Ultimate deactivation techniques enable the highest inertnesswith this silica based LC column, allowing analysis of traceamounts of the most demanding basic and acidic compounds.

Excellent peak shapes for both strong acidic andbasic compounds

GL Sciencesʼ newly developed endcapping technique ideallydeactivates the silanol groups, suppressing the adsorption ofcommon basic compounds as well as strong ones, which leadsto sharper peaks.

The new endcapping technique prevents the adsorption of strongacidic compounds as the surface of the packing materialbecomes neutral.

Haloacetic acid is a harmful substance generated when tap wateris chlorinated. With its strong acidity, it is easily adsorbed atbasic sites on the surface of the packing material.

High-sensitivity analysis by LC-MS/MS has become increasinglypopular. A column free of adsorption is required since the adsorp-tion of even a very small amount can adversely affect the preci-sion of such highly sensitive quantitative analysis.

Inertsil® ODS-4 is ideal for LC-MS/MS analysis of strong acids(see Figure 1).

Antihistamines are strongly basic and therefore will easily beadsorbed to the residual silanols on the packing material, result-ing in deterioration of peak shapes.

Generally, in LC analysis of strong basic compounds, highly-con-centrated buffer or ion-pair reagents are introduced to the eluentto prevent adsorption of strong basic samples on the packingmaterial. However, in LC-MS/MS, such eluent conditions are notrecommended and a packing material with superb inertness isrequired (see Figure 2).

Inertsil® ODS-4 prevents the adsorption of strong basic com-pounds without using the above mentioned eluents. This featuremakes Inertsil® ODS-4 an ideal column for LC-MS/MS.

Excellent analysis stability of strong metalchelating compounds

For stable analysis of strong metal chelating compounds, sam-ples were injected repeatedly before the analysis to mask theadsorption active sites on the surface of the packing material oraddition of EDTA to the eluent were required.

Column equilibration was also time consuming.Since the trace amount of metals has been eliminated from thesurface of the packing material, it allows a stable analysis fromthe 1st injection requiring no masking process (see Figure 3).

Tetracyclines are antibiotics broadly used as pharmaceuticals foranimals. The residues in livestock and sea product are a majorissue today.

High-sensitivity analysis by LC-MS/MS has become a large fieldof interest. As tetracyclines have a very strong chelating charac-teristic, it has been anticipated that the adsorption on the columndeteriorates quantitative precision.

Inertsil ODS-4 is an ideal column for LC-MS/MS analysis ofchelating compounds since it has completely removed metalsfrom the surface of the packing material.

Excellent analysis stability under 100% aqueouseluents

Under the condition of a mobile phase containing very littleorganic solvent, the mobile phase can easily come out frompores in the packing material, resulting in the dewettingphenomenon. As shown in Figure 4 (next page), such waterrepellency makes the analysis results unstable over the reten-tion time. Inertsil® ODS-4, with an ideal chemical processing,minimizes such dewetting phenomenon, achieving reliableelution with 100% aqueous eluents

Inertsil® ODS-4 minimizes the dewetting phenomenon whilemaintaining the same retention characteristics to that of com-mon ODS columns. Therefore, Inertsil ODS-4 can be used atease under 100% aqueous eluents.

Packing material that achieves a stable analysis using 100%aqueous eluents provides another advantage. There isreduced equilibration time returning to an initial eluentcondition with a gradient separation.

InertSustain™ C18Introducing a radically new type of silica

It was not possible to end-cap 100% of residual silanols using tra-ditional chemical modification procedures.GL Sciences studied the possibility of developing a radically newtype of silica, a silica that would provide both high intertness(base deactivation) and durability at a wide range of pH.

Our focused R&D effort lead to the development of “EvolvedSurface Silica”, in which adsorption sites and silanol distributionare strictly controlled.

This process enables delivery of an HPLC column with the mostadvanced combination of base deactivation and chemical com-patibility (see Figure 1).

Rigorous tests for base silicaInertsil® base silica is manufactured using the purest reagentsavailable, in a process that excludes contaminants during the for-mation of the silica bead structure. The resulting base silica isultra pure and shows improved efficiency and sample recoveryfor samples that interact with metals.The silica particles are rigid and have a smooth surface, pre-venting structural fragmentation when operated at pressures inexcess of 30Mpa.To ensure extremely low metal levels of the silica, analysis byAtomic Emission and ICP is applied to every batch. ScanningElectron Microscopy is also used for the assurance of the surfacesmoothness. Particle sizes are measured by NitrogenAdsorption and Laser Ray Particle Analyzer. Every batch of syn-thesized silica gel is subject to rigorous inspection, and onlythose batches meeting the strict specifications are used for thesubsequent bonding procedure.

High inertnessIt is well known that residual silanol groups on the surface of thesilica gel will cause severe peak tailing and adsorption, especial-ly with nitrogen-containing compounds. In the production ofInertsil® ODS-3, the silanols are eliminated through a proprietarybonding-endcapping technology developed at GL Sciences. Toensure maximum inertness, each batch of Inertsil® ODS-3 mustpass a new quality control program using 29Si NMR.The 29Si NMR spectrum (Fig. 1 next page) shows that isolatedsilanols on Inertsil® ODS-3 packings have been completely elim-inated, thereby ensuring excellent inertness and optimum perfor-mance of the phase. There are many new generation ODSphases on the market which still show residual silanols using thistest.Inertsil® ODS-3 shows excellent inertness for both basic andacidic compounds. In addition, chelating compounds also elutewith good peak shape owing to the high purity of the base silica.As typical examples of these compound types, pyridine, ethyl-aniline, benzylamine and aminopyridine (basic compounds),formic acid and acetic acid (acids) and oxine copper (chelatingcompound) are generally employed for QC tests. Good peakshapes are obtained on Inertsil® ODS-3 columns. Conversely,there are many columns on the market which are referred to asnew generation columns but show significant tailing for thesecompounds (see Fig. 2).

High durabilityA long column lifetime is one of the most important factors con-tributing to cost efficiency of an analysis. In general, routine usein the pH range of 2 to 7.5 is recommended for silica-based pack-ings. The durability of Inertsil® ODS-3 at pH 2 and pH9 has beentested.

The graph below shows the variations in retentivity of naphtha-lene on Inertsil® ODS-3 compared with other ODS columns.Inertsil® ODS-3 was stable at pH 2 (0.1% TFA) over 500 hours.

Inertsil® ODS-P is a polymerically bonded phase which isdensely bonded with octadecylsilane groups. It shows uniqueseparation capabilities and offers an alternative selectivity toInertsil® ODS-3. Inertsil® ODS-P is particularly suited to the sep-aration of polycyclic aromatic hydrocarbons, and shows addi-tional retentivity for planar compounds.

Figure 2 shows the separation of components from toad venom.Figure 3 shows the separation of terphenyl isomers on the threetypes of ODS phases. On the polymeric Inertsil® ODS-P phase,the planar p-terphenyl is retained longer than the non-planaro- and m- isomers.

Inertsil® HILICHydrophilic Interaction Chromatography is a variation of normal-phase mode where a high concentration of non-polar solvent isused in a mobile phase. Inertsil® ODS-SprintSharp peak shapeSharp peak shapes can be observed due to GL Sciencesʼ newly-developed bonding technology and column hardware.3µm packed columns are also available for further sharp peakshapes.

Inertsil® ODS-sprint does not show any tailing with complete baseline separation, resulting in excellent quantitative determination.Peak tailing of large volume compounds (unreacted derivatizingreagent) disturbed the detection of the target compound onCompetitor E, F and G columns (see Figure 3).

No adsorption of peaksGL Sciencesʼ intensive endcapping is performed to eliminateresidual silanols, creating an exceptionally inert column forreverse-phase chromatography. Excellent peak shapes for awide range of polar analytes, including both basic and acidiccompounds, can be obtained.Inertsil® ODS-Sprint shows excellent peak shapes of basic andacidic compounds. Other competitorʼs columns are showing tail-ing peaks of both compounds. Some columns may be able toanalyze basic compounds, but show peak tailing for acidic com-pounds (see Figure 4).

Sulfa C18Analysis of antibiotic drugs and drug residues in products of ani-mal origin has become an important element of ensuring foodsafely.Inertsil® Sulfa C18 is suitable not only for the analysis of sulfadrugs but also for the screening of synthetic antibacterial agentswhich are directed in the food safety investigative guidelines.

The Inertsil® Diol phase is a dihydroxypropyl bonded silica whichcan be used in both reversed-phase and normal-phase modes.The retentivity is higher than that of silica in normal-phase mode.Figure 1 shows the analysis of free amino acids on an Inertsil®Diol column.

The new Inertsil® Peptide C18 column was developed for theanalysis of Peptide mapping. 100Å pore size high purity silicagels are bonded with octadecyl groups and the residual silanolsare drastically reduced. Metabolic analysis to Peptide analysisare performed with great reproducibility on Inertsil® Peptide C18.Figure 2 shows the application of this column for the analysis ofcytochrome C tryptic digest.
