(Microfiltration)

Introduction
Oros system
Oral somotic

History
History
Osmotic pressure, a colligative property, depends on the conc. of solute
(neutral molecular or ionic species) that contributes to the osmotic pressure.
Solutions of different conc. having the same solute and solvent system exhibit
an osmotic pressure proportional to their conc..
☞ A constant osmotic pressure and thereby a constant influx of water, can be
achieved by an osmotic delivery system that results in a constant release rate
of drug.
∴ Zero-order release, which is important for a controlled release delivery system
, is possible to achieve using the platforms.
History

Applying the principles of osmotic pressure to a new generation of controlled drug
delivery devices with many advantages over other existing controlled drug delivery system
Rationale for design of osmotic controlled Drug delivery system
Ideal situation in drug therapies
Maintenance of a uniform conc. of drug at the site of action , since the
pharmacological action is proportional to its conc. at the site of action
Many forms of controlled drug delivery systems have been designed
but , all these controlled drug delivery system suffer from one or more
disadvantages
☞ pH , gastric motility , food effect
Theeuwes & Higuchi : Oros drug delivery system (1974)
☞ New generation of controlled drug delivery devices
☞ Systems that provide a sustained delivery rate , preventing the sudden
increases in plasma conc. that may produce side effects, as well as
sharp decrease in plasma conc. that may lower the effectiveness of
the drug.

But, water-insoluble
H = WH / WP
[ WH : weight of water imbibed per weight of dry polymer WP ]
* Degree of hydration : ratio of initial polymer weight to water uptake
Key parameters that infuluence the design of osmotic controlled drug delivery system
(Smax)
☞ Smin < Do < Smax
Orifice size
Cross-sectional area of orifice S0 should be maintained between the minimum
and maximum values
Key parameters that infuluence the design of osmotic controlled drug delivery system
Smin = 5 [(L/Pmax) μ (dV/dt)] ½ dV / dt : volume flux thrtough orifice
L : length of orifice [ usually the same as the thickness of membrane
μ : viscosity of drug solution flowing through orifice
Pmax : maximum tolerated pressure difference across membrane
before the occurrence of deformation of the housing
Estimation of minimum cross-sectional area
Orifice size
Key parameters that infuluence the design of osmotic controlled drug delivery system
Smax = Mtz f L / DsCs M tz : amount of drug delivered in zero-order release
Ds : drug diffusion coefficient in the permeating solvent
f : zero-order pumping rate
[ In practice , a fraction smaller than 0.025 generally is necessary to
minimize diffusion contributions ]
Estimation of maximum cross-sectional area
Orifice size
Key parameters that infuluence the design of osmotic controlled drug delivery system
Release rate depends on the solubility of the solute inside drug delivery

Evolution of Oros [ II ]
Higuchi-Theeuwes Pump (1970)
Rose-Nelson
Rose –Nelson

Membrane & Orifice Uniformity ?
Laser Drilling System Operation
To begin the process, tablets are first introduced on to a single line conveyor from a bowl feeder.
A color sensor views each tablet to determine which side is facing up.
In the specific case of osmotic pumps, tablets typically are colored brown on the push layer side with a pink or yellow drug layer side.
The hole thus needs to be drilled only in the yellow (or pink) side.
Next, a presence sensor detects the passage of a tablet and then triggers the laser drilling process if the results from the color sensor were that the tablet was facing right side up
Tablets then pass through a machine vision inspection system
A digital image of each passing tablet is acquired and compared against the four possible outcomes, listed in the table
Two of these outcomes constitute a “pass” and two are considered a “reject.”
Laser Drilling System Operation
Laser Drilling System Operation
Laser Drilling System Operation
Rejected tablets are removed from the conveyor by an air activated blow off system
Because of the speed at which the conveyor moves and the physical response time of the blow off system, the reject mode is activated as soon as a failed tablet is sensed by the vision system.
This typically causes one or two tablets ahead of the rejected unit to be expelled as well
Then, the reject state is usually left on until the system sees five tablets in a row that meet either of the two pass criteria.
An additional presence sensor downstream from the blow off verifies that no tablets are passing through the system when the reject condition is set to “on.”
Despite the fact that some good tablets are rejected by this necessarily rigorous approach, the system still typically operates at 98% efficiency (tablets in/tablets out).
After transiting the first laser drilling station, tablets pass single file through an inverter, and then continue on the conveyor through a second laser drilling station.
This second laser drilling station operates in exactly the same way as the first.
Its function is to drill any tablets that were wrong side up when they passed through the first drilling station.
Alternately, to drill both sides of a tablet, the color sensors in both stations are set to trigger the drilling process regardless of tablet orientation.
Also, in that case, the vision inspection system is programmed to reject tablets only when no hole is detected.
At the end of the line, processed tablets are fed into a collection drum, ready for final coating and printing.

Enhanced Bioavailablity By targeting specific areas of the gastrointestinal tract, OROS® technology may provide more efficient drug absorption and enhanced bioavailability L-OROS® technology can also enhance the bioavailability of drugs with low solubility
Patterned Delivery For many drugs, zero-order is not the optimal delivery profile
OROS® can be tailored to meet patterned delivery profiles to optimize a drug's therapeutic efficacy.
Reduced VariabilityThe osmotic driving force of OROS® and protection of the drug until the time of release eliminate the variability of drug absorption and metabolism often caused by gastric pH and motility

How OROS® technology produced Pfizer's first $1 billion-a-year product, and how OROS® technology produced the first methylphenidate treatment to offer 12-hour ADHD symptom control:
A pharmaceutical company's dream scenario : instead of suffering a monumental loss when an important drug goes off patent, the company launches a new, vastly improved version that eclipses both the original drug and its generic competition.
That's exactly what Pfizer did when it partnered with ALZA to develop Procardia XL®, which incorporates ALZA's OROS® osmotic technology.
ALZA's technology gave Procardia XL® several major advantages over Pfizer's original product, Procardia®, and generic nifedipine
As a result, the label indication for Procardia XL® was expanded to include treatment of both angina and hypertension.

OROS : 성공 사례
The use of OROS® technology can provide the desired plasma profile of the active agent and allows OROS® products-such as Concerta® for the once-daily treatment of Attention Deficit Hyperactivity Disorder (ADHD), and Ditropan XL®, the first once-a-day treatment in the U.S. for overactive bladder-to provide either patterned-release of medication or consistent levels of medication throughout the day
Until Concerta®, there was no once-daily methylphenidate medication for ADHD that worked effectively throughout the day
Concerta® uses an advanced OROS® patterned-release delivery system to achieve the desired improvement in symptoms, eliminating the need for in-school and after-school dosing.

시판제품 : COVERA-HS
The COVERA-HS formulation has been designed to initiate the release of verapamil 4–5 hours after ingestion
This delay is introduced by a layer between the active drug core and outer semipermeable membrane
As water from the gastrointestinal tract enters the tablet, this delay coating is solubilized and released
As tablet hydration continues, the osmotic layer expands and pushes against 2 the drug layer, releasing drug through precision laser-drilled orifices in the outer membrane at a constant rate This controlled rate of drug delivery in the gastrointestinal lumen is independent of posture, pH, gastrointestinal motility, and fed or fasting conditions
The biologically inert components of the delivery system remain intact during GI transit and are eliminated in the feces as an insoluble shell

of Risperidone
Invega
INVEGA® uses osmotic pressure to deliver paliperidone at a controlled rate
In the gastrointestinal tract, the water-dispersible color overcoat erodes quickly
Water is then absorbed through the semipermeable membrane at a controlled rate, which then controls the rate of drug delivery
The hydrophilic polymers of the core hydrate and swell, creating a gel containing paliperidone that is then pushed out through the tablet orifices
