“and what exactly is it you are doing in Thuringia?” Udo Winkels, the hotel director of the Eisenacherhof in Eisenach,was asking me. - Answer: “empty bottles...small empty bottles from glass for Chromatography.” - Question: “for what?”

This conversation started because Udo asked me whether I would become the fi rst international member of the LutherAssociation. Eisenach with Wartburg and Bach house bound to the Middle Ages endepts you to your adopted country,so I agreed to join. Yet, how did this Swiss – German friendship between Glastechnik Gräfenroda GmbH and infochroma ag start?

In autumn 1992, three years after the Berlin Wall came down, infochroma was looking for a new vial supplier. After some “detours” we found the address of Glastechnik Gräfenroda GmbH, a company founded after the Reunition, and after several phone calls we decided to travel to the “wild East” to visit our future supplier.

After a never ending trip with traffi c jams, road works, detours, road blockages and other incidents, we fi nally arrived in Gräfen-roda. We viewed the site which at this time consisted of a small workshop, a production hall under construction and one machine for the production of vials. The negotiations were quick and an agreement was found until the question arose: “Would you like some more coffee?” - Answer: “Nein Danke, ich bin bedient.” which to the German listener means why bother lets get out

of here as quickly as possible. Luckily my cousin from the so called “old German states” was with me and acted as translator. He explained: “Please don’t get this wrong. He is Swiss and in Switzerland this is an absolutely polite reply to your question. It doesn’t mean that we’ve had enough of you guys, it just means we don’t wan’t any more coffee.“

After overcoming this language obstacle, the cooperation prospered. The factory has grown, several products have been patented and much was paid for experience. But the rebuilding of East Germany, although on a small modest scale, has succeeded in Gräfenroda and meanwhile many empty bottles are produced, bottles that are fi lled with products by customers all over the world – and used not just forChromatography.

Autosampler Vials for Chromatography with 12 x 32 mm dimension are used in most instruments. If there is only little sample volume or as much sample as possible should be taken from the vial, an optimised solution is needed.

Chromatography Vials for Optimised Microsampling

Chromatography autosamplers generally use 12 x 32 mm vials. These vials can take 1 ml sample. However, if the available sample volume is limited or a maximum amount of sample should be withdrawn from the vial, an optimised solution is needed. A limited volume vial that stands without support and keeps the outer dimension so that it can still be used in common autosamplers.

We developed three solutions that fulfi l these requirements. To fi nd the proper vial for a certain application means to ask oneself one important question. How much sample volume do I have and how much sample do I need for the analysis. The difference between the two, the volume that remains in the vial, we call residual volume. This residual volume defi nes which vial is needed.

Many vial suppliers advertise the dead volume, the quantity of sample that cannot be withdrawn from the vial; however

this volume cannot be defi ned as simply as that. The insertion depth of the needle, the geometry of the needle, how well the needle hole is covered with sample so that no air is sucked up, all these factors defi ne the dead volume of an application. We fi nd that the defi nition of the dead volume without the exact specifi cation of those parameters is useless.

We defi ne the volume in the foot of the vial at 4 mm fi lling level, with the provision that an autosampler needle inserts down to about 1-2 mm above base and that the needle should be covered with 2 mm sample to guarantee a reliable sampling. The actual dead volume of our vials is usually a lot smaller than the residual volume that we publish, yet if an autosampler with side hole needle is used, the published value can also be too small. A regular 12 x 32 mm standard vial for example has a residual volume of 400 μl.

Which one of our 3 microvials should be used?

the 1.5 ml Vμ-Vial our „Vase Vial“ Our Vase Vial or Vμ-Vial has the largest volume. It takes 1.5 ml up to it‘s shoulder and has 15 μl at the foot. Since our vase vial is the most economic solution, we recommend this vial for all applications that go with a residual volume of 15 μl. The vase is also the most versatile vial, it is available with all common closure versions including 8-425 screw top which we call „NM“ or „Narrow Mouth“ as these vials have a narrow opening. Notabene the Vase Vial is our very own invention and we patented the form as well as the production process. Now we also produce bigger Vase Vials, the VMax-Vial which you will fi nd on pages 20 - 21 of this catalogue.

the 350 μl μ-Vial Our fi rst microvial, historically speaking, is the i3μ type with a fused insert that has one base with the outer vial. It‘s like a vial and insert combination, only that vial and insert are pre-assembled, fused and the insert is well centred. The total volume is 350 μl with 10 μl at the foot.

the 250 μl insert Vial iV2μ Our latest microvial is the iV2μ type. It‘s different because the insert wears the head of the vial so that the insert itself is closed. That means no sample can escape into the outer vial. The body of the vial is a simple tube that is fused with the base of the insert. We produce these vials on a one-station machine, we especially developed, a machine that allows us to draw the insert longer and more narrow than is possible with common techniques. The total volume is 250 μl with 8 μl at the foot. The iV2μ is especially suited for volatile micro samples or gas chromatography applications and rounds our limited volume range off at the bottom.

Non-Pigmented Silicone/Natural PTFE

Bleed Comparison Between Natural and Red PTFE Sample Using Gas ChromatographyIntuitively, one would surmise that pigmented materials (i.e. those that are coloured in some way) would be more likely to pose as a contaminant source when compared to materials that are not pigmented. This study compares two (2) 0.1 mm thick samples of PTFE. One is red in colour and the other is non-pigmented or natural. The Total Peak Area illustrates that the Natural (non-pigmented) PTFE shows a value of (0) zero, while the Red PTFE yielded a total peak area of 315. Using lower bleed materials provides truer readings, eliminates another potential error source, decreases sample contamination, and improves productivity.

iV2μ-Vial

12 x 32 mm Vial

Like the μ-Vial, the iV2μ-Vial has the insert integrated into the body of a 2 ml vial, but the insert itself is closed with the cap. Because of the directly closed 250 μl insert, the iV2μ-Vial is suited for use with volatile samples, since no sample vapour escapes into the hollow space between vial and insert.

With our specially developed one-station machine we can draw the insert substantially longer and more narrow than was pos-sible until now. Therefore the iV2μ-Vial is also suited for

autosamplers with side port needle or smallest sample volume of a few microlitres. We recommend the iV2μ-Vial when a dead volume of < 5 μl is needed. Depending on the geometry of the needle and needle height, however, the dead volume can be much lower than 5 μl.

Standard Caps with Rubber/PTFE Septum, although used for routine analysis, should have the necessary purity and should be produced within the required tolerances. We found that the cheapest varieties don‘t necessarily fulfi l the quality requirements of our customers.

That‘s why our standard caps are batch tested for extractable substances in co-operation with Interlaber Belp and for an easy handling in the laboratory they are supplied in clear PET screw top jars.

The difference between the three Screw Vial TypesDescription of the Screw Threads and the nomenclature that goes with them

Most Autosampler screw threads are GPI (Glass Packaging Institute) Threads, an US standard for screw threads of glass vials. The following screw threads are used for 2 ml Autosampler Vials:

Agilent Technologies compatible (HP) Screw Caps (pages 11)This is a 9 mm thread which was especially developed for Agilent Autosamplers so that the Robotic Arm System can handle screw vials.

Headspace Screw Vials with Magnetic Screw CapIn Headspace Chromatography, the vial closure has to be pressure tight. This takes some effort and when closing larger series and is tiring. Our newly developed screw-top Headspace Vial with a glass fi bre fi lled polyamid screw cap requires a torque of less than 1 Nm for a pressure tight closure whereas common metallic solutions require a torque of more than 8 Nm.

In addition, the septa, which is the price defi ning factor, has a smaller diameter. This makes the new closure more cost effective.

VMax-Vial

The VMax-Vial has a cylindrical foot. We chose a „cylindrical foot“ over a „cone foot“ as we think a cylindrical foot is technically supe-rior for the sample taking in an autosampler.

A sampler usually pierces the sealing septum with a needle which descends to a defi ned height to take the sample. The needle has two functions, to pierce the septum and to withdraw the sample. The needle should be formed so that it easily pierces the septum. If the needle has a bevel tip it mustn‘t hit the bottom of the vial or the needle tip will be bent which will negatively infl uence the further piercing of the septum. If the needle has a cone tip it mustn‘t hit the bottom of the vial either as the needle whole could become sealed by the vial bottom so that no sample is taken. If a needle with side hole is used the system may be adjusted so that the nee-dle tip rests on the bottom of the vial but still the needle hole is above the bottom of the vial. Independent of the needle geometry the highest point at which the sample enters the needle defi nes the death volume of a vial.

In order to get the maximum sample return, air should usually not be sucked into the needle with the sample. This requires minimal sample coverage over the needle hole. For most samplers, it can be said that the needle descends to a defi ned level above the intended withdrawing level (commonly 2.5 mm above withdrawing level). As the withdrawing level is at the bottom of the vial, the vial producer must assure that the vial bottom has a reproducible inner height. The dead volume can be derived with the measure that the bottom of the vial has a radius r (=øfoot/2) and the needle hole is above the bottom at height h.

The death volume of the cone version forms a cone stump with the side angle α; the cylindrical version, as produced by GTG, forms a cylinder.

This means that the dead volume of the cone version is bigger by a share of (r*h2*tan(α) + h3/*tan2(α) ). With an angle α bigger 45°, tan(α) > 1 and increases considerably. The same results when the needle hole has to be covered with sample. In thiscase height h = height h(needle above base) + c(coverage with sample).

in this example applies: death volume to coverage r = 1.5 mm, h = h + c = 1.5 mm + 2.0 mm = 3.5 mm and α = 45° -> tan(α) = 1VKS= 86.0 μlVZyl = 24.7 μl

It can be concluded that in order to minimise death volume tan(α) one has to aim towards zero which means that a cylindrical solu-tion is an advantage. The actual death volume has to be reduced by the displacement volume of the needle but the infl uence is the same for both cylindrical and conical and therefore is not shown in this calculation.

We are able to form the foot with a thorn such as is required by the geometry of the needle under the adherence of certain measurements.

FlAmp

FlAmp - the resealable Ampoule

- sample can be kept after breaking the ampoule- tight seal, less then 0.5 % loss within one month- ampoule can be resealed over 50 times

If a sample or a sample mix is to be stored over an extended period of time a glassampoule is the best sealing option as it is the only one that will guarantee a maximum storage time. It is a disadvantage though that ampoules, once broken, cannot be resealed. Often, however, sample is left over as it is not needed straight away but used over a period of time.

This is where the FlAmp comes in. When we developed the FlAmp our aim was to develop a sealing option, which allows the user to use up the sample over a period of time after breaking the ampoule, and at the same time have the guarantee that sample loss is a little as 2 % or less over that period of time. After 18 months of research and development we had the FlAmp.

And we had achieved more than what we had aimed for. An extensive study carried out by Interlabor Belp (www.interlabor.ch) showed that the loss of substance within a month is on average less than 0.1% and this with Methylenchloride as solvent and 800m Bar ambi-ent pressure. The absolute highest detected loss was 2 mg in a month which proves the excellent sealing characteristics of the plug‘s sealing rings in combination with the conical sealing surface of the ampoule. This result, however, can only be achieved if the plug is pushed in hard enough for the two sealing rings to seal visibly. If this is not the case the seal will be less tight. The sealing rings of the plug wear off but they showed a standing time of 50 closures at the same high level of tightness.

The FlAmp would typically be used for Chromatography Standards used to test columns; over a period of several days the calibration standard of one ampoule is used up. In the example below we ran a GC test and for one week took a daily sample from the ampoule. As you can see from the Chromatogram, the sample becomes slightly more concentrated as the week carries on, due to evaporation of the solvent during handling. The composition of the standard, however, has remained the same which makes the FlAmp the perfect choice for this purpose.

The FlAmp is available in amber with 1ml, 2 ml and 5 ml volume. The plug is made from inert Fluoropolymere and can be used for over 50 closures.
