presented the first ambulatory infusion system suitable for GLP canine

toxicity studies. Nine years later, I founded SAI with the goal of helping other

researchers implement reliable infusion techniques that are less stressful

Rodent Tethered Infusion Systems

Tethered systems require anin-dwelling catheter, a harnessor jacket, spring tether, swivel,swivel holder, extension linesand infusion pump. In the past,the fluid-carrying componentsof these systems (the catheter,extension lines and swivel) wereall connected by friction fittinga stainless steel cannula withthe internal diameter of flexiblepolymer tubing. New catheterand harness designs have madethe entire process more reliableand less time-consuming. Today,the most advanced systemavailable, the patented QuickConnect™ system, eliminatesall of the sizing issues inherent inolder systems and replaces thefriction connections with secureluer locks. These fittings can beconnected and disconnectedquickly and cleanly allowingfor intermittent infusion andsampling without continuoustethering or restraint. Highqualitypolyurethane has become the preferred catheter material due to its pliability and thrombo-resistant nature. Recent tapered catheter designs greatly increase success rates, allowing for smaller diameter distal endsthat insert easily into vessels andthicker walled proximal ends thatconnect directly to the harnessor swivel.

Tethered Rabbit Infusion Systems

Tethered infusion systems are well suited for studies that require large dosing volumes for extended periods of time or when repeated interaction with the pump is necessary. With large dosing volumes, syringe pumps may not be optimal since the largest standard syringe only holds 60ml. Volumetric pumps can handle a wide range of flow rates and can accommodate reservoirs of 2 liters or larger. Reservoirs and delivery ‘tubing sets’ are more expensive but the versatility and convenience are well worth it. The tethered system requires an in-dwelling catheter, either a tunneled exteriorized catheter or access port, jacket, metal tether, swivel, extension lines, and an infusion pump.

One system is an exteriorized catheter that luer-locks to the extension line. The alternative system utilizes a vascular access port accessed by a Huber needle or catheter and extension line.

Self-Administration Systems in Rodents

Self-administration, a form of operant conditioning, often provides the reward as an intravenous dose of drug. Both the test agent and the addictive drug can be administered remotely through chronically implanted intravenous catheters using swivel and tether systems.

Standard self-administration systems use threaded connections and friction fittings at the junctions in the fluid path. Self-administration systems which provide luer lock fittings (Quick Connect) reduce the potential for contamination and provide a secure, locking fit between the components of the fluid path, including harness or skin buttons, tethers, swivels and extension lines.

SAI’s split-septum systems provide rapid needle-less connection and disconnection and eliminate the torque on the harness or skin button when connecting and disconnecting either threaded or luer-lock systems.

Our self-administrationcomponents are designed for use with any standard operant self-administration system including but not limited to MEDAssociates or Panlab operantchambers.

Insulin & Glucose Clamp System

Components for Insulin and Glucose Clamp System in Rodents

SAI carries a complete set of components for researchers using the Glucose, or Insulin Clamp Technique (sometimes called euglycemic or hyperinsulinemic clamp) in rodents to assess Insulin Sensitivity. These clamp methods are typically employed in research designed to measure the effects of test agents or other types of interventions on metabolic disease. These systems require specialized infusion and sampling components to ensure meaningful data collection. Dual infusion lines for glucose and insulin, as well as a separate sampling line for blood measurements are often used concurrently in these complex systems. Accurate and easy-to-use syringe pumps are also essential in these studies, as the rate of infusion may be frequently adjusted throughout the Clamp procedure.

Fusion Touch Pumps Our line of Fusion laboratory syringe pumps are designed for accuracy and ease of use and monitoring. The Touch series features precise flow rates, combined with syringe recognition, and USB, RS232 and TTL for PC control. These pumps allow for Simple and Smart Advanced Flow Control. The new Fusion touch syringe pump has one of the newest ARM processors, allowing it to load/save methods, control motor force, and run an array of complex flow profiles. Users can program in (without a PC): Constant flow, Variable/Punctuated flow profiles, Gradient flows and Advanced loops.

Fusion Touch features an overview hybrid touchscreen and keypad interface. Mouse Tail Vein Catheters The SAI tail vein access system consists of a very small catheter and stylet that easily inserts into the tail vein of a restrained rodent. The size allows for quick, mistake-free access to the tail vein and once inserted, the flexibility keeps vessel trauma to a minimum. Unlike sharp, stiff needles this catheter moves freely up the vein to ensure the dose goes into the bloodstream, not into the surrounding tissue or back out through the insertion site. It bends naturally with the tail, reducing injury and allowing for continuous infusion or repeated bolus dosing with less chance of ‘losing’ the tail. These catheters are often used for MRI, PET and CT applications and can easily be modified to eliminate all metallic components.

Butterfly Needles These specialty needles (often called Winged Infusion Sets) offer a highly effective means of accessing a peripheral blood vessel for infusion or sampling in small and large animals. When compared to traditional straight needles, butterfly needles are often shorter, include wings on both sides of the needle for stability and have a tube attached to the end for easy connection to syringes or collection devices. The wings provide added grip and accuracy when handling the needle. These come in a wide variety of needle sizes and tubing lengths and can be customized. Often used to infuse in rodents via the tail vein, or in the cephalic or saphenous veins in larger animals they are biocompatible and resistant to organic vehicles. Femoral & Jugular Groshong® Catheters The FDA-approved “for human use” Groshong® line of central venous catheters by Bard offers the latest in technology. Unlike traditional open-ended catheters, the Groshong® catheter has a rounded, closed tip and features the patented Groshong® valve. The Groshong® valve opens inward for blood aspiration and outward for infusion but remains closed when not in use. Because the valve remains closed when not in use, the inside of the catheter is sealed off preventing it from coming in contact with the animal’s blood. The ultra low volume, top access port is designed to be accessed using a standard Huber or bullet point needle. Omni Harnesses and Tethers

The OMNI Harness

The word Omni is defined as “all” and that is exactly what the new Omni harness can do. By “all” we mean this harness can accept all types of needles or cannulas. Gone are the days where researchers are tied to a high-cost, specific or hard-to-get needle used to access their current harnesses. The Omni harness can accept a blunt needle, Huber needle, hypodermic needle, pencil point, luer stub adaptor, or a plastic cannula. The Omni harnesses use a special sealed septum to allow for all the needles to be used. This new septum will not leak, produce ‘cores’ or wear out over time with any of the needles used. SAI created the first Quick Connect harness and we have again re-invented the wheel.

The Omni harness comes in two configurations, single septum and dual septum. This will allow researchers to use single and dual cannulated rodents for sampling, infusion or both. We have a bile re-circulation harness as well. Each septum is color-coded so you know exactly what you are connecting.SAI has also gone one step further in the rodent harness design. We have incorporated a self-sterilization feature in our harness caps. We have developed a cap to which the user can add a small amount of 70% isopropyl alcohol in order to keep the septum sterile between samples or use. SAI is the first and only company to offer this technology.

Skin Buttons Rodent Skin Buttons are used for externalizing a catheter and/or for attaching a spring tether to the animal as an alternative to a harness or jacket. The polyethylene button is covered with a 1 in. Dacron patch for tissue ingrowth to provide a secure attachment. The polyethylene button center has an inner diameter of 0.123 in. which makes a perfect friction fit with a standard stainless steel spring tether with an OD of 0.124 in. All SAI tethers fit this skin button. Rodent Skin Buttons for use with auto samplers: With all the features of the standard skin button, the skin button for auto samplers has a wire hook for attachment to an auto sampler arm. The animal’s catheter can exit the animal through the center of the button and be attached to the sampler. The auto sampler’s arm connects to the wire hook to form a secure attachment.

HDS: Heparin/Dextrose Catheter Lock Solution Cath-Loc HDS acts as a non-toxic anticoagulant lock solution for in-dwelling catheters for pre-clinical research. This is one of the most common formulations used as a reliable pre-clinical lock solution to maintain catheter patency in vascular catheters. Used in both large and small animals, this solution combines the high viscosity of 50% dextrose with the anticoagulant activity of Heparin to promote and extend implanted catheter patency. The high osmolarity of dextrose prevents bacterial colonization and the high viscosity reduces washout from the catheter tip. Since both dextrose and Heparin are physiological constituents which occur naturally in the blood, any excess Cath-Loc HDS injected is quickly metabolized.

Surgical ServicesRegardless of species, continuous infusions lasting longer than two weeks require surgical procedures to insert and anchor in-dewelling catheters. Frequent intermittent infusions of long duration or with test formulations that require central venous or arterial delivery also require cut-down procedures for catheter or port implantation.

It is difficult, if not impossible, to maintain the skilled surgical staff necessary to perform these procedures without regular training or hands-on practice. SAI’s experienced surgeons understand this and offer surgical consulting and practical training to ensure your animals are successfully instrumented and maintained for the duration of the infusion.
