Ceva’s marek’s vaccines has been proven to give protection against all marek’s field viruses including the very virulent + marek’s disease virus.

to control marek’s disease, reducing early exposure in the field to marek’s disease virus is key, Ceva has developed a check list in order to reduce early exposure.

Marek’s disease and it’s economic impactmarek’s disease (md) is a common and widespread disease in poultry caused by a herpes virus. marek’s disease was first described in 1907 by Jozsef marek as Polyneuritis or Nerve inflammation1. Over the years the disease became more virulent leading to higher mortality, transient paralysis and to lymphoid tumors in the visceral organs.

economical impact of marek’S DiSeaSe marek’s vaccines have proven to be reducing mortality, which could reach 60% in a non protected layer flock. Non protected broiler flocks may experience losses from 0,1% to 0,5% and condemnations of 0,2% or more2.

Before the introduction of universal vaccination in commercial flocks, marek’s disease was widespread in chickens. vaccination reduces the mortality rate, but the disease remains one of significant importance. Recently the economic deficits caused by marek’s disease were estimated close to 1 billion US $3.

claSSification of marek DiSeaSe viruSeSthere are 3 serotypes of md historically described in literature; these have more recently been renamed4.

Currently the vaccines used to reduce the clinical signs of the disease are hvt (serotype 3), SB 1 (sero-type 2) and Rispens (serotype 1).

In short living birds (broilers) in several parts of the world hvt or hvt + SB1 vaccines are used, while in long living birds (long living broilers, layers and breeders), mainly hvt + Rispens, hvt + SB1 or Rispens vaccines are used.

controlling marek’S DiSeaSeIn order to fully control marek’s disease there are 3 essential points to consider.

1 vaccination of birds with a immune-potent vaccine.2 Correct storage, preparation and application of marek’s disease vaccine. great care should be

taken to ensure the storage conditions, preparation and application of the vaccine are as advised in order to obtain optimal protection.

3 Reduction of the risk of early infection on the farm: as it takes 8-21 days before the vaccine gives full immunity, it is important to reduce the risk of infection by the virus in the environment for up to 21 days.

Vectormune ND & rispens vectormune Nd & Rispens* is a vector hvt vaccine with the F protein of a Newcastle virus as an insert in the hvt, the Cevac md Rispens strain is added to the vaccine. the vaccine combines strong protection for marek’s disease (serotype 1 & 3) with the proven protection against Newcastle disease.

Strong, long laSting nD protection7

In order to demonstrate the efficacy against Newcastle disease virus, vectormune Nd** was mixed with a Rispens (CvI 988 strain) vaccine and applied to commerical brown day old layers. these birds were split into different groups.

group 1: vectormune Nd was given at one day old. No further Nd live or inactivated vaccines were applied.

group 2: vectormune was given at one day old. at one day old Cevac vitabron was applied by spray followed by a booster Nd killed (Cevac Nd IB edS k) at 15 weeks.

group 3: Control group non Nd vaccinated.at various ages the birds were challenged with two genotype vII Newcastle virus isolates. all groups were observed for mortality or other Newcastle clinical signs. egg production drops due to Nd challenge and viral shedding of the Nd virus were measured as well (data available on request).

.immuno-potent Marek’s vaccineIn order to get strong protection against very virulent and very virulent + marek’s viruses present in the field, it is necessary to vaccinate birds with a strong low passage Rispens + hvt vaccine. the strain used in the Ceva Rispens based marek’s vaccines is derived from the original CvI 988 strain isolated in chickens in 1972 in the Netherlands4.

Low passage strains are more immunogenic than high passage strains as they are less adapted to the chicken cells and the birds immune system will have a stronger response to the vaccine resulting in a stronger immunity.

SPF layers were split in 4 groups which were vaccinated at day old with Cevac md hvt & Rispens, a commercial hvt vaccine, or no vaccine. all groups, except the negative control group were challenged by s.q injection at 5 days old with the very virulent RB1B marek strain and observed for 45 days post challenge for marek clinical signs or lesions. after the trial all birds were culled and examined for lesions or other marek clinical signs.

Strong protection againSt very virulent + marek’S DiSeaSe6.In this trial, commercial white layers were vaccinated with Cevac md Rispens, a commercial hvt, or received no vaccine. all groups were mixed with non vaccinated shedder birds. these shedder birds have been challenged with a very virulent + marek’s disease virus (648a). the vaccinated and shedder birds were mixed at day old and the different groups birds were followed for marek clinical signs up to 18 weeks of age. this model provides an intense challenge, since the shedder birds are massively shedding high level of marek’s disease virus and continuously infecting the vaccinated birds.

Conclusion

Cevac md hvt & Rispens gives strong protection against a very virulent marek disease challenge strain*.

● hvt gives reduced protection against this marek disease challenge strain.*Strong protection against very virulent marek’s disease was also demonstrated when Cevac md hvt & Rispens was applied in-ovo.

Conclusion

By 3 weeks of age significant (~75%) Newcastle protection was seen which attained almost complete (95%) protection by 4 weeks of age regardless of the vaccination program.

100 % clinical Newcastle protection was achieved from 6 to 72 weeks of age.

vectormune Nd was also able to protect against production drops due to Nd challenge and reduced the shedding of the Nd field virus*.* data available on request

Conclusion

Cevac md Rispens provided efficacious protection against very virulent + marek’s disease, while the hvt vaccine provided only partial protection against this marek disease strain.

Prevention of early infectionStrict bio-security practices to limit the extent of early mdv exposure, are a crucial and cost-effective control method in combination with vaccination. Full immunity for marek’s disease in day old vaccinated chickens appears 8-21 days post vaccination; therefore prevention of early infection is important in order to let the vaccine build up its strong long lasting immunity. as marek’s disease can last up to 1 year in feathers and dust, proper cleaning and disinfection of the hatchery, materials, truck and farms is necessary.

actionS to prevent early infection

Prevent infection by other immune-suppressive diseases such as Cav and gumboro by vaccina-tion of the parentstock (PS) against these diseases.

hatchery level

Proper cleaning and disinfection of all materials (chick-boxes, equipment, etc before handling the day old hatched chickens.

Leave birds after vaccination for 4-6 hours before transporting, which will allow the replica-tion process of the vaccine to commence.

Reduce stress on birds during loading and transportation.

make sure the truck and other transporting materials are well cleaned and disinfected.

Farm level

Cleaning and disinfection of house where day old chicks will be placed.

Single age rearing is preferred.

In multi age rearing, place chicks in a separate house and prevent staff from visiting the house with the youngest birds after visiting older birds.

Check farm for recent marek’s disease outbreaks.

Reduce stress and immune-suppression due to disease, nutrient or climate issues.

Correct storage, preparation and application of the vaccineCell associated marek vaccines are stored in liquid nitrogen, which preserves the vaccine quality. For preparation of the vaccine, the ampules have to be thawed and injected in the diluent bags. during this process several factors can influence the vaccine quality and vaccine titer.

In order to improve the vaccination process, Ceva has set up the Ceva hatchery Immunization Control keys or C.h.I.C.k program.
