PRODUCT OVERVIEW This product must be installed in accordance with the applicable installation code by a person familiar with the construction and operation of the product and hazards involved.

ScopeLED® B-Series Illuminator Overview ScopeLED

® B-series offers high brightness, tunable color and adjustable intensity in a compact,

flexible, and convenient package. The advanced thermal design provides a vibration free fan-less cooling with a high intensity optically stable light output. Different colored LED chips packed into DiCon’s Dense Matrix LED™ Array provides high efficiency and ultimate performance to replace conventional halogen light sources used for bright-field, dark-field, phase contrast, polarized light and DIC in transmitted and reflected light applications, illuminating your sample with selectable colors to enhance contrast, protect wavelength sensitive samples, and optimize viewing comfort. The LEDs do not emit in the UV and IR spectral regions, eliminating the need for special filters. Undesirable light and sample interactions such as photobleaching in fluorescence and unintended curing in photosensitive adhesives can be avoided by selecting a color channel that doesn’t interact with the sample. ScopeLED

® B-series also provide versatile operations that can be readily adjustable to most

systems. It can be simply controlled manually or can be automated and synchronized with other hardware via USB or External triggering, making it a great fit for automated production lines and robotics. The RGB model allows any quantitative black and white camera to function as a high-speed color imaging system with separate channel External trigger control, eliminating the need for specific software drivers. Its non-phosphor based DiCon’s Dense Matrix LED™ array is inert to laser excitation allowing it to be used as a self shuttering LED transmitted light source with any confocal or laser based fluorescence microscopy system. The ScopeLED

® B-series combine optical power, flexibility, and usability in a compact form and is

a great choice for your application.

Serial Commands

Message Structure The message formats is the same in both the send and receive directions. Each message starts with a sentinel Start byte followed by a message length byte, and ends with a checksum byte and sentinel Stop byte. The message length value excludes the checksum byte and the start and stop bytes. The checksum value is the negative two’s complement of the sum of all the data in the message including the length byte, but excluding the Start and Stop bytes. The value of the Start byte is 0xA9, and the value of the Stop byte is 0x5C. The drawing below illustrates the message format for the B-Series illuminator.

Byte Number

Description

1 Start Byte. This sentinel value signals the beginning of a message. The value is 0xA9.

2 Message length. This value is the count of all of the bytes, excluding the Start byte, checksum, and Stop byte. The length includes the length byte itself. In the illustration above, the length value is (N-3).

3 Command byte. This value is the command code for the message.

4 to N-2

Messages to the device Messages from the device

Parameter data. This is optional data for possible command argument parameters.

In messages from the device to the host, byte B:4 will contain the command result code. Refer to Section 6 for the meaning of the result code. The remaining data, bytes B:5 to B:N-2, is optional reply data. For example, the answer data to a query command would be included here.

N-1 Checksum. The checksum is the negative sum of all the data in the message including the length byte, but excluding the Start and Stop bytes. In the illustration above, it would be the negative sum of bytes B:2 to B:N-2.

N Stop Byte. This sentinel value signals the end of a message. The value is 0x5C.

EXTERNAL TTL/ANALOG

External Input Connector Overview The external TTL and Analog inputs have the highest priority of all modes of operation and will override any settings on the the manual controller and PC. This allows you to easily interface with hardware such as National Instruments DAQs and Arduinos. The external input connector operates though a 15-pin D-Sub connector depicted in figure below. Pin references are listed in Table1. Pin 1 to Pin 4 is for Transistor-Transistor Logic (TTL) signal input. Pin 7 to Pin 10 is for Analog control. Pin 5/6/11/12/16 is ground. Both TTL and Analog controls are with individual channel or Master (preprogrammed color mode).

Table 1: External Input Pin Configuration for Mode Control

Pin Function Input Signal Type

1 TTL1 (CH1) TTL (0-6V)

2 TTL2 (CH2) TTL (0-6V)

3 TTL3 (CH3) TTL (0-6V)

4 TTL Master TTL (0-6V)

7 Analog 1 (CH1) Analog (0-6V)

8 Analog 2 (CH2) Analog (0-6V)

9 Analog 3 (CH3) Analog (0-6V)

10 Analog Master Analog (0-6V)

5/6/11/12 Ground

NOTES:

Ground connection pulled to each Analog and TTL BNC cable.

The response time from the high voltage trigger to the output response of the Microscope Illuminator can be less than 2 us. (TTL shutter switch time). The Analog sampling rate is 5 Hz. (us = micro second, Hz = Hertz)

The analog control is from 0 to 6 volts, which corresponds to 0% to 100% light intensity. Please do not drive with more than 6 volts, or you may damage your unit.

2. Connect the DE-15P to 2-BNC Male Cable (for single color type) or DE-15P to 8-BNC Male Cable (for RGB color type) to D-Sub connector on the LED illuminator (if TTL or Analog control is necessary.)

3. Set Manual Controller to “On” position to turn the illuminator on.

NOTES: The illuminator is now ON with a shutter closed. No light output will be observed now unless you manually open the shutter via the shutter button or trigger via TTL. When triggering using TTL, Shutter button must be in CLOSE state (not illuminated green)

4. Use the Color Tuning (color/mode) knob to select the desired preprogrammed colors, white light CCT values. These knob settings are also referred to in this manual as preprogrammed modes. (See Preprogrammed Modes for mode descriptions. Rotating the Color Tuning knob clockwise will progress through these modes in the order listed.)

5. The Intensity knob is used to manually adjust the intensity or brightness of the modes without changing its color parameters. Turning the knob fully clockwise results in maximum brightness of the selected mode. Turning the knob fully counter-clockwise results in dimming the selected mode to zero output.

6. Connect the Male BNC connectors to a BNC breakout box or TTL signal generator for the Channel or Channels desired to be triggered

7. Ensure that the Shutter Button is CLOSE (not illuminated).

8. Send TTL signal to the desired channel for TTL triggering.

Master – Triggers all three channels for white light CCT channel selection TTL1 – Triggers the Red Color Channel only TTL2 – Triggers the Green Color Channel only TTL3 – Triggers the Blue Color Channel only

9. The intensity Dial will work during the TTL triggering and intensities can be changed

during trigger generation for illumination control.

Use of TTL and Analog Control for Shutter and Intensity

1. Follow the Uses Manual Controller to Set Channel Intensity and TTL to Shutter instruction to STEP 7.

2. Connect the desired Channels TTL and Analog BNC connectors to BNC break out box or to a TTL signal generator and a power supply output.

TROUBLE SHOOTING

Problem Solution

Illuminator does not turn on

Check that the green LED Power Indicator on the illuminator is on and verify that all of the connections to the adapter and the Microscope Illuminator are properly connected.

Turn the Intensity knob to maximum intensity and cycle through the different modes using the color-tuning knob. The color-tuning knob can be in a custom mode position where the custom mode has not been programmed. For your mode of operation, ensure that a higher priority input is not interfering. If you’re using the manual controller only, check that the “shutter” button is OPEN (illuminated green)

Illuminator is not emitting all of the colors (Not applicable to 5600K model)

Turn the intensity knob to maximum intensity, then cycle through each preset mode using the color-tuning knob. If any of the colors appear unusually dim, please contact the manufacturer.

Illuminator is flickering

Flickering is often a result of the driver not receiving a high enough DC voltage or current. Be sure to use the power adapter from ScopeLED to avoid power shortage issue.

Illuminator has shut down automatically and is hot to touch

Make sure there is nothing obstructing the airflow to the Microscope Illuminator. It is required that a minimum of eight cubic feet of airspace is provided around the device and the unit will auto shutdown if it gets too hot.
