Single Molecule Imaging and Nanometry: Fluctuations and Function of Life Since biological molecular machines such as molecular motors, cell signal processors, DNA transcription processors and protein synthesizers are only nanometers in size and have a flexible structure, they are very prone to thermal agitation. Furthermore, the input energy level is not much different from that of average thermal energy, kT. Molecular machines can use this thermal noise with a high efficiency of energy conversion for their functions. This is in sharp contrast to man-made machines that operate at energies much higher than thermal noise. In recent years, single molecule imaging and nano-technologies have rapidly been expanding to include a wide range of life science applications. The dynamic properties of biomolecules and the unique operations of molecular machines, which were previously hidden in averaged ensemble measurements, are now being unveiled. The aim of our research is to approach the engineering principle of adaptive biological systems by uncovering the unique operation of biological molecular machines. Here, I review our single molecule experiments designed to investigate molecular motors, enzyme reactions, protein dynamics and cell signalling, and discuss how thermal fluctuations (noise) play a positive role in the unique operation of biological molecular machines allowing for flexible and adaptive biological systems including muscle and brain.

Capillary models for liquid crystal fibers, membranes, films, and drops An overview will be presented of the capillary modeling science of nematic liquid crystals and its applications to the stability, structure, and shape of films, membranes, fibers, and drops. Liquid crystals are anisotropic viscoelastic materials possessing long range orientational order, and hence these models are relevant to the capillary science of anisotropic soft matter. A systematic multiscale approach is used to derive the equations that govern the shape of interfaces and contact lines. These shape equations generalize the surface Laplace and the contact line Neuman equations by introducing long range orientational order, gradient elasticity, surfactant adsorbants, magnetic and electric fields. The thermodynamics of capillary systems is used to reveal novel cross-effects such as adsorption-driven shape changes of surfaces and contact lines. The capillary models are used to analyze the structure and stability of films, membranes, fibers, and drops, of direct relevance to the processing and performance of structural and functional liquid crystals. Novel soft materials and mechanisms analyzed in this paper include: (1) stabilization of freely-suspended nematic films by orientation and molecular order heterogeneities, (2) orientational defects in polymer dispersed liquid crystals films, shown to originate from surface anchoring transitions, (3) electric field-induced curvature in membranes for sensor and actuator applications, (4) new helical morphologies of thin nematic filaments driven by strong interfacial anchoring, (5) tunable partial wetting through contact angle modification via gradient and anchoring elasticity, (6) liquid crystal nanoemulsion shape control through anchoring effects, and (7) magnetic shaping in liquid crystal colloids. Readily accessible applications to biological liquid crystal materials and processes indicate that capillary modeling science will be a most active area of research in the very near future.

Next-Generation Colloids for Therapeutic Delivery: From Assembly to In-Vivo Application Responsive polymers play an important role in the development of particle carriers for biomedical applications and biochemical reactions. This presentation will focus on nano-engineered polymer-based particle delivery systems assembled through the sequential deposition of polymers, and their utilization for the encapsulation, protection and release of oligonucleotides and peptides. The delivery of capsules to antigen presenting cells to stimulate an immune response and the specific binding of antibody-functionalized capsules to cancer cells will be highlighted. The preparation of synthetic hydrogel capsules that possess size and charge exclusion properties to allow for selective permeability of reaction components and facile control over capsule architecture, affording the continuous modification of nucleic acids and the assembly of liposome/polymer composites with a subcompartmentalized structure reminiscent of cells, will also be presented.

Swelling of constrained hydrogels: New approaches to active surfaces and polymer micro-objects The stresses developed when soft solids are swelled in the presence of constraints can lead to a variety of different elastic shape instabilities, depending on the constraining geometry. In one example, we study the surface 'creasing' instability of thin layers confined in two-dimensions by attachment to a rigid substrate. We have verified that the conditions for instability are quite insensitive to both material properties and layer thickness, in agreement with simple predictions. Using temperature-responsive gels, we seek to elucidate the mechanisms of crease formation and growth, and take advantage of this instability as a way to create dynamic polymer surfaces with reconfigurable chemical patterns. In a second example, we consider the behavior of externally unconstrained gel sheets containing laterally varying extents of swelling. A newly developed method for patterning these materials provides access to opportunities for studying the controlled folding and buckling of gel micro-objects.

Matthew E. Helgeson obtained his B.S. in Chemical Engineering at Carnegie Mellon University, Pittsburgh (2004) and PhD in Chemical Engineering at the University of Delaware (2009) under the supervision of Profs. He is presently a Postdoctoral Associate at the Department of Chemical Engineering at Massachusetts Institute of Technology (MIT). His research interests involve the design and characterization of self-assembling colloidal mixtures with tailored meso-scale structure and rheology, along with the development of novel fluidic-based approaches for the synthesis of functional and stimuli-responsive colloids and nanomaterials. Prior to winning the Victor LaMer Award (2011), he received several prizes and awards, among which the Excellence in Graduate Polymer Research of the ACS POLY division (2007), the Neutron Scattering Society of America prize for outstanding student research (2008) and the Center for Composites Materials, Roy L. Temporary (and not so temporary) networks from colloids in self-assembling media Self-assembling surfactant and polymer solutions encounter colloidal species (nanoparticles, emulsions, proteins, etc.) throughout their formulation, processing, and application. In such mixtures, complex interactions at the solution-colloid interface can result in dramatic changes in behavior of the bulk fluid. Thus, understanding these interactions represents both a challenge toward successful product design as well as an opportunity for the discovery of new materials in venues ranging from petroleum production to nanotechnology to human health. This talk will highlight how characterization of interfacial interactions and self-assembly leads to unique control over the rheology and stability of a variety of colloids in self-assembling media. First, I will discuss mixtures of wormlike micelles (WLMs) and model nanoparticles, in which interactions between micelles in the bulk and adsorbed surfactant monolayers give rise to reversible micelle-nanoparticle junctions. These junctions affect the rheology of WLMs in a number of ways, including increased viscosity, polymer-like viscoelasticity, and suppression of flow instabilities such as shear banding. Furthermore, they allow for dynamic control of the stability of the dispersed nanoparticles through micelle-mediated bridging interactions. I will also discuss a unique class of oil-in-water nanoemulsions, where interfacial self-assembly of a polymeric gelator leads to surprisingly solid-like behavior in an otherwise liquid system. The ability to tune the gelation process, as well as render the fluid polymerizable, through the choice of gelator chemistry allows for the preparation of highly structured “organohydrogels” with widely tunable mechanical and transport properties.
