The Sysmex XS-1000i AutomatedHematology System utilizes thepower of fluorescent flow cytometry(FFC) and hydrodynamic focusingtechnologies. Using a unique,stable, avalanche photodiode witha laser bench, Sysmex fluorescentflow cytometry provides the sensitivityneeded for measuring and differentiatingcell types in whole blood.
Hydrodynamic Focusing(DC Detection)
Fluorescent technology andhydrodynamic focusing enablethe analyzers to consistently classifynormal WBC (white blood cell),RBC (red blood cell), and PLT(platelet) populations from abnormalpopulations, thereby decreasing thenumber of manual interventions.

RBCs and PLTs
RBCs and PLTs are counted in adedicated channel using Impedanceor Direct Current (DC) detectionmethod combined with hydrodynamicfocusing technology. Challenges tocell counting such as coincidence orrecirculation are circumvented andautomatic discriminators separatethe two cell populations.
Even with samples at extremelylow or unusually high concentrations,the Sysmex system analyzes RBCsand PLTs with uncompromisedprecision and accuracy.
High Quality Hemoglobin(HGB) Analysis
The XS-1000i utilizes the non-cyanidereagent, Sodium Laurel SulfateHemoglobin (SLS). The end productis a colored compound that ismeasured spectrophotometrically.Since hemoglobin determinations areperformed from a dilution and in itsown separate chamber, there is nointerference from high WBC counts,lipemia or abnormal proteins.
Direct Hematocrit(HCT) Measurement
The cumulative pulse heights of allthe RBC counts yield the HCT. This isbased on the principle that the pulseheight (voltage change) produced bycells passing through the aperture isproportional to cell volume.

The combination of side scatter(cell complexity), forward scatter(size) of nucleated cells provides aconcise and precise image of eachdetected peripheral blood cell.
This 3-dimensional blood cell analysisprovides unique accuracy and precision.Fluorescence labeling of peripheralblood cells is a milestone for theroutine leukocyte differential.
Fluorescent technology enablesthe XS-1000i to reliably differentiatenormal WBC populations fromabnormal WBC populations. Thesensitivity of the unique applicationof fluorescent flow cytometry givesthe lab a high level of confidence inreporting accurate WBC differentials,even on critical patient samples whenthe WBC count is low.
