Zoology Vertebrate Destructive Sampling Request

The Denver Museum of Nature & Science has specific protocols for destructive sampling of vertebrate and parasite specimens for genetic, stable isotope, and other analyses. This material may include and is not be limited to frozen or fluid tissues, bone, teeth, feathers, hair, dried skin, and muscle. Such material represents a finite resource that is expensive to obtain and house, thus will be provided only to qualified researchers following the guidelines and requirements listed below.

Request for destructive sampling will be evaluated on a case-by-case basis by the curator of the collection. o For mammal, amphibian and reptile, and parasite collections: John Demboski o For bird (including egg) collections: Garth Spellman

Requests for destructive sampling must contain compelling reasons why the project cannot be completed without the use of Museum specimens. Requested samples are intended to reasonably supplement research materials obtained from other sources such as field sampling of extant taxa. If material from historic specimens (e.g., 100-yr old study skins) is being requested, the research question being addressed must explicitly state why the use of those specimens are required, or that the taxa cannot be sampled directly from the field. In addition, the rarity and ability to replace the samples might be considered as well.

Requests for destructive sampling should provide evidence that the investigators have experience with the proposed analytical procedures (e.g., DNA extraction, sequencing, isotope analyses, or working with historic material), and that the proposed project will likely generate usable data given destructive sampling of specimens.

Unless other arrangements are made, the actual sampling will be performed by Museum staff.

Should the loan be approved, a Federal Express account number should be provided to cover shipping charges (and dry ice if needed) unless other arrangements are made.

Specimens used in publications, reports, or presentations should be included in a "Specimens Examined" section and listed in the following formats:

Mammals: ZM.XXXXX

Birds (non-egg): ZB.XXXXX

Eggs/Nest: ZEN.XXXXX

Herps: ZAR.XXXXX

Parasites: ZP.XXXXX

DNA sequences obtained from Museum specimens should be submitted to GenBank so that they are accessible to other researchers. These sequences must be referenced to the DMNS catalog number in the following triplicate format: DMNS:MAMM:XXXXX or DMNS:BIRD:XXXXX in the “specimen-voucher” modifier from the pull-down menu. Please contact us about this format as this allows our database Arctos to talk to GenBank. See an example here: https://www.ncbi.nlm.nih.gov/nuccore/NC_032373.1

The Denver Museum of Nature & Science shall be acknowledged in any publications that result from the use of its specimens. Acknowledgement should be given as "Denver Museum of Nature & Science” and an electronic copy (PDF) should be sent to us.

Material is non-transferable to 3rd parties without prior permission and unused portions of tissues and resulting products (e.g., DNA extracts) must be returned to the Museum, unless other arrangements are made.

Student visitors, please ask your advisor to email us to provide a brief letter of introduction that vouches for your ability to work responsibly in museum collections.
