BACKGROUND

SingHealth Advanced Bioimaging aims to establish a bioimaging hub for investigators in SingHealth and Singapore. This is by providing a wide variety of imaging platforms and services in both light and electron microscopy.

KEY OBJECTIVES

Provide guidance and expertise in image acquisition and analysis to investigators

Provide imaging services to investigators in SingHealth and Singapore

Educate and train investigators in bioimaging techniques through conducting workshops, seminars, and hands-on training sessions

CORE STRENGTHS & SERVICES

SingHealth Advanced Bioimaging provides imaging services for both light and electron microscopy. We focus on super-resolution light microscopy, including:

The table below summarises the lateral (XY) and axial (Z) resolutions that can be obtained with each technique:

Imaging Technique Lateral Resolution (XY) Axial Resolution (Z)

SIM 120 nm 250 nm

STED-3X 50-70 nm 70-130 nm

3D-STORM 10-30 nm 50-70 nm

3D-PALM 10-30 nm 50nm

Structured Illumination Microscopy (SIM)

Stimulated Emission Depletion microscopy (STED)

3D-Stochastic Optical Reconstruction Microscopy (3D-STORM)

3D-Photoactivated Localisation Microscopy (3D-PALM)

For image processing, we provide two HP Z420 workstations installed with the following scientic image rendering and analysis software: NIS-Elements, ImageJ, Imaris, Huygens, LAS-AF Lite, MetaXpress, ZEN, and ZEN Lite.

For electron microscopy, we provide ultra-thin tissue sample preparation and imaging services for Transmission Electron Microscopy (TEM). SingHealth Advanced Bioimaging owns a Joel JEM 2100 TEM and state-of-the-art EM preparation equipment from Leica. The EM preparation room is supplied with necessary chemicals and reagents for ultra-thin tissue sectioning, staining, coating and treatment. Below is a list of our equipment:

Leica EM KMR3 glass knife maker

Leica EM ACE200 sputter coater

Leica EM CPD300 critical point dryer

We have a fully functioning cell culture room, equipped with a CO2 incubator, a phase contrast microscope and a laminar ow hood for live cell/tissue treatment.

The following list of platforms can be booked via our Google calendar:

Nikon N-STORM (BF, WF, TIRF, live cell, d-STORM & N-STORM)
