The NRF SARChI Chair in “Development and Optimization of Wastewater Treatment Technology for Developing Economies” is based at the Institute for Water and Wastewater Technology. Positions in the following areas are available. Post-doctoral: Parasitology PhD with documented background in parasitology (soil transmitted helminths) and laboratory quality assurance and control. Experience in instruments like Flow Cytometer, HPLC, UV/VIS are advantages Strong verbal and written communication skills required. Post-doctoral: Environmental/Molecular Microbiology PhD with documented experience in Environmental and Molecular Microbiology . Areas and competence to consider includes Microbial Source Tracking, Risk Assessment and a documented publication record in microbial methods and/or Flow Cytometry. PhD/Masters candidates: Area 1: Microbial source tracking methods applied in South African rivers. Microbial source tracking (MST) may differentiate sources (e.g., human, cattle, and bird) of fecal contamination in water. Methods include antimicrobial resistance markers, Bacteroidales and Bacteriophages markers, host specificity (like Cryptosporidium and for viruses), mitochondrial DNA and chemical markers. The focus areas will be South African rivers impacted by humans, wildlife, and pasturing cattle. Area 2: On-site detection system for multi-antimicrobial resistant bacteria and/or investigations of emerging chemical pollutants and drugs. On-site testing allow an earlier detection of potential disease agents, and a more precise identification of the origin of microbial contaminations. The aim of this study will be to develop and validate onsite detection strategies (e.g. Loop- mediated isothermal amplification: LAMP) for detection of targeted antimicrobial resistant bacteria and validate the developed assay in real life water samples. Wastewater reflects the use of drugs and organic pollutants in society, which will be the focus of a chemical study. Area 3: Emerging trend in host-phage interaction in environment and their public health impact Phagehost interaction and infection dynamics based on host range, cross infectivity, strong lytic activity, and electron micrographs for DNA sequencing and proteomic analysis. Integrative approaches and standard tools including Flow Cytometer, FISH, qPCR, HPLC, UV visible spectrophotometer etc. would be used to enable study of phage infection of bacteria from environmental samples and to discriminate between phagehost interaction outcomes (e.g., lytic, chronic, lysogenic). Post-doctoral: Parasitology

Integrative approaches and standard tools including Flow Cytometer, FISH, qPCR, HPLC, UV visible spectrophotometer etc. would be used to enable study of phage infection of bacteria from environmental samples and to discriminate between phage–host interaction outcomes (e.g., lytic, chronic, lysogenic).
