Usefulness and limitations of taste sensors in the evaluation of palatability and taste masking in oral dosage form
Presented by Arantha j joseph
First year MPharm
This is a review taken from Asian journal of pharmaceutical sciences done by TamamiHaraguchi from school of pharmaceutical seience mukogawa womens university japan published on 31 August 2015.
In various types of formulation solid oral forms are most common
Advantages• Dose accuracy .• stability.• modify drug release profile .• delay or sustain therapeutic effect.
Tablets even plain associated with problems like difficulty in swallowing due to
large size. sticking to the throat mucosa . irritation of the pharyngeal region. coughing or choking.
One way of eliminating this is the use of orally disintegrating tablets(ODT)
In ODT acute drug concentration reaches the oral cavity is high due to the immediate disintegration of tablet in saliva. There for taste masking technology must be involved if the active ingredient is bitter or astringent.
o Disintegration time is an essential factor for determining its palatability
Advantages if ODT
Quick disintegration after contact with saliva in mouth.
facilitating swallowing ease of delivary increase patient adherence
Human sensation test is a major method of evaluating palatability
It should be accurate and precise Ethical issues are for testing drugs like
anti neoplastic agents in humans
So we are using artificial taste sensors Insent taste sensing system Alpha-ASTREE electronic tongue
are using.
Insent Taste Sensing System It composed of lipid polymer membrane Insent taste sensing system exhibits
different taste qualities like Bitterness Sourness Saltiness Umami sweetness
Advantages
Highly skilled Each taste sensor membrane response
particular taste
Disadvantages
Some substances are not detected
Antibiotics like clarithromycin dry syp have been evaluated using this system, a good correlation was obtained between the results of human gustatory sensation testing
Suppression of unpleasant taste also can be predicted
ALPHA-ASTREE ELECTRONIC TONGUE It is a quantitative sensor Evaluate overall taste of a product by using
electrodes Consist if 7 liquid cross sensitive electrodes An auto sampler Interface electronic module Each electrodes contain an organic membrane Any interaction at the interface of this
membrane is detected and convert into electric signal
Advantage
Evaluate overall taste Qualitative evaluation Compare elucidation distance Represents similarly in taste
EVALUATION OF OTDTs 3 different apparatus are used for
estimating disintegrated time OTD-101 TRI CORP TESTER OD MATE
Combined taste sensor and apparatus to estimate disintegration time of ODTs The combination of taste sensing system
and an apparatus to estimate disintegration time to predict the Palatability of ODT
Eg: Combination of incent taste sensing and
OD mate Alpha ASTREE electronic tongue an OD
mate
conclusion As per the comparative study the
authors reported that the both systems have their own usefulness and limitations
Although the combination of taste sensing system and an apparatus to estimate disintegration time would be useful for prediction the palatability of OTDs
