Syringe filters are used for filtration of suspended matter from liquid sample or gases. Rapid purification and removal of particles is very simple: just place the filter on a syringe, and you are ready for filtration. Contamination of sensitive instrumen-tation by solid impurities can be avoided. Also the lifetime of chromatographic columns and equipmend can be increased.

Solid-phase extraction (SPE) is a powerful method for sample preparation that is used today in most laboratories. 5. Thin layer chromatography (TLC)

The principle of TLC is known for over 100 years. TLC is still a popular technique in the laboratory for reaction, quality and identity control.

Advantages of TLC are:

• High sample throughput possible in a short time

• No complex sample preparation required

• Simple method without expensive instruments

IMPLEMENTATION OF A TLC SEPARATION SELECTION OF THE SOLVENT:The solvent is selected by elution strength. The most appropriate solvent can be determined very easily.

SAMPLE APPLICATION:A common method is to use a glass capillary to place a point or a short line on the TLC plate. The placement of the substance has to be narrow, because, if the point or line is too broad this leads to a poor separation, since the zones become broader while processing. After application, the solvent is allowed to evaporate, this can be accelerated by the use of a heat gun.

DEVELOPMENT:A common method is to use a developing chamber. The eluent is filled into the bottom of the chamber. For a reproducible running distance, the saturation of the chamber with the running medium vapor is necessary. Then the TLC plate with the sample is positioned in the chamber. The separation is completed when the eluent has ascended to the upper end of the TLC plate. The TLC plate can now be removed and dried accordingly.

EVALUATION:The evaluation depends on the aim of the chromatographic analysis. In the case of qualitative determinations, it must be allowed to run alongside the simplest comparative substances. Quantitative evaluations are possible via appropriate calibration measurements. In this case, the area of the substance spots is used or a photometric evaluation is carried out on the layer. In the case of the latter, however, the complexity of the apparatus is greater.

6. Tablet Dissolution AccessoriesCANNULA FILTER: Filters are important to hold back particles of the tablet. All filters are USP compliant. By using cannula filters the hydrodyna-mics in the vessel may be affected, advantage is the large surface area. If high particulates are present blockage of sample probes can be prevented, due to the high flowrate. There are different porosity ranges available. For different manufacturers different filters are recommended, they are not interchangeable due to the differences in diameter. For some manufacturers also filter discs or filter tips are available.

7. Sampling Cannulae

Sampling position: Important for test with baskets, it is different for 500 ml and 900 ml tests.

Cannula length: The cannula varies in length depending if it is a 500 ml or 900 ml test.

Cannula Material: They are made from stainless steel or PEEK, in order to guarantee that there is no adsorbtion or interference with the active ingredient. Stoppers: Stoppers can be used to make sure that the sample is always been taken from the same depth. The stoppers are attached to the cannula and they sit in a hole in the vessel cover.

STANDARD GLAS VESSELS DISTEK

High quality vessels are important to obtain reproducible and correct results. They are the key element of the dissolution test. They have to meet the dimensions of the USP, also the bottom should be perfectly rounded without obvious efects of the glass on the inside. The thickness of the wall has to be even and reproducible. Vessels associated for different dissolution baths can vary in dimensions, although the dimensions are specified by the USP. The tolerances are quite wide. In general standard vessels are used, there are also Ultracenter Precision Vessels available.

Glass vessels have the advantage of good thermal conductivity. They have a high resistance to scratching which means they don t have to be replaced so often. Amber vessels are used if light sensitive analytes are tested. They are available in glass and plastic.

AGILENT COMPATIBLE PADDLES

Paddles are used for the Apparatus 2, commonly known as the paddle method. If it is important to get consistent results from vessel to vessel the dimensions and tolerances of the paddle can be critical. According to USP the paddle has to rotate smoothly without significant wobble. No sharp edges in the contours of the paddle blade wanted. They could produce a turbulent instead of a laminar flow pattern.

Types:

• Electropolished Stainless Steel they are strong and resistant to bending. Surface impurities are removed by electropolishment, they are resitstant to corrosion.

• PTFE coated: Paddles are coated according to the suggestion of the USP to enhance inertness.

• Solid Fluoropolymer: a single piece molded design assures reduced wobble.

10. Baskets

The basket method was the first official method. This method is now named Apparatus 1. The metallic shaft is connected to the cylindrical basket. The basket is positioned inside the vessel, kept at a constant temperature. The vessel solution is stirred smoothly by the rotating basket. Normally the USP method required a 40 mesh screen unless otherwise specified.

Especially in the case of headspace caps, it is im-portant not to over-crimp these. If the aluminum is pulled too far under the

rolling edge, the stops of the breaking lines sufferfrom too much pressure and break even at a low internal pressure of the

cylinder (less than 3 bar). In the worst case, they tear apart even next to

the breaking lines.

17. Guard columns by Altmann AnalytikIf you would like to protect your column by adding a guard column cartridge, you also always need a guard column holder. The following diagrams show the connection possibilities for a guard column to an HPLC column (column connectors have to be ordered separately):

The series of tungsten halogen lamps are specifically for use in analytical applications. Special quartz envelopes enable high transmission below 380 nm and the position of the tungsten filament is closely controlled through precise alignment. The lamps offer high colour temperature, luminous efficacy and long life. Tungsten lamps are filled with a halogen gas mixture specific to their final application and range from 5 W – 200 W, with typical lifetimes of more than 2,000 hours.

Hollow cathode lamps (HCL) are discharge lamps designed for use in Atomic Absorption (AA) instruments. They consist of a cathode made from the element of interest, an anode and an inert filler gas contained in a glass envelope. 32. Cuvettes

It is particularly important for a problem-free operation with cuvettes that the light can only penetrate the interior. Therefore, the light beam cross-section should always be selected appropriately and the cuvette must be exactly aligned in the holder. The light beam cross-section must be selected to be smaller than the inner width. This ensures that neither the side walls nor the ground are touched by the light beam.

In photometry, the macrocuvette has largely established itself as a standard. It is defined as follows:

• The designation macro cuvette applies according to DIN58963, if the inner space is wider than 5 mm in a stretch cuvette. • For a cylinder cuvette, the inside diameter must be over 8mm, so that it can be counted in this category.

QS: QS is the identification logo which indicates that Quartz of the highest purity and homogeneity has been used for the window. Because it is produced from a silicon compound, it is also called synthetic Quartz. Hellma use Quartz SUPRASIL from Heraeus Quarzglas GmbH, with which gives transmission values of more than 80 % over a spectral range of between 200 nm and 2500 nm for an empty cell.

OS: OS indicates that SCHOTT type UK 5 is used for cells referred to in the catalogue as manufactured from “Special Optical Glass”. This crown glass is made from exceptionally pure raw materials, which gives an improved transmission in the near ult-raviolet range. An empty cell will give a transmission of more than 80 % over a spectral range of between 320 nm and 2500 nm.

For example, it will be very difficult to prime a 10 µL syringe if the dead volume in the needle is greater than 10 µL. NEEDLE POINT STYLE:Several different needle point styles are offered on Hamilton syringe and needle products depending on the application. The correct point style varies depending on the intended application. Below are a few examples of common applications.

Manual GC Injections: Historically a Point Style 2 was required to achieve efficient septum penetration with minimized coring. With Hamilton’s new line of GC septa there is evidence that indicates the Point Style AS will consistently extend septa life by as much as 10-fold.

Manual HPLC Injections: Most manual injection valves are designed to be used with a 22 gauge Point Style 3 needle.

Drawing Sample: Any point style will efficiently draw a sample, but when it is important to get the last drop out of a vial use Point Style 3 or AS to reach all the way to the bottom.

Animal Injections: Animal injections require the sharpest needle possible. The recommended needle point style for this application is a Point Style 4 at a 12° bevel. For applications where injection site depth is critical or the target is small, a more vertical bevel as shown below, such as a 30 or 45°, is suggested.

Point Style 2: Sharp, beveled, curved, non-coring needle point, recommended for septum penetration

Point Style 3: Blunt needle point for use with HPLC injection valves and for sample pipetting

Point Style 3T: Blunt needle point for thin layer chromatography (TLC) applications. Needles are coated with PTFE ¾“ up from the needle tip.

Point Style 4: Standard 12° beveled needle point is recommended for life science applications.

Point Style 5: Conical needle with side port for penetration of septa, thin-gauged vinyls and plastics without coring.

AS: Special conical style needle point used on autosampler syringes the non-coring needle point is recommended for septum penetration.

GASTIGHT 1000 SERIES SYRINGES:The 1000 series syringes are a mid volume gastight syringe. This series of syringes is ideal for dispensing volumes from 100 μL up to 100 ml. A variety of different terminations and needle options are available in this series.

If the syringe is used manually, then a manual syringe is sufficient if it is to be installed in an instrument. Make sure that the selected syringe matches the instrument. Syringe size: For good reproducibility and accuracy, no more than 10 % of total injection capacity should be injected.

MANUAL OR AUTOSAMPLER SYRINGES:

Fixed needles are the first choice for experienced users or analyzes in the trace area. Fixed needles are suitable for auto-samplers where there is little risk that the needle will bend. Solid needles are very easy to handle. Interchangeable needles can save costs over time because, with bent needles, the needles can be replaced separately. The needles can also be exchanged for different applications.

PERMANENT OR REPLACABLE NEEDLES:

Beveled:

Is the first choice when manually inserted into a septum. Beveled tips are also known as Point Style 2.

Cone:

This type of tip is specifically designed for multiple injections or when the lifetime of the septum in the autosampler is to be extended. The conical shape effectively perforates the septum during the piercing process without cutting it in. Conical need-les are also known as Point Syle AS.

HPLC:

These straight needles are used in HPLC and are also known as Point Style 3.

Side Hole:

The samples are injected through the side hole to prevent clogging of the needle. This shape is ideal for large-volume gas injections and is also known as Point Style 5 needles.

Sheathed bevelled needle:

This needle corresponds to the chamfered needle, except that the needle is still strengthened by a sheath. These needles are also known as Point Style 4 needles.

DIFFERENT TYPES OF NEEDLES:

Metal Plunger:

These plungers can not be exchanged, stainless steel plungers with special fitting into the injection body. Standard for chromatographic applications. Used for injection volume between 5 and 500 μl. Plunger with PTFE Tip:

Can be used for gaseous and liquid injections. The plunger is interchangeable, easy to remove. Used for injection volumes between 10 μl and 100 ml. Conical needles are also known as Point Syle AS.

Flexibler Plunger:

Very robust plunger, ideal for student use. Used for injection volumes between 5 and 10 μl.

The ferrule is a small, sealing connector within the GC system. Often underestimated, however, the ferrule is of central importance. On the other hand, the ferrule ensures that the GC column is held securely and correctly positioned in the GC apparatus. Ferrules are available in different sizes, materials and shapes and can thus be optimally adapted to the respective GC-Column.
