Cross Reactivity

The term cross-reactivity refers to two distinct antigens that are recognized by the same antibody, thereby eliciting a

similar immune response. Cross-reactivity in allergic reactions occurs when the proteins in one allergen are similar to

the proteins found in other allergen.

Typically, cross-reactivity occurs between species in the same taxonomic genus or family. For example:

Our Scientifi c Affairs Department is available for all of your science questions

Science Hotline: 855-782-9323 (Option 4)

Online helpdesk: https://alkinc.freshdesk.com

ALK strives to improve the scientifi c knowledge base for allergy in the veterinary fi eld.

Many cross-reactivity relationships have been established in the scientifi c literature for human immunotherapy, and

our Scientifi c Affairs department can provide substitution options available for ALK allergens as well as cross-reactivity

charts by request.

TREES

Members of the Betula genus (e.g. White Birch, Red Birch and Black Birch)

are cross-reactive.

WEEDS

Ragweed species (Ambrosia genus) are highly cross-reactive, and exhibit

moderate cross-reactivity with other members of the Asteraceae family (e.g

Mugwort, Marhshelder, and Cocklebur).

GRASSES

Timothy grass is expected to be cross-reactive with Pooideae subfamily,

including Orchard, Sweet Vernal, and Perennial Rye grasses.

MOLDS

Molds containing the same genus name (e.g. Aspergillus niger and

Aspergillus fumigatus) are considered cross-reactive, and these molds can

be readily substituted for one another.

Weight to Volume

Allergenic extracts are available commercially in multiple formulations as well as concentrations. All extracts are

initially generated by combining source material (e.g., pollen, mold, etc.) with set volumes of solutions designed

to extract allergenic proteins. Weight-to-volume (w/v) is a manufacturer’s designation that identifi es the amount of

source material and extraction buffer used to make the product. Additionally, w/v extracts may be diluted to a certain

Protein Nitrogen Unit (PNU) and sold at specifi c concentrations. Each formulation has unique advantages, and both

are successfully utilized for immunotherapy nationwide. In order to determine which formulation is best for your

specifi c practice, we have outlined the differences between PNU and w/v extracts below.

Weight-to-Volume (w/v)

Most w/v extracts are generated at the 1:10 w/v aqueous strength. This formulation serves as the mother lot from

which most other extract formulations are derived. Glycerin extracts may be generated two ways: 1) addition of equal

parts of 100% glycerin to the aqueous mother lot, yielding a 1:20 w/v extract; 2) direct extraction using 50% glycerin

solutions. The concentration of w/v extracts is typically much higher than diluted PNU formulations, conferring greater

protein stability for long-term storage. This equates to more stable potency over the lifespan of the product.

Protein Nitrogen Unit (PNU)

PNU was one of the fi rst measurements used to describe extract strength and indicates the concentration of total

protein in a vial. PNU identifi es the total nitrogen present in a sample measured by the Kjeldahl assay, as specifi ed

by the FDA. Nearly all aqueous extracts are subjected to PNU determination as part of FDA-required quality control

measures, therefore this measurement is also used to dilute extracts to specifi c PNU concentrations (i.e. 10,000,

20,000, and 40,000 PNU/mL). These dilutions are useful for formulating immunotherapy dosage by PNU.

There are a few issues to note when using PNU for immunotherapy applications. Not all of the components identifi ed

by the Kjeldahl assay are relevant allergenic proteins, therefore the PNU concentration on the label may overstate

the actual amount of true allergen in the vial. To further complicate the issue, studies have shown that in some cases

not all of an allergen protein is identifi ed by Kjeldahl assay, causing PNU to understate allergenic content. May et al.

reported that only half of the short ragweed in solution was captured by Nitrogen determination1. Because of this, the

FDA does not recognize PNU as a potency measurement and therefore is not a reliable parameter for standardization.

Nevertheless, knowledge of PNU concentration does provide some useful information, and is often utilized as a unit

for therapeutic dosing.

Converting between PNU and w/v Extracts

Transitioning between PNU and w/v extracts is a relatively straight-forward process, but does require two simple

calculations. Before getting started, acquire the PNU value of the w/v extract that will be diluted; identify the desired

(diluted) PNU concentration, and determine the total volume of diluted extract that will be created. Most aqueous

extracts state the PNU concentration on the vial label. For glycerin extracts or those extracts without a labeled PNU

concentration, contact ALK customer service at 800-444-2370 for the historical PNU concentration.

It is important to note that when the desired PNU concentration is greater than the w/v extract PNU concentration,

a dilution cannot be made. For instance, Mountain Cedar 1:10 W/V typically has a PNU concentration below 10,000.

Therefore a 20,000PNU/mL vial cannot be formulated. When issues such as these arise, feel free to contact Medical

Scientifi c Affairs for advice on extract formulation.

ALK Commitment

ALK is committed to helping our customers maintain uniformity of care for their patients. Please do not hesitate to

contact Scientifi c Affairs Team at 855-782-9293 should you have additional questions or concerns regarding a transition

between ALK products.

1. May, et.al JACI 63:89 (1979)

To create a specifi c PNU concentration from a w/v product:

Volume of W/V extract needed = (desired PNU ÷ PNU of W/V extract) X Total vial volume

Divide the desired PNU concentration by the PNU concentration of the w/v extract; multiply the resulting number by the total volume of diluted extract needed.

Example : (10,000PNU/mL desired ÷ 80,000PNU/mL w/v extract) X 10mL = 1.25mL of w/v extract needed

Next, determine how much diluent is needed to dilute the extract above in a new vial:

Volume of diluent needed = Total vial volume – Volume of w/v extract needed (determined above)

Subtract the volume of w/v extract needed (answer from previous calculation) from the total vial volume that will be created (i.e. 10mL in above example).

Example: 10mL – 1.25mL = 8.75mL of diluent

Finally, use the two numbers from the formulas above to create a diluted extract in a sterile empty vial by

combining the w/v extract with diluent

ACTT® Serum Testing

Proven TechnologyThe ACTT® ELISA technology identifi es allergen-specifi c IgE with precision and accuracy. Our affi nity purifi ed,

anti-IgE antibody detects IgE only; it does not bind to IgG. Test scores are not subject to miss-identifi cation or

false-positive due to IgG interference. Our laboratory employs rigorous quality control methods to ensure accurate and

reproducible test results for each patient.

No Steroid InterferenceACTT® Allergy Test results are not affected by steroid use. Your patient does not need to be withdrawn from steroid

or anti-histamine therapy prior to serum allergy testing, making this modality a safe and accurate testing alternative to

intradermal testing.

ACTT® Allergy Testing Services

Successful allergy management begins by identifying all aspects of the animal’s allergy exposure. The ACTT® Allergy Test

offers a comprehensive list of testing options:

The ACTT® ELISA test results are the foundation for a successful immunotherapy treatment program.

Allergen-Specifi c Immunotherapy Treatment

Immunotherapy is the only allergy treatment that actually reverses the natural course of allergic disease, thereby

reducing the body’s sensitivity to allergens. Because every allergy case is unique, we custom-formulate treatment

for each animal, based on species, age, weight, breed and clinical history. This unique feature separates ALK from

other laboratories. Benefi ts of custom treatment include:

• Fewer patient reactions to injections and drops

• Quicker improvement of the patient’s symptoms

• Increased owner compliance

Distinguishing Features of ACTT® Immunotherapy

Careful Allergen Selection

ALK focuses on treating immuno-dominant allergens, which provide cross protection for any allergens in related

families. Allergen cross-reactivity occurs within grass, tree, weed, mold, dust mite and insect families.

Separation of Molds, Mites and Insects

ALK follows the recommendations of the AAAAI for allergen groupings when formulating immunotherapy treat-

ment.

Custom Treatment Protocols

Immunotherapy protocols are custom-formulated according to the species, age, weight, breed and clinical history

of each animal.
