Our mission and basic philosophy

After the experience of laboratory animal's rearing management for over 10 years, we have been handling the manufacture and sales of animal blood and sera. At our initial foundation period we have won over a lot of trusts through biotechnology service business, particularly establishing the position as the raw material supply base of the monoclonal and polyclonal antibodies.

Recently, we have been working on the development and manufacturing of microbiological media as well as tissue culture media even while industrial infrastructures of biotechnology was expanding, we especially concentrate on making development of the tissue culture media and challenging to a new creation at a vision.

YEARS

History

1981 Established as Kohjin Co., Ltd. at Suginami-ku, Tokyo, with the capital of JPY 10,000,000. Started on manufacture of animal blood/sera and culture media for bacterial test.

1982 Built a factory at Hanakage-cho, Sakado, Saitama and the manufacturing base moved there.

1986 Increased the capital to JPY 20,000,000. Built a new factory at Asabano, Sakado and started the manufacture of tissue culture media. Obtained permission as “Poisonous and deleterious substances manufacturing permit”

1989 Renamed to Kohjin Bio Co., Ltd. Increased the capital to JPY 28,000,000. Launched on production service of customer-oriented monoclonal and polyclonal antibodies.

1991 Increased the capital to JPY 48,000,000. Started manufacturing in vitro diagnostic agents. Obtained Medicine manufacturing permit.

1996 Moved the head office to Asabano. Increased the capital to JPY 70,000,000.

1997 Received the authorization on “Promotion of a creative action by small- and medium-sized businesses” from Saitama Prefecture.

1998 Received the commendation for “Technology development” from the Technology Development Foundation.

2000 Received the authorization on “Small- and medium-sized business innovation” from Saitama Prefecture.

2001 Built a new factory at the present address (Chiyoda, Sakado) and the manufacturing base moved there together with the head office.

2003 Acquired ISO 9001. Obtained the first to receive the authorization on “Management innovation model enterprise” from Saitama Prefecture.

2005 Increased the capital to JPY 98,000,000. Obtained cosmetics manufacturing permit.

2006 Acquired ISO 13485. Launched on development and manufacture of normal human dermal keratinocytes.

2007 Established Osaka office.

2008 Increased the capital to JPY 105,000,000.

2009 Obtained medical equipment manufacturing permit, the third-class medical equipment manufacturing sales permit, and the third-class medical equipment manufacturing sales permit. Establish Kohjin Japan Medical Biology Technological (Beijing) Co., Ltd as 100% subsidiary company in Beijing China.

MEM is also used to culture cells in a dihydrofolate reductase(DHFR) selection system.

DMEM Dulbecco's Modified Eagle's Media

R. Dulbecco and G. Freeman modified Basal Medium Eagle (BME) for culturing mouse embryonic cells in 1959. The modified BME, which is referred to as Dulbecco's Modified Eagle's Medium (DMEM), characteristically contains a 4-fold concentration of amino acids and vitamins. Furthermore, DMEM enriched with glucose from 1000 mg/L to 4500 mg/L is widely used for not only a conventional culture but also a large-scale high density culture of many cell lines.

Media 199 Earle

J. F. Morgan and his co-workers developed Medium 199 in 1950 to study nutritional requirements for culture of chick embryonic fibroblasts. Medium 199 characteristically contains certain vitamins, nucleic acid-related compounds, sugars, and antioxidants in addition to common amino acids and vitamins. The medium is frequently used as a basal cell culture medium for virus production.

McCoy's 5A Modified Medium without L-Glutamine

T. A. McCoy and his co-workers modified Basal Medium 5A in 1959 for culturing Novikoff rat hepatoma cells. The modified medium is referred to as McCoy's 5A Modified Medium. McCoy's 5A Modified Medium contains animal organ extract in addition to common amino acids and vitamins. The medium has been extended to culture other types of cells derived from a variety of organs such as bone marrow, kidney, lung, and spleen.

DMEM/F-121:1 Mixture of Dulbecco’s Modified Eagle’s Medium and Ham’s F-12

It has been thought that serum is an essential cell growth-promoting factor in culture. Since 1979 G. Sato and his co-workers have postulated a new concept that molecules replaceable serum in mammalian cell culture are considered as hormones where the term "hormone“ does not mean a classical endocrine hormone but is a growth factor. Representative hormones include serum albumin, transferrin, insulin, polypeptides growth factors, amine compounds, and trace elements. Several combinations of these hormones at appropriate concentrations support growth of many types of cells in serum-free medium. A 1:1 mixture of Dulbecco's Modified Eagle's Medium and Ham's F-12 (DMEM/F-12) has been widely used as a representative serum-free basal medium.

IMDM without L-GlutamineIscove's Modified Dulbecco's Medium without L-Glutamine

L. Guilbert and N. Iscove modified Dulbecco's Modified Eagle's Medium (DME) in 1976 for culturing B cells, T cells, and hybrid cells in the presence of appropriate stimulators. The modified medium contains potassium nitrate, selenium, sodium pyruvate, and HEPES in addition to common amino acid and vitamins formulated to DME. This medium is referred to as Iscove's Modified Dulbecco's Medium (IMDM). IMDM has been widely used as a defined basal serum-free medium particularly when high-density cell culture is required.

Glutamine synthetase (GS) selection is often used by culturing mammalian cells such as CHO cells to express recombinant proteins. 51Cr release assay has been so far widely used to evaluate cytotoxicity in immunology, pharmacology, toxicology and other medical and biological fields. However, the use of isotopes involves a variety of limitations. Radioisotopes must be treated in registered radioisotope facilities and used within their half-life. Furthermore, their quantity to be deposited or used at one time as well as the ways of safekeeping and waste disposal must be followed the governmental or local radioisotope regulations.

TeraScan is a microfluorocytometer developed for measuring cytotoxicity using a standard routine technique without limitations stated above. TeraScan consists of three pieces of apparatus, i.e. an inverted epifluorescent microscope, a control unit, and a data processor.

TeraScan assay requires fluorescent dye as a cytosolic marker of target cells, such as Calcein-AM. The routine procedure of the fluorometry is much simplified. Separation of the cells and incubation medium is not necessary after the target cells contact the effector cells, enabling kinetic analysis.

Not only NK cell- or CTL-mediated cytotoxicity but also antibody-dependent cellular cytotoxicity can be assessed by the TeraScan microfluorocytometer.

"KBM" Anaerobic Rabbit Blood Agar is used to culture the obligate anaerobe from a variety clinical specimens. This medium is excellent in the growthperformance of anaerobic cocci, such as Genus, Bacteroides that high detection rate, Clostridium, and Peptostreptococcus that phastidious organisms. Moreover, it is easy to make a differentiation and optimum for isolation and cultivation of phastidious anaerobic bacteria from the clinical specimen for Gram-negative rods (black pigment producing bacteria) such as Porphyromonas and Prevotella.
