Capillary Electrophoresis Technology


Technology

Electrophoresis is a separation technique that is based on the differential migration of charged compounds in a semi-conductive medium under the influence of an electrical field. When using capillary electrophoresis (CE) or capillary zone electrophoresis (CZE,) separation is done inside a narrow bore capillary (with an internal diameter between 20 and 100 µ).

The capillaries are made of fused silica and filled with an electrolyte or gel. For high separation efficiency the current may be as high as 30 kV. A cooling system will remove excessive Joule heating. Detection is performed online on the capillary with an UV, LIF (Laser Induced Fluorescence), or CCD (Contactless Conductivity Detection) detector. CE is also connected to MS.

The mean characteristics of Capillary Electrophoresis are: simplicity, selectivity and low cost. Furthermore, Capillary Electrophoresis offers higher efficiency than HPLC and thus greater resolution power for separating various components. It is recognized as an ideal method to separate proteins but also small molecules.
