We propose here extension of this technique to the field of plant downstream processing (DSP), where plants are used as a bioreactors for recombinant protein production. The main problem in plant-based bioreactors is the presence of chlorophyll and phenolic compounds in plant extracts, which tend to precipitate and denature proteins. Their removal from the extracts is essential, since they have a large impact on the DSP performance. In the present work we studied the application of an electrocoagulation based technique to clarify tobacco leaf extracts by removing chlorophyll and phenolic compounds. By manipulating the pH of electrocoagulation, 90% of chlorophyll, 65% of phenolic compounds, and only 23% of total protein were removed from the extract. Therefore, the aim of the present study, part of FIBRA (the Frailty in Brazilian Elderly study), was to investigate relationships between frailty and measurements of physical activity. The study used a probabilistic sample of 689 elderly (72.28 +/- 5.40 years; 68.21% women). An adapted version of the Minnesota Leisure Time Physical Activity Questionnaire was used and two criteria were established for measuring physical activity: regular physical exercise and weekly calorie expenditure during physical exercise and household activities. Of the total sample, 15.09% scored on weight loss, 17.13% on fatigue, 16.96% on low grip strength, 15.87% on slow gait, and 43.02% on comorbidities. According to the calorie expenditure criterion, 83.55% of the elders were classified as active, as compared to 45.27% according to the physical activity criterion. Low calorie expenditure was associated with low grip strength, slow gait, and advanced age. Sedentary lifestyle (limited physical activity) was associated with comorbidities. However, ACC-HGT can also undergo transformation to adenocarcinomas which are not poorly differentiated. ACC-HGT is generally considered to be an aggressive variant of ACC, even more than solid ACC. This study was aimed to describe the genetic changes of ACC-HGT in relation to clinico-pathological features, and to compare results to solid ACC. Genome wide DNA copy number changes were analyzed by microarray CGH in ACC-HGT, four with transformation into moderately differentiated adenocarcinoma (MDA) and two into poorly differentiated carcinoma (PDC), and five solid ACC. In addition, Ki67 index and p53 immunopositivity was assessed. ACC-HGT carried fewer copy number changes compared to solid ACC. Two ACC-HGT cases harboured a breakpoint at 6q23, near the cMYB oncogene. The complexity of the genomic profile concurred with the clinical course of the patient. Among the ACC-HGT, p53 positivity significantly increased from the conventional to the transformed (both MDA and PDC) component. Specifically, we present an application of a methodology that allows interpreters to obtain effective porosity 3D maps from post-stack 3D seismic amplitude data, using measured density and sonic well log data as constraints. In this methodology, a 3D acoustic impedance model is calculated from seismic reflection amplitudes by applying an L(1)-norm sparse-spike inversion algorithm in the time domain, followed by a recursive inversion performed in the frequency domain. A 3D low-frequency impedance model is estimated by kriging interpolation of impedance values calculated from well log data. This low-frequency model is added to the inversion result which otherwise provides only a relative numerical scale. To convert acoustic impedance into a single reservoir property, a feed-forward Neural Network (NN) is trained, validated and tested using gamma-ray and acoustic impedance values observed at the well log positions as input and effective porosity values as target. The trained NN is then applied for the whole reservoir volume in order to obtain a 3D effective porosity model. While the particular conclusions drawn from the results obtained in this work cannot be generalized, such results suggest that this workflow can be applied successfully as an aid in reservoir characterization, especially when there is a strong non-linear relationship between effective porosity and acoustic impedance. NO. 5111; MERIDIANI-PLANUM; PALEOMAGNETIC POLES; POLAR WANDER; DEPOSITS; STRATIGRAPHY; REGIONAB Gale Crater contains a 5.2 km-high central mound of layered material that is largely sedimentary in origin and has been considered as a potential landing site for both the MER (Mars Exploration Rover) and MSL (Mars Science Laboratory) missions. We have analyzed recent data from Mars Reconnaissance Orbiter to help unravel the complex geologic history evidenced by these layered deposits and other landforms in the crater. Results from imaging data from the High Resolution Imaging Science Experiment (HiRISE) and Context Camera (CTX) confirm geomorphic evidence for fluvial activity and may indicate an early lacustrine phase. Analysis of spectral data from the CRISM (Compact Reconnaissance Imaging Spectrometer for Mars) instrument shows clay-bearing units interstratified with sulfate-bearing strata in the lower member of the layered mound, again indicative of aqueous activity. The formation age of the layered mound, derived from crater counts and superposition relationships, is similar to 3.6-3.8 Ga and straddles the Noachian-Hesperian time-stratigraphic boundary. Thus Gale provides a unique opportunity to investigate global environmental change on Mars during a period of transition from an environment that favored phyllosilicate deposition to a later one that was dominated by sulfate formation. One of the challenges in these studies is the identification of the cross-linked peptides. The interpretation of the MS/MS data generated in cross-linking experiments using N-hydroxy succinimide esters is not trivial once a new amide bond is formed allowing new fragmentation pathways, unlike linear peptides. Intermolecular cross-linked peptides occur when two different peptides are connected by the cross-linker and they yield information on the spatial proximity of different domains (within a protein) or proteins (within a complex). In this article, we report a detailed fragmentation study of intermolecular cross-linked peptides, generated from a set of synthetic peptides, using both ESI and MAID! to generate the precursor ions. The fragmentation features observed here can be helpful in the interpretation and identification of cross-linked peptides present in cross-linking experiments and be further implemented in search engine's algorithms. The three variables studied (molar ratio of hesperidin to decanoic acid, [bmim]BF(4)/acetone ratio and lipase concentration) significantly affected the conversion yield of acylated hesperidin derivative. Verification experiments confirmed the validity of the predicted model. The lipase showed higher conversion degree in a two-phase system using [bmim] BF4 and acetone compared to that in pure acetone. One of the possible pathways of H(2)O(2) production involves oxalic acid (Oxa) degradation by apoplastic oxalate oxidase. Here, we analyzed the production of H(2)O(2), the presence of calcium oxalate (CaOx) crystals and the content of Oxa and ascorbic acid (Asa) the main precursor of Oxa in plants in susceptible and resistant cacao (Theobroma cacao L) infected by the hemibiotrophic fungus Moniliophthora perniciosa. We also quantified the transcript level of ascorbate peroxidase (Apx), germin-like oxalate oxidase (Gip) and dehydroascorbate reductase (Dhar) by RT-qPCR. We report that the CaOx crystal amount and the H(2)O(2) levels in the two varieties present distinct temporal and genotype-dependent patterns. Susceptible variety accumulated more CaOx crystals than the resistant one, and the dissolution of these crystals occurred in the early infection steps and in the final stage of the disease in the resistant and the susceptible variety, respectively. High expression of the Glp and accumulation of Oxa were observed in the resistant variety. The content of Asa increased in the inoculated susceptible variety, but remained constant in the resistant one. The susceptible variety presented reduced Dhar expression. The role of H(2)O(2) and its formation from Oxa via Apx and Glp in resistant and susceptible variety infected by M. perniciosa were discussed. (C) 2011 Elsevier Masson SAS. A slice of epidote natural monocrystal was taken from the (010) plane and then divided into five pieces. Each piece was then irradiated with ions whose incidence angles (zenith angles) were of 15 degrees, 30 degrees, 45 degrees, 60 degrees and 75 degrees with respect to y-axis. The azimuthal angle of incidence of the ions was the same for the pieces 15 degrees, 60 degrees, 75 degrees and 180 degrees apart for the pieces 30 degrees and 45 degrees. Etching times were of 10, 20, 30, 40 and 50 min (HF 40%, 35 degrees C). The results show that etching velocities of ion tracks are higher in directions closer to the y-axis. The mean lengths of the ion tracks, regarding the angles, were of 23.14 +/- 0.21 (15 degrees); 19.89 +/- 0.08 (30 degrees); 19.39 +/- 0.04 (45 degrees) and 16.59 +/- 0.10 mu m (60 degrees). Since no tracks were identified in the 75 degrees aliquot it was assumed that the epidote has a critical angle, for recording of ion tracks with this mass/energy ratio, between 60 degrees and 75 degrees. Despite the evidence of a defective proprioceptive sensory input in SN, the prominent gait and truncal ataxia raises the question of a concomitant involvement of the cerebellum. Objective: To evaluate cerebellar atrophy in SN. Method: We analyzed MRI-based volumetry of anterior lobe (paleocerebellum) and total cerebellum in patients with non-paraneoplastic chronic SN and compared to age- and gender-matched controls. Results: Cerebellum and anterior lobe MRI volumetry were performed in 20 patients and nine controls. Mean anterior lobe and cerebellar volume were not statistically different. Three patients (15%), however, had an abnormal anterior lobe and cerebellar volume index (values outside 2.5 standard deviations). One of them also had a specific atrophy of the anterior lobe. All these patients had infectious or dysimmune associated SN. Conclusion: Cerebellar atrophy is infrequently associated with SN, but can be found in some patients with SN related to infectious or immune mediated conditions. Method: We applied the Fugl-Meyer Assessment (FMA), Nottingham Sensory Assessment (NSA), and several motor and sensory tests: Paper manipulation (PM), Motor Sequences (MS), Reaching and grasping (RG) Tests Functional (TF), Tactile Discrimination (TD), Weight Discrimination (WD) and Tactile Recognition of Objects (RO). Results: We found moderate correlations between the FMA motor subscale and the tactile sensation score of the NSA. Additionally, the FMA sensitivity was correlated with the NSA total; and performance on the WD test items correlated with the NSA. Conclusion: There was a correlation between the sensory and motor functions of the upper limb in chronic hemiparetic stroke patients. Method: It was studied 14 patients with hemiplegia who were submitted to a program with FES during fourteen weeks. Brain perfusion SPECT was performed before and after FES therapy. These patients were further separated into 2 groups according to the hemiplegia cause: cranial trauma and major vascular insults. All SPECT images were analyzed using SPM. Results: There was a significant statistical difference between the two groups related to patient's ages and extent of hypoperfusion in the SPECT. Patients with cranial trauma had a reduction in the hypoperfused area and patients with major vascular insult had an increase in the hypoperfused area after FES therapy. Thirty-two 18-month-old adult male pigeons (Columba livia), weighing on average 350 g, were trained under different conditions: with tone-shock associations (experimental group; EG); with shock-alone presentations (shock group; SG); with tone-alone presentations (tone group; TG); with exposure to the training chamber without stimulation (context group; CG), and with daily handling (naive group; NG). The number of immunoreactive nuclei was counted in the whole TnA region and is reported as density of Zenk-positive nuclei. This density of Zenk-positive cells in the TnA was significantly greater for the EG, SG and TG than for the CG and NG (P < 0.05). The objective of the present investigation was to examine the thalami of patients with juvenile myoclonic epilepsy using a combination of multiple structural neuroimaging modalities. The association between these techniques may reveal the mechanisms underlying juvenile myoclonic epilepsy and help to identify the neuroanatomical structures involved. Twenty-one patients with juvenile myoclonic epilepsy (13 women, mean age = 30 +/- 9 years) and a control group of 20 healthy individuals (10 women, mean age = 31 +/- 8 years) underwent MRI in a 2-T scanner. The volumetric three-dimensional sequence was used for structural investigation. Evaluation of the thalamus comprised voxel-based morphometry. automatic volumetry, and shape analysis. Comparisons were performed between patient and control groups. Voxel-based morphometry analysis identified areas of atrophy located in the anterior portion of the thalamus. Post hoc analysis of automatic volumetry did not reveal significant differences between the groups. Shape analysis disclosed differences between patients and controls in the anterior and inferior portions of the right thalamus and in the anterior portion of the left thalamus. The present investigation confirms that thalami of patients with juvenile myoclonic epilepsy are structurally abnormal with impairments located mainly in the anterior and inferior sections. St. - HILL), found in Brazilian cerrado, has been used in the initial ripening stage as flour and/or starch in popular medicine due to its hypoglycemiant action. The aim of this work was to study the occurrence of phytochemical compounds, physical, chemical and technological characteristics of fruit of wolf flour and starch in the initial ripening stage. Flour and starch were extracted using known popularly and experimental methods (with sodium bisulfite) and chemical composition and technological characteristics were analyzed. The results were comparatively assessed by Tukey's test (p 200 mg/dL, high-density lipoprotein is < 35 mg/dL, low-density lipoprotein is > 130 mg/dL, and triglycerides are > 150 mg/dL. Dyslipidemia can be found in patients with malnutrition, a severe condition that needs prompt nutrition intervention. This report describes a case of malnutrition causing severe dyslipidemia in a newborn. Primary dyslipidemia was excluded by the presence of primary malnutrition, normal response to a postheparin lipoprotein lipase activity test, a favorable clinical course after nutrition intervention, and relatives' blood lipid levels close to normal that did not indicate familial dyslipidemia. The child was fed fat-free milk formula supplemented with medium-chain triglycerides and had adequate weight gain with a decrease in blood lipids. Subsequently the formula was changed to regular milk-based formula, and the child maintained adequate growth rate. Although blood lipids never returned to normal values for age and sex, they were lower than before treatment. Merrill] seeds. Utilising the alpha prime (alpha') subunit of beta-conglycinin tissue-specific promoter from soybean and the alpha-Coixin signal peptide from Coix lacryma-jobi, we obtained transgenic soybean lines that expressed the mature form of hGH in their seeds. Expression levels of bioactive hGH up to 2.9% of the total soluble seed protein content (corresponding to approximately 9 g kg(-1)) were measured in mature dry soybean seeds. The results of ultrastructural immunocytochemistry assays indicated that the recombinant hGH in seed cotyledonary cells was efficiently directed to protein storage vacuoles. Specific bioassays demonstrated that the hGH expressed in the soybean seeds was fully active. The recombinant hGH protein sequence was confirmed by mass spectrometry characterisation. The data used to define management zones were the chlorophyll index measured by SPAD sensor and the nutritional coffee leaf analysis performed in laboratory. The study was conducted in Jatoba Farm, located in Paula Candido, Minas Gerais state, Brazil. The data was collected in November, 2007 when the coffee fruits were starting their development. The coffee variety was Coffea arabica Catuai, and the total analyzed area was 2.1 ha. The management zones were generated using different set of data: the SPAD values; the N, P and K leaf concentrations; the N and Ca leaf concentrations; the N, Zn and B leaf concentrations; the N, P, K, Ca and S leaf concentrations; and the N, Ca and S leaf concentrations. The management zones generated by using K-means and Fuzzy C-Means did not present difference in management zone delimitation.
