Initiation & Clean Up Starting from tubers, initiated 20 new cultivars into tissue culture. Used heat and chemical treatment to clean viruses from 13 of these.

New Cultivars Received 32 additional varieties in tissue culture for growers.

Breeding Program/Special Projects Initiated new cultivars into tissue culture, maintained varieties kept for breeding/research purposes, and cut plantlets for the breeding program greenhouse. Research projects (cryotherapy and alginate bead storage) consumed approximately 35 days during the year.

Record Keeping Extensive paperwork to document all testing done on active and long-term storage clone banks, orders cut for growers and other labs, and to track the virus clean-up process.

Update on direct tuber testing for PVY-PCR:

- Various buffers, enzymes, anti-body dilutions, core location trials

- Preliminary results (see figure below) suggest test near ready *

- * Cores positive were taken directly under sprouts, viral dormancy may be issue after harvest

- Putative PVY strains in samples detected so far by multiplex IC-RT-PCR: ‘O’, ’N-WI’, & possibly ‘NTN’

- Direct testing, of tubers without sprouts, will be necessary for final validation

- Estimated cost of PCR confirmation, of wells testing ELISA-PVY+, $11 first well, $6/addl.well

- As protocols are streamlined, costs will come down for complete lot direct tuber testing
