IoLine: 2 triple junction glass electrodes with iodine/iodide reference • No contamination of sample with metal ions (Tris buffers) • 3-chamber reference system allows for stability in all media and at
changing temps; used in variety of applications • 1 micro version for small sample size
ScienceLine: 3 double junction electrodes with high electrolyte outflow • Unique applications requiring particular junction • Ground joint: highest outflow, best for solutions with a lot of solids,
useful for solutions with low ions • Platinum: high outflow to prevent clogging, but not as high as ground
joint; micro size available • Fast response; double junction reduces the chances that the
reference will touch the sample, avoiding possible contamination.

Measurement Basics: Nernst Equation
• ISEs measure ion activity (i.e. “effective concentration”) -Ion activity < actual ion concentration due to interaction of ions in solution, reduces ions available to react
• ISEs are designed to measure specific ions, but other ions can interfere
-Adding ionic strength adjustor acts to minimize/mask effect of interfering ions
• pH electrode is a Hydrogen ISE
• ISE output is in mV; follows Nernst Equation
• Slope of electrode: 2.303 ∗ 𝑅𝑅𝑅𝑅𝑛𝑛𝑛𝑛
-Divalent (e.g. Ca2+) ~ 29 mV/decade (typical 26-29)
-Must have decade (i.e. order of magnitude) difference in calibration standards; can “fill-in” between standards if many used

ISE Electrode Types
Half cell ISEs:
• Contain only the ion sensor (half cell); separate reference must be used
-Both have ion exchange with the sample
-mV output determined by difference in potential b/w the sensing half cell and the reference
Combination ISEs:
• Contain the sensing electrode and reference electrode in the same body
• Much easier to use; preferred by most customers

Measurement Methods
Direct measurement:
• Simple and most commonly used ISE method
• Measure mV output in samples and determine concentration based on calibration (i.e. just as is done with pH)
Incremental methods:
• Standard addition, standard subtraction, sample addition, sample subtraction
• Methods of spiking the sample
• Sample ion concentration calculated from change in potential
• YSI TruLab 1320 and MultiLab 4010-2/3 have all incremental methods preprogrammed

• Range: 0.02-17,000 mg/L • Slope: 54-59 mv/decade • pH range: Must be 11 or higher to make NH3 gaseous. ISA has blue color at this pH
• Interferences: few, volatile amines • Applications/Notes: Popular for wastewater, aquaculture, agriculture, industrial. pH shift caused by ammonia gas is correlated to concentration

• Technology: Solid State • Features: Refillable, non-replaceable sensor • Range: 0.6-6,400 mg/L • Slope: 26-29 mv/decade • pH range: 2-6 • Interferences: Hg2+, Ag+, Fe2+, Br-, Cl- • Applications/Notes: Industrial, environmental. Also known as Cupric ISE.
Copper (Cu2+)
• Technology: Solid State • Features: Refillable, non-replaceable sensor • Range: 0.2-260 mg/L • Slope: 54-59 mv/decade • pH range: 10-14 • Interferences: I-, Br-, Cl-, S2- • Applications/Notes: Industrial, environmental. Standard solutions not included in electrode package (safety issue)
Cyanide (CN-)
• Technology: Solid State Crystal • Features: Refillable, non-replaceable sensor • Range: 0.02 mg/L - saturation • Slope: 54-59 mv/decade • pH range: 5-7 at low range, 11 at higher range • Interferences: OH- • Applications/Notes: Very popular for drinking water, industrial. Lanthanum fluoride crystal is the sensor.

TruLine ISE Tips
• Takes ~1-2 mL to fill electrode
• One order of magnitude (i.e. decade) difference between two standards; can include standards between
• Add ISA to both samples and standards to ensure similar ionic strength
• Short term (1-3 day) storage: place in low standard and fill the reference
• Long term: store dry (i.e. drain reference) and avoid sunlight
• Fill hole should be open during calibration and measurement, closed during storage
