동결건조에 향을 미치는 Factors
2. 시료의 두께 :
동결건조하고자 하는 시료의 두께는 1~2cm 정도로 얇을수록
좋습니다.

1. 최적의 진공도 세팅 :
진공이 많이 걸릴수록 기체의 체적은 오히려 증가하기 때문에
최적의 진공조건(eutectic point보다15~20℃ 낮은 진공)보다
고진공이 형성되면 동결건조 시간은 오히려 늘어나게 됩니다. 따
라서 적절한 진공도 세팅은 중요합니다.
� 모든 Christ 제품들은 진공도 세팅이 가능.

3. 선반 Heating :
시료에 에너지 투입효과가 생겨 건조시간을 줄여줍니다. 단, 이과정에서 시료가 녹지 않도록 하는 것이 중요합니다.
� 부분의 Christ 제품이 Shelf heating 이 가능하며, 시료의 Thawing(해동)을 방지하기 위한 Safety Pressure 기능이 있습니다.

1. 최적의 진공도를 맞춰 동결건조를 하기 위해서는 Eutectic point를 아는 것이 가장 중요합니다.
Freezing Point (or Eutectic point)를 알아내기 위한 여러가지 방법이 있습니다.
- 열학적 수식에 한 계산 (ex. VDI Thermal Altas.)- Cryo-microscope- DSC (differential Scanning Calorimetry)-
measurement of temperature & resistance during freezing phase
→ Christ에서는 Lyo control sensor를 이용하여 시료가 어는 동안의 온도와 전기저항성 변화를 측정하여 eutectic point를 찾아

2. Freezing 속도와 동결건조 :
일반적으로 1 ~ 1.5 ℃/min 정도의 속도로 얼릴 때 Freezing concentration 현상이 생기지 않아 동결건조가 가장 잘 됩니다.
→ Epsilon 모델의 경우 프로그램을 이용하여 선반의 Freezing 시간을 설정하여 Freezing Speed를 조절 할 수 있습니다.

동결건조의 완료시점의 결정
동결건조는 primary와 secondary drying으로 나누어지는데
*primary : 시료중의 수분을 95% 이하까지 제거하며, eutectic temperature보다 (main) 15℃ 낮은 온도의 진공으로 실시하는게
이상적입니다.
*secondary : primary drying 이후 남아있는 소량의 수분을 제거하기 위한 단계로(final) 시료중의 수분을 99% 이하까지 제거하는
단계입니다. 이 과정은 물리적인 desorption 과정이기 때문에 가능한한 낮은 온도의 진공으로 세팅하며, shelf heating을
통해 에너지를 공급해줍니다.
1. product와 shelf의 온도를 비교하는 방법 :
product sensor의 온도와 shelf의 온도가 거의 비슷해지는 시점이(3~5도 차이) primary drying이 완료되는 시점이며, final
drying으로 진행하면 됩니다.
2. Pressure increase test를 이용한 방법 :
CHRIST LSC 모델 가능(intermediate valve 장착요)
prodcut chamber와 ice condenser 를 물리적으로 분리시켜, 시료에서 승화되어 나온 수분이 ice condenser로 달라붙지
못하게 함. Drying이 완료되지 않았다면 승화된 water vapour의 분압이 증가하여 압력이 올라가기 때문에 drying의 완료여
부를 알 수 있습니다.
Primary -> secondary 로 옮겨갈지 여부를 알려주거나, 최종적으로 건조가 완료됐는지를 알 수 있는 중요한 tool입니다.

�RVC 는 Sample 의 Evaporation, Drying, Purification 과 Concentration 을 위해 사용됩니다.
�온도에 민감한 미생물이나 clinical 물질을 Drying 시키고자 할 때, 1,350 에서 1,550 min-1 의 원심력으로 샘플에
아무런 향 없이 Drying을 가능하게 해주며, 이 때의 원심력은 sample이 Spitting 되거나 bumping되는 것을 막
아줍니다.
�증발된 Solvent는 Trap에 모이고 필요할 경우, 재사용이 가능하며 폐기도 가능합니다.
�아래의 표에서도 볼 수 있듯이, solvents에 진공을 걸어 주면 끓는점이 낮아지므로 온도에 민감한 샘플의 Drying에
적용 가능합니다. (Application Data 참조)

< Cooling Traps for Larger Volumes >�Evaporation 은 energy input에 향을 받을 뿐만 아니라, Diaphragm pump의 suction capacity에 의해서도 향을 받습니다.
�더 큰 용량의 vacuum pump를 사용하는 신에 Cryo-pump로서 Cooling-trap 을 사용하는 것이 더욱 효율적일 수 있습니다.
�Cooling-Trap 을 사용할 경우, Pumping Speed 가 향상 되므로 Sample 의 Drying Time을 줄일 수 있습니다.
그러나 Pump에 비해 가격이 약간 비싸므로 Application에 따라 고려하시기 바랍니다.
