parameters. The promovendus also developed a new grading scale for the severity of the disorder based on his findings. The newly developed parameter may prove to describe the severity of the disorder known as sleep apnea more accurately. The degree Doctor of Philosophy

In her thesis, Development of a breast cancer spheroid model for use in drug discovery and cancer biology research, the promovenda established a 3D in vitro breast cancer cell culture system that allows us to mimic the microtumour architecture of breast cancers. She developed efficient methods to assess spheroid survival and used these to determine the efficacy of a number of developmental anti-cancer compounds. This has allowed for the rapid screening of compounds saving money and time in drug development. Furthermore, she investigated the biology of this system to show that the adhesive mechanisms of cells differ substantially in 2D vs 3D and these changes can have dramatic effects on drug efficacy. In conclusion, the promovenda has established the first 3D breast cancer culture system in South Africa to be used as a high throughput screening platform for anticancer drugs and a tool to study breast cancer biology within a in vivo-like environment.

In her thesis, Assessing modified chitosan wound dressings to enhance wound healing in the porcine model, the promovenda synthesised a hydrophobically modified chitosan and combined it with collagen and platelet-rich plasma to produce a filler type wound dressing that promoted growth of new cells and reduced healing times in wounds involving large areas. In vitro assays were utilised to characterise the product and to prove the material’s biocompatibility and sustained release of platelet-derived growth factor. Observations from in vivo full-thickness porcine skin wounds demonstrated enhanced initial haemostatic properties and increased collagen density with stimulated growth of new epithelial cells in the wound bed of the modified dressing when compared to the control treated wounds. Overall, reduced healing times and less scarring was observed using the modified chitosan dressing. These findings provide evidence that chitosan-based wound dressings could reduce the requirement for autologous skin grafts for extensive wounds. The degree Doctor of Philosophy

In the thesis, Evaluation of a strategy based on multi-drug targeting of cancer proteins in breast cancer cell lines, the promovenda assessed the in vitro combinatorial effects of several novel compounds for antineoplastic activity in two distinct breast cancer cell lines and a non-cancer cell line. The promovenda discovered that combining a polypharmacological antimitotic estradiol analogue with a novel bromodomain inhibitor resulted in synergistic in vitro antineoplastic activity. Furthermore, the combination was selective towards triple negative breast cancer (TNBC) cells compared to hormone responsive breast cancer and non-cancer breast cells. By using a series of scientifically sound techniques the study contributes towards an understanding of the mode of action of the combination that leads to downstream pathway activation, which culminates in both apoptotic and autophagy-related cellular demise. Candice van Wyk In her thesis, In vitro mechanistic study of a novel small molecule inhibitor of Candida albicans from Ptaeroxylon obliquum, the promovenda investigated the mechanism of action of a novel antifungal compound, obliquumol, isolated from the South African sneezewood tree in order to develop this compound into a natural product to combat candidiasis. Obliquumol caused structural and morphological damage to the cytoplasm of C. albicans. Obliquumol demonstrated no selectivity for cancerous cell lines compared to non-cancerous cell lines. Therefore, obliquumol is regarded as safe and its preparations have the potential to be used as an oral rinse or gel in the treatment of oral candidiasis. Phenotype MicroArrays identified key microbial enzymes that represented targets for the antifungal compound, obliquumol. Candida albicans treated with obliquumol were unable to utilise various substrates necessary for metabolism and virulence. Obliquumol would therefore be an ideal candidate for therapeutic interventions against C. albicans infection.

In her thesis, A multi-method study to explore perceptions and attitudes towards neuroanatomy in an undergraduate medical curriculum, the promovenda utilised qualitative and quantitative methods to explore the attitudes of anatomy lecturers, undergraduate and postgraduate medical students towards the teaching and learning of neuroanatomy in the South African medical curriculum. She further examined the current perceptions of these stakeholders about the relevance of neuroanatomy in the medical curriculum and the current teaching approaches employed. The findings indicate that some medical students have a dislike or fear towards the neurosciences (neurophobia), which could be ascribed to the current teaching approaches used and limited exposure to neuroanatomy. The research emphasises that no single best teaching method for neuroanatomy exists but that teaching should be student-centred. By reflecting on the perceptions, attitudes and teaching approaches to neuroanatomy, and by making the necessary changes, students could be assisted overcoming their fear of the neurosciences.

Liezl Balfour In her thesis, Implementation and evaluation of a clinical pathway for non-invasive ventilation in critical care: a person-centred practice development approach, the promovenda used a mixed-method design through a person-centred practice development approach utilising emancipatory action research. This study aimed to implement the NIV clinical pathway for critically ill patients collaboratively. Analysis of quantitative and qualitative data indicated that workplace culture directly impacted nurses’ ability and motivation to implement evidence-based practice through innovative strategies. The collaborative utilisation of a person-centred practice development approach aided the implementation of the NIV clinical pathway and the identification of workplace culture and moral injury among critical care nurses as limitations to effective research evidence application. The study highlights the complexity of transferring evidence into practice. It will assist nurse leaders in engaging nurses in critical conversations to collaboratively develop strategies to achieve moral reconciliation and adapting workplace culture to enhance evidence-based practice, improve patient outcomes and multidisciplinary collaboration.

Christian Lokotola Lueme In his thesis, The interaction effect of temperature and air pollution on unintentional injury, respiratory and cardiovascular disease mortality and hospital admissions in Cape Town, South Africa, the promovendus observed that apparent temperature and air pollution significantly increased respiratory disease and cardiovascular disease hospital admissions. The study was the first of its kind in South Africa. The risks posed by air pollution differed on cold, normal and warm days. Cold and warm apparent temperatures had different effects on unintentional injuries in Cape Town, Durban and Johannesburg. Long-range transported air pollution posed no clear risk to cardiovascular disease mortality, but increased respiratory disease mortality on warm days. The results of this thesis may assist policy makers and public health interventions dealing with climate change and air pollution health risks.

In his thesis, Development of a PBPK-based health risk assessment framework for exposure to cerium, gold, silver and titanium engineered nanoparticles, the promovendus developed and implemented a novel integrated approach for inhaled engineered nanoparticles risk assessment using several health risk assessment frameworks. The study demonstrated that exposure to nanoparticles through inhalation could cause disruption of physiological homeostasis and lead to adverse health effects. The study used physiologically based pharmacokinetic (PBPK) modelling to determine biokinetics of nanoparticles and assessed the suitability of PBPK models for use in risk assessments. PBPK modelling demonstrated that biokinetics of nanoparticles depended on the size, dose, concentration, duration of exposure and coating of nanoparticles. A novel health risk assessment framework was constructed using integration of current health risk assessment and a health information integration framework. The novel health risk assessment was applied to inhalation exposure to gold nanoparticles. The findings suggest that nanoparticle toxicity is dose-dependent.

In her thesis, Establishing a heuristic framework for effective attenuation of traffic noise transmission in typical naturally ventilanted classrooms in urban schools in Gauteng, South Africa, the promovenda developed a heuristic framework to guide architects and provincial school planners regarding traffic noise attenuation. The framework was developed using acoustic simulation software validated by field measurements. To reduce the background noise level in classrooms adjacent to busy roads, two attenuation interventions were modelled in various scenarios. The interventions proposed were a noise barrier and an increased distance between the classroom and the noise source. The resultant framework presents a selection matrix for scenarios of various barrier heights and classroom distances from the noise source that are predicted to result in a suitable background noise level in the classroom. A reasonable background noise level is essential for effective communication, learning and academic performance of learners. Classrooms exposed to traffic noise and ventilated through open windows, as many in Gauteng are, bear the risk of either poor acoustics or poor ventilation or both, to the detriment of learners and teachers.

In his thesis, Selection hyper-heuristics for population-based meta-heuristics in continuous dynamic environments, the promovendus investigated the ability of selection hyper-heuristics to select the most appropriate optimisation method over time to solve dynamic optimisation problems (DOPs) better than the individual optimisation methods were able to. The heterogeneous meta-hyper-heuristic (HMHH) framework was extended to solve DOPs: criteria were established to identify appropriate meta-heuristics for DOPs, global and island neighbourhoods that govern heuristic visibility of the population were introduced, and various heuristic selection triggering mechanisms were presented. A new performance measure for DOPs, namely the relative error distance, was proposed to improve existing popular performance measures. A new measure for heuristic diversity in population-based meta-heuristics was introduced. Additional measures were proposed that consider the heuristic reassignment frequency and the heuristic reassignment volume over time. Empirical studies examined the performance and behaviour of various hyper-heuristic selection operators using the proposed measures.

In her thesis, A critical analysis of public discoveries on context-awole technologies and of their informed use:The case of users in a developing country, the promovenda examined the information asymmetry that occurs between creators and consumers of context-aware technologies such as smartphones. Although ostensibly created to untether the consumer from devices and improve user efficiency, these increasingly autonomous technologies dynamically gather and analyse data about the consumer, often without their knowledge and consent. Much of what one knows results from socialisation, which takes place mostly through communication. Communicative practices of technology providers play a significant role in the production of ideologies. Ideologies, which exemplify the exertion of power, are used to shape perceptions that may be contrary to the interests of those who hold them. Indeed, the needs arising from the unique circumstances of developing countries are shown to make consumers more vulnerable and susceptible to common technological ideologies. Powerful creators are shown to distort the discourse surrounding these technologies and, as a consequence, impair rational decision-making regarding their use. Adriana Botha Johanna Maria In her thesis, A learning managment system based framework for higher education quality programme review, the promovenda developed the PAIR framework (Programme Outcomes Alignment, Implementation and Reporting) to improve student success at a South African institution of higher education. The goals tool of the Blackboard Learning Management System was incorporated into PAIR. The framework allows universities to contextualise outcomes coverage and achievement reporting to improve programme quality and to inform intervention strategies for the advancement of effective student learning. PAIR offers the possibility of an effective working partnership between academic staff, technical support staff and educational professionals to determine institutional student learning outcomes consistent with the programmatic collection of data on student performance. The comprehensive programme assessment praxis provides departments with a uniquely systematic process for reporting on the assurance of learning during an accreditation cycle. Through international collaboration, the inclusion of a programme design phase elevated PAIR’s delivery processes to an institutional level that can be applied across different university systems.

Huaqiong LLiu In his thesis, A systematic design of e-commerce logistics from collaborative management theory perspective, the promovendus proposes a novel method for the systematic design of e-commerce logistics. A new framework was developed using relevant theories and methods of systems science, systems engineering, project management and collaborative theory. The resulting design of a logistics system also comprised a four-stage radial-spoke logistics network. As part of the design, the sites of e-commerce hubs were selected by applying useful mathematical modelling methods. A comparative analysis and a simulation method were employed to improve supply chain facilities and equipment, optimise transportation processes and integrate information. Through his research, the candidate contributes to bridging the gap between the first- and last-mile delivery in the supply chain, improving logistics efficiency, and reducing risks in the logistics process. His contribution by using a design structure matrix method approach is relevant to the operation, information platform, e-commerce regional distribution centres, e-commerce hub, chain stores and convenience stores. Rina Roux Nicolene In her thesis, A roadmap for the titanium metal industry of South Africa, the promovenda proposes a novel method for producing a roadmap for the fragmented South African titanium metal industry. The roadmapping framework was developed by subjecting each stage of a value chain to the generic industry technology roadmap layout for market, product, technology, research and development, and resources. Roadmaps were first produced for each value chain stage. These roadmaps were then either included or excluded from the overall South African titanium metal industry technology roadmap based on local requirements. Despite South Africa’s large titanium mineral reserves, the country does not have a complete titanium metal value chain. South Africa exports low-value raw materials and then imports high value-added products. This research considered which stages of the titanium metal value chain should be localised in the next decade and suggested a roadmap to advance these stages. The degree Doctor of Philosophy
Dafeng Xu In his thesis, A critical evaluation of reisk on large scale logistics projects, the promovendus proposes a logistic alliance risk identification and analysis system based on the Particle Swarm Optimization (PSO) algorithm, combined with the Fuzzy Comprehensive Measurement Method (FCMM) and PSO-Analytic Hierarchy Process (AHP). The adapted method transformed the PSO into the PSO-AHP method, which was then applied and evaluated in the risk assessment of large-scale logistics projects. The candidate not only combined AHP and PSO but also incorporated the Work Breakdown Structure (WBS) method to build a novel risk indicator system. With the development of Chinese economics and a special relationship with South Africa, the connection between the two countries relies heavily on the development of logistics projects. An important contribution of the research is that it provides a way of limiting the risk inherent to large-scale logistics projects. The candidate focuses on measuring as well as analysing risks and provides meaningful new results for actual practical logistics project cases by comparing risk results using different novel assessment approaches.
