Protocol for Arabidopsis seed sterilization

1. Sterilization solution: To 90ml of 5% sodium hypochlorite

solution, 5ml 37% !l is added slo"ly and carefully. The solution

is prepared in a #lass $ottle and ept open in a closed cham$er

or desiccator.&. The seeds are ept in an 'ppendorf tu$e (fe" seeds) or plate

(more seeds). The tu$e*plate is ept in the closed cham$er or

desiccator $y the side of the sterilization solution $ottle

immediately.

+The seeds are ept "ell spread so that more or less the entire

surface area of the seeds comes in contact "ith the #as or fumes

produced from the sterilization solution.

3. The cham$er*desiccator is closed and sealed immediately andept for & hours. The seeds then can $e used for #ermination.

Note: The sterilization solution can also $e made "ith -0ml of 5%

sodium hypochlorite solution and 10ml 37% !l. n that case the time

for "hich the seeds can $e ept in the cham$er should not e/ceed 1.5
