TurboSEC Size Exclusion Chromatography Software Introduction TurboSEC is an optional 32-bit software package that provides complete and flexible molecular weight distribution processing and reporting of TotalChrom“ data. TurboSEC methods may be run automatically, as part of a TotalChrom sequence, to generate Size Exclusion Chromatography (SEC) reports and plots at the completion of each data acquisition. In size exclusion, or gel permeation chromatography, there is an intrinsic relationship between the molecular weight of a component and its retention volume. PerkinElmer’s TurboSEC software option for TotalChrom Chromatography Data Systems is a Microsoft® Windows® based SEC processing and reporting application. TurboSEC software is designed to take full advantage of the power inherent in today’s 32- bit operating environments. TurboSEC methods permit calibration against narrow or broad molecular weight standards. TurboSEC methods are linked to TotalChrom data acquisition methods to facilitate automatic generation of the chromatographic run (Figure 1). TurboSEC’s reporting options provide a wide variety of report formats and layouts, complete with a Print Preview option. All standard features of TotalChrom software can be utilized during the collection of data for SEC calculations. Familiar Data Acquisition SEC analyses are acquired via the standard TotalChrom methods. TotalChrom manages all instrument control parameters and data acquisition through methods that are passed to PerkinElmer’s standard interfaces (NCI or LINK) for instrument control and data acquisition. All interfaces incorporate buffered RAM memory to ensure data acquisition or PC interruptions. Since each interface’s memory is segmentable, multiple chromatographic runs may be stored in each interface, if necessary. The multitasking architecture of TotalChrom software permits data acquisition from conventional gas and/or liquid chromatographs during acquisition of SEC data. Figure 1. Intuitive TurboSECMethods

TurboSEC methods contain all theprocessing information needed togenerate an SEC report from aTotalChrom result file. Methodsare easily created via aGenerate/Review feature in theSEC method editor. This approachguides the user through all agesand parameters of a method so thatnothing is overlooked.Alternatively, the user may godirectly to a specificpage/parameter of an existingmethod that might need selectivemodification. Online context-sensitive help is always availableto assist the user as methods arecreated or modified. After youcomplete a TurboSEC method thesoftware automatically creates acalibration plot for your review.Different curve fit equations maybe easily selected to generate thebest fit to your data. Data pointsmay also be annotated with labelsfor clarity. Nothing could be moreintuitive.

Easy Calibration

TurboSEC method calibrations arebased on different polymerstandard types—narrow rangestandards with calibration curve fitchoices of point to point or apolynomial regression fit from firstto ninth order, and broad rangestandards based on the IntegralBroad Standard or Hamielectechnique. The Universal Narrowstandard calibration procedure isalso available. A flow markercomponent may be added tostandards and unknown samples toprovide correction for variations incolumn flow during an analysis.

A spreadsheet table is used to enterelution volumes and correspondingmolecular weights.Retention/elution values may beentered manually or loadedautomatically from an existingTotalChrom result file. Calibrationcurves and fit statistics may bedisplayed on screen or printed(Figure 2). Calibration data may beautomatically updated as needed ifthe TurboSEC method is includedas a user program in theTotalChrom data acquisitioncalibration run. Nothing couldbe easier.

Graphical TurboSECProcessing

Processing parameters that offer awide range of options areincorporated into each TurboSECmethod. The TurboSEC processingparameters work in tandem withthe TotalChrom method to providea total SEC data processingsolution. For example, raw datamay be smoothed prior to SECprocessing by using the Savitsky-Golay smoothing algorithms nativeto the TotalChrom software. Thismay be accomplished inTotalChrom’s Graphical MethodEditor (GME) which is alreadyfamiliar to TotalChrom users.

Also from within TotalChrom,negative peaks, often encounteredin refractive index detectors, canbe processed or ignored. To ensurethe most precise and reproduciblemolecular weight distributionresults, TurboSEC methods permitthe setting of processing limits andbaseline placement in the SECchromatogram graphically (Figure3). For detailed examination ofSEC data and baseline assignments,the user may zoom in on a portionof the chromatogram for a closerlook at the data.

WYSIWYG Reports andPlots

A wide variety of user-selectablereports and plots are available froma TurboSEC method. All SECreports and plots may be displayedon screen, prior to printing, basedon the report/plot options selectedin the TurboSEC method.

Report formats include a molecularweight average report, a peakmolecular weight averages report, aweight slice report, and anabove/below molecular weightlimits report (Figure 4). Themolecular average report mayinclude Mw, Mn, Mz, Mw/Mn

(Polydispersity), Mv, (z+1) AverageMw and Intrinsic Viscosity values.Reports may be stored as an ASCIItext file and printed at a later time.

TurboSEC plot formats include theability to display the SECchromatogram with molecularweight average values, peakmolecular weight values or peakretention times. The SECchromatogram may also bedisplayed in a time slice format.A number of slice report curves,including cumulative Area %,cumulative Mn%, cumulativeMw% and cumulative Mz%,may be overlaid on theSEC chromatogram,if desired (Figure 5).

Additionally, the SEC calibration curve may beoverlaid on the chromatogram. The TurboSECplot may also be sized to fit any portion of aprinted page. This feature facilitates printing theplot and report on the same page. All plots maybe reviewed in Print Preview mode.
