INERTSIL® (GL Sciences)
Inertsil®Manufactured by GL Sciences, Tokyo, Japan, Inertsil® LCcolumns represent state-of-the-art silica manufacturingtechnology and are unrivaled for their physical and chemicalstability.
Inertsil® silica gels are free of dents and cracks, which mightweaken the spherical particles or lead to inefficient columnpacking. The uniformity of the base silica, with a purity of99.999%, greatly contributes to the chemical stability andexcellent resistance to high and low pHs.
SEM photographs of Inertsil® silica gels are shown below. There are many silica gels on the market which have rough sur-faces and cracks. In contrast, Inertsil® silica gels have a highly smooth surface andspherical shape, which provides durability and high inertnesswith good reproducibility over a long period of time, making themideal for LC applications.A rigorous test by SEM is employed for base silica gels forInertsil® packings.
What makes the column back pressure different?Size of silica gel, uniformity of the particle size and particle shape
SEM photograph - Inertsil® Silica Gel
Why is the particle size distribution different?Every commercially available column has a particle size distribution, but its distribution widths can be quite different.
Particle size, surface area, pore diameter and pore volume arecontrolled by using nitrogen adsorption and laser ray particleanalyzer.
Packings with an uniform size and smooth surfaceshow very low column back pressure.

Inertsil® 4 Series
Inertsil® Series 4 phases
The Inertsil® 4 series is GL Sciencesʼ latest generation of LC columns. This newly developed chemical bonding technology allowsInertsil® ODS-4 to provide high efficiency and good peak shape where competitive columns fail.

Inertsil® ODS-4• Improved peak shapes and heights• Enhanced sensitivity• High resolution• Fast equilibration• Compatible with 100% aqueous eluents
Ultimate deactivation techniques enable the highest inertnesswith this silica based LC column, allowing analysis of traceamounts of the most demanding basic and acidic compounds.
Strictly classified particle size with narrow distribution width andideally designed carbon loading and its procedure allows GLSciences to guarantee the high plate count of +100,000/m.
Combined with the synergistic effect from the improved peakshape based on the high inertness of the packing material,Inertsil® ODS-4 can be used as a tool for enhancing sensitivityand high separation efficiency.
Excellent peak shapes for both strong acidic andbasic compounds
GL Sciencesʼ newly developed endcapping technique ideallydeactivates the silanol groups, suppressing the adsorption ofcommon basic compounds as well as strong ones, which leadsto sharper peaks.
The new endcapping technique prevents the adsorption of strongacidic compounds as the surface of the packing materialbecomes neutral.
Haloacetic acid is a harmful substance generated when tap wateris chlorinated. With its strong acidity, it is easily adsorbed atbasic sites on the surface of the packing material.
High-sensitivity analysis by LC-MS/MS has become increasinglypopular. A column free of adsorption is required since the adsorp-tion of even a very small amount can adversely affect the preci-sion of such highly sensitive quantitative analysis.
Inertsil® ODS-4 is ideal for LC-MS/MS analysis of strong acids(see Figure 1).
Antihistamines are strongly basic and therefore will easily beadsorbed to the residual silanols on the packing material, result-ing in deterioration of peak shapes.
Generally, in LC analysis of strong basic compounds, highly-con-centrated buffer or ion-pair reagents are introduced to the eluentto prevent adsorption of strong basic samples on the packingmaterial. However, in LC-MS/MS, such eluent conditions are notrecommended and a packing material with superb inertness isrequired (see Figure 2).
Inertsil® ODS-4 prevents the adsorption of strong basic com-pounds without using the above mentioned eluents. This featuremakes Inertsil® ODS-4 an ideal column for LC-MS/MS.

Excellent analysis stability of strong metalchelating compounds
For stable analysis of strong metal chelating compounds, sam-ples were injected repeatedly before the analysis to mask theadsorption active sites on the surface of the packing material oraddition of EDTA to the eluent were required.
Column equilibration was also time consuming.Since the trace amount of metals has been eliminated from thesurface of the packing material, it allows a stable analysis fromthe 1st injection requiring no masking process (see Figure 3).
Tetracyclines are antibiotics broadly used as pharmaceuticals foranimals. The residues in livestock and sea product are a majorissue today.
See Figure 5 (next page) for an inertness comparison onHinokitiol.
High-sensitivity analysis by LC-MS/MS has become a large fieldof interest. As tetracyclines have a very strong chelating charac-teristic, it has been anticipated that the adsorption on the columndeteriorates quantitative precision.
Inertsil ODS-4 is an ideal column for LC-MS/MS analysis ofchelating compounds since it has completely removed metalsfrom the surface of the packing material.
Excellent analysis stability under 100% aqueouseluents
Under the condition of a mobile phase containing very littleorganic solvent, the mobile phase can easily come out frompores in the packing material, resulting in the dewettingphenomenon. As shown in Figure 4 (next page), such waterrepellency makes the analysis results unstable over the reten-tion time. Inertsil® ODS-4, with an ideal chemical processing, minimizes such dewetting phenomenon, achieving reliableelution with 100% aqueous eluents
Inertsil® ODS-4 minimizes the dewetting phenomenon whilemaintaining the same retention characteristics to that of com-mon ODS columns. Therefore, Inertsil ODS-4 can be used atease under 100% aqueous eluents.
Packing material that achieves a stable analysis using 100%aqueous eluents provides another advantage. There isreduced equilibration time returning to an initial eluentcondition with a gradient separation.

InertSustain™ C18Introducing a radically new type of silica
It was not possible to end-cap 100% of residual silanols using tra-ditional chemical modification procedures.GL Sciences studied the possibility of developing a radically newtype of silica, a silica that would provide both high intertness(base deactivation) and durability at a wide range of pH.
Our focused R&D effort lead to the development of “EvolvedSurface Silica”, in which adsorption sites and silanol distributionare strictly controlled.
This process enables delivery of an HPLC column with the mostadvanced combination of base deactivation and chemical com-patibility (see Figure 1).
The introduction of Evolved Surface Silica and our cutting-edgechemical bonding technology make InertSustain™ C18 compati-ble with100 % aqueous mobile phases, while maintaining strongnon-polar retentivity. InertSustain™ C18 offers superb separationpower with unmatched inertness to virtually any type of analyte.

• Excellent reproducibility• Exceptional stability and column lifetime• Low column back pressure• Ultra pure and rigid silica• Rigorous physical property tests• High inertness
High reproducibility with longer column lifeRigorous quality control of the physical properties of the basesilica, proprietary bonding and endcapping chemistry, as well asexcellent chromatographic tests for inertness and selectivity, contribute to the production of columns with good reproducibilityand long column lifetimes. The inherent lower column back pressure of Inertsil® ODS-3enables a 5µm, 250mm x 4.6mm column to be operated below10Mpa even when using water/methanol as the mobile phase.
Rigorous tests for base silicaInertsil® base silica is manufactured using the purest reagentsavailable, in a process that excludes contaminants during the for-mation of the silica bead structure. The resulting base silica isultra pure and shows improved efficiency and sample recoveryfor samples that interact with metals. The silica particles are rigid and have a smooth surface, pre-venting structural fragmentation when operated at pressures inexcess of 30Mpa.To ensure extremely low metal levels of the silica, analysis byAtomic Emission and ICP is applied to every batch. ScanningElectron Microscopy is also used for the assurance of the surfacesmoothness. Particle sizes are measured by NitrogenAdsorption and Laser Ray Particle Analyzer. Every batch of syn-thesized silica gel is subject to rigorous inspection, and onlythose batches meeting the strict specifications are used for thesubsequent bonding procedure.

INERTSIL® (GL Sciences)
High inertnessIt is well known that residual silanol groups on the surface of thesilica gel will cause severe peak tailing and adsorption, especial-ly with nitrogen-containing compounds. In the production ofInertsil® ODS-3, the silanols are eliminated through a proprietarybonding-endcapping technology developed at GL Sciences. Toensure maximum inertness, each batch of Inertsil® ODS-3 mustpass a new quality control program using 29Si NMR.The 29Si NMR spectrum (Fig. 1 next page) shows that isolatedsilanols on Inertsil® ODS-3 packings have been completely elim-inated, thereby ensuring excellent inertness and optimum perfor-mance of the phase. There are many new generation ODSphases on the market which still show residual silanols using thistest.Inertsil® ODS-3 shows excellent inertness for both basic andacidic compounds. In addition, chelating compounds also elutewith good peak shape owing to the high purity of the base silica. As typical examples of these compound types, pyridine, ethyl-aniline, benzylamine and aminopyridine (basic compounds),formic acid and acetic acid (acids) and oxine copper (chelatingcompound) are generally employed for QC tests. Good peakshapes are obtained on Inertsil® ODS-3 columns. Conversely, there are many columns on the market which are referred to asnew generation columns but show significant tailing for thesecompounds (see Fig. 2).
High durabilityA long column lifetime is one of the most important factors con-tributing to cost efficiency of an analysis. In general, routine usein the pH range of 2 to 7.5 is recommended for silica-based pack-ings. The durability of Inertsil® ODS-3 at pH 2 and pH9 has beentested.
The graph below shows the variations in retentivity of naphtha-lene on Inertsil® ODS-3 compared with other ODS columns.Inertsil® ODS-3 was stable at pH 2 (0.1% TFA) over 500 hours.

• C18 with polar embedded group• Compatible with 100% aqueous eluents• Alternative selectivity to C18• Ideal for LC-MS
Inertsil® ODS-EP contains a polar functional group embeddedbetween the silica surface and the C18 group. The embeddedpolar group makes the C18 phase stable in 100% aqueous elu-ents without “phase collapse”. This phase is also extremely basedeactivated and provides superior peak shape for acids andbases in organic eluents as well as acidified eluents typicallyused in LC-MS.

Inertsil® HILICHydrophilic Interaction Chromatography is a variation of normal-phase mode where a high concentration of non-polar solvent isused in a mobile phase. It is an ideal method for separating andretaining highly polar analytes unretained by traditional reversed-phase mode. HILIC columns become widely used for criticalseparations in demanding drug metabolism, drug discovery andcombinatorial chemistry. In addition, volatile mobile phases areused for HILIC mode and afford enhanced ESI-MS sensitivityresulting in lower limits of detection.
Application for HILIC mode- Amino acids- Water-soluble vitamins- Drug metabolites and drug discovery- Combinatorial chemistry- Promoted retention of compounds unretained by reversed-
phase mode
Advantages of HILIC mode- Low column back pressure analyses can be performed due to
high organic solvent composition- A high organic solvent concentration of the mobile phase will
lead to a high sensitivity LC-MS analysis- The benefit of less ion suppression effect in LC-MS analysis- Precision determination for polar compounds due to the fast
elution of hydrophobic contents

Inertsil® ODS-Sprint• Well balanced retention of both polar and
non polar compounds• Superior inertness and base deactivation• Compatible with completely aqueous or neat
organic eluents• Low operating back pressure• 20mm x 2.1mm column with new Sprint HG
hardware is ideal for rapid LC-MS separations
Inertsil® ODS-Sprint is an important new addition to the Inertsil 3column series. Inertsil ODS-Sprint is extremely base deactivatedand optimally bonded to retain polar compounds without exces-sive retention of non-polar compounds. This allows you toachieve improved separations faster.ODS-Sprint operates at low back pressure even at high flowrates, shows outstanding longevity and is reproducible fromcolumn to column and batch to batch. To complement this new phase, GL Sciencesʼ has created azero-dead-volume column hardware format, known as Sprint-HGhardware, further enhancing peak symmetry. For additional information, please contact Canadian LifeScience.
Moderate retentivityGL Sciencesʼ newly developed chemical bonding technologyallows Inertsil® ODS-Sprint to retain hydrophilic compounds, butelutes hydrophobic compounds faster. It offers 1/2 of Inerstil®ODS-3 retention for hydrophobic compounds (see Figure 1).
Short equilibration timeA highly uniformed low carbon loading ODS enables shorterequilibration time for gradient analysis and can be used for high-throughputs. The time for purging and cleaning the column canbe reduced in isocratic analysis. In Figure 2, Inertsil® ODS-SP shows a complete equilabrationafter 10 mins with stable retentivity.

Inertsil® ODS-SprintSharp peak shapeSharp peak shapes can be observed due to GL Sciencesʼ newly-developed bonding technology and column hardware.3µm packed columns are also available for further sharp peakshapes.
Inertsil® ODS-sprint does not show any tailing with complete baseline separation, resulting in excellent quantitative determination.Peak tailing of large volume compounds (unreacted derivatizingreagent) disturbed the detection of the target compound onCompetitor E, F and G columns (see Figure 3).
No adsorption of peaksGL Sciencesʼ intensive endcapping is performed to eliminateresidual silanols, creating an exceptionally inert column forreverse-phase chromatography. Excellent peak shapes for awide range of polar analytes, including both basic and acidiccompounds, can be obtained.Inertsil® ODS-Sprint shows excellent peak shapes of basic andacidic compounds. Other competitorʼs columns are showing tail-ing peaks of both compounds. Some columns may be able toanalyze basic compounds, but show peak tailing for acidic com-pounds (see Figure 4).

The Inertsil® WP300 series of columns has been developed forthe analysis and purification of proteins and peptides.
High durability under TFA eluent conditionsInertsil® WP300 Series shows high durability against acidic elu-ents, including TFA solutions (see Fig. 1).Protein and peptide analyses can beperformed using formic acid as a modifierExceptional inertness of Inertsil® WP300 Series prevents irre-versible adsorption of proteins under formic acid eluent condi-tions, providing optimum conditions for LC-MS.
Column choice depends on target ofmolecular weightInertsil® WP300 C8 and C18 show high separation efficiency forpeptides and proteins of medium molecular size.Inertsil® WP300 C4, having high partition efficiency for veryhydrophobic proteins, elutes them with sharp peak shapes.
Inertsil®WP300 C4Recovery of proteins with highhydrophobicityOn Inertsil® WP300 C4 you can get a linear recovery under diffi-cult eluent conditions. Three typical proteins with high hydropho-bicity show good linear recovery (see Fig. 1).Inertsil®WP300 DiolInertsil® WP300 Diol is designed for GFC collection and purifica-tion of proteins (see Fig. 2).
