Form the 16th Century to, today, chemicalresearch teams have used glass containers fora very basic reason the glass containers istransparent, almost invisible. And so the contentsand the reaction are clearly visible, But becausechemists must heat, cool and mix chemicalsubstances, ordinary glass is not alwaysadequate for laboratory works.

Laboratory works requires apparatus madein a glass - which can readily be moulded intoany desired shape or from, which offers maximuminertness when in contact with the widest rangeof chemical substances, which can withstandthermal shock with fracture and hightemperature work without deforming, and whichwill be resillient enough to survive the everydayknocks to which it will be subjected in normallaboratory handling, washing and sterilizingprocesses.

Chemical CompositionLOBAlife - Glassco Glassware is a low alkali

borosilicate composition. Its typical chemicalcomposition is given under. It is virtually free ofmagnesia-lime-Zinc group and contains onlytraces of heavy metals.

In general the ‘Strain Point’ should beregarded as the maximum safe operatingtemperature of LOBAlife - Glassco glassware.When heated above 500°C the glass mayacquire permanent stresses on cooling. AllLOBAlife - Glassco labware is annealed inmodern ovens under str ict ly control ledconditions to ensure minimal residual stress inthe products.

Chemical DurabilityLOBAlife - Glassco Glassware in highly

resistance to water, neutral and acid solutions,concentrated acids and their mixtures as wellas to chloride, bromine, iodine, and organicmatters. Even during extended period ofreaction and at temperatures above 100°C, itschemical resistance exceeds that of mostmetals and other materials.

It can withstand repeated dry and wetsterilisation without surface deterioration andsubsequent contamination. Resistance to attackof various chemicals is shown under. Onlyhydroflioric acid, very hot phosphoric acid andalkaline solutions increasinglly attack the glasssurface with r is ing concentration andtemperature.

Fabrication with 3.3 Borosilicate GlassDue to low expansion of glass and easy

workability, this glass can be shaped, formed,joined into complicated apparatus. It can bedone even by an analyst in his own laboratory.He can keep on changing till he gets what heneeds. HEATING AND COOLING• Glass may suffer damage in three ways :• It may break under thermal stress in thesteady state, that is when there is establishedconstant thermal gradient through the glass.

• It may break under the transient stress of a‘thermal shock’, that is sudden heating or cooling• It may, i f heated beyond certaintemperature, acquire a permanent stress oncooling which could cause subsequent failure.

The following precautionary measures will assistin avoiding failures during heating and coolingprocedures.

1. Never leave vessel unattended whenevaporation work is being carried out. The vesselmay crack or explode as dryness condition isapproached if the heat source is not adjustedcorrectly. Lower the temperature gradually asthe liquid level drops.2. Always use caution when removingglassware from a heat source and avoid placingon a cold or damp surface. Although the warecan withstand extreme temperatures, suddentemperature changes may cause the vessel tobreak.3. Always cool vessels s lowly to preventthermal breakage.4. Never apply heat to badly scratched oretched vessel as the thermal strength will havebeen greatly reduced.5. Never apply point source heating to a vesselas this wil l greatly increase the chance ofbreakage.6. Always diffuse the heat source by using ametal gauze or air/water bath. Alternativelyensure even heating of the vessel byslowmovement of the vessel in relation to the heatsource.7. Adjust Bunsen burner to get a large softflame. It will heat slowly but also more uniformly.Uniform heat is critical factor for some chemicalreactions.8. Ensure that the flame contacts the vesselbelow the liquid level. Heating above that levelwill invite breakage of the vessels.9. Always use anti-bumping devices in thevessel, such as powered pumice or glass woolrapid heating of the vessel and contents isrequired.10. Never use material with sharp edges such asbroken porcelain as an anti-bumping device.This will cause internal abrasions and reduce themechanical and thermal strength of the vessel.11. Thick walled glassware should not besubjected to direct flame or other localised heatsource. Vessels of this type are best heated withthe use of an electric immersion heater.12. Avoid heating glassware over electr icheaters with open elements. Uneven heat of thistype can include localised stress and increasethe chance of breakage.13. Remember that the hot plate will retain heatlong after the appliance has been switched off.

14. Always ensure that the surface of the hotplate is larger in area than the base of the vesselbeing heated. An under-sized plate of the jobin hand will invite uneven heating and promotebreakage of glassware.15. Always ensure that manufacturer’sinstructions are Bollowed when electrical heatsources.

Mixing and Stirring1. Always use a policemen’s or similar deviceon stirring rods to prevent scratching the insideof a vessel.2. When using a glass vessel with a magneticstirrer always use a covered follower to preventabrading the inside of the vessel.3. When using glass or metal mechanical stirrerin a glass vessel always predetermine the heightof the stirrer before use to ensure there is nocontact between the stirrer blade and thebottom or sides of the vessel.4. Never mix sulphuric acid and water inside aglass measuring cylinder. The heat of reactioncan break the base of the cylinder.

Vacuum and Pressure1. Never use a glassware beyond therecommended safe limit.2. Always use a safety screen when workingwith glassware subjected to pressure of vacuum,3. Never subject glassware to sudden pressurechanges. Always apply and release positive andnegative pressures gradually.

Joining and Separating glass apparatus1. When storing glass stopcocks and joints,insert a thin strip of paper between joint surfacesto prevent sticking.2. Never store stopcocks for long periods withlubricant still on the ground surfaces.3. Glass stopcocks on Burettes and SeparationFunnels should be lubricated frequently toprevent sticking.4. If a ground joint sticks, separation cangenerally be achieved by carefully recking thecone in the socket, or gently tapping of thesocket flange on a wooden surface, or byheating the socket and not the cone in alocalised flame. The use of penetrating oil willoften prove useful in aiding separation.5. In using lubricants it is advisable to applylight coat of grease completely around theupper part of the joint. Use only a small amountand avoid greasing that part of the joint whichcontacts the inner part of apparatus.6. Three type of lubricants are commonly useon standard taper joints A) Hydrocarbon greaseis the most widely used. It can be easily removeby most laboratory solvents, including acetone.(B) Because hydrocarbon grease is so easilyremovable, silicon grease is often preferred for

higher temperature or high vacuumapplications. It can be removed readily withchloroform. (C) For long term reflux or extractionreactions, a water soluble, organic and insolublegrease, such as glycerin, is suitable. Water willclean glycerin.

There are other type of greases which canbe used specifically when certain reagents areused in the Burettes or Separating Funnels. 7. The use of water, oi l or glycerol isrecommended on both tubing and rubber bungwhen inserting glass tubing into a bung. Alwayswear heavy protective gloves or s imi larprotection when carrying out this operation.8. Always fire polish rough ends of glass tubingbefore attempting to insert into flexible tubing.The lubricants recommended above may alsoprove useful.9. Never attempt to pull a thermometer out ofa rubber bung. Always cut the bung away.

PERSONAL SAFETY1. Use tongs to asbestos gloves to remove allglassware from heat. Hot glass can cause seversburns.2. Protective gloves, safety shoes, aprons, andgoggles should be worn as safety chemicalaccidents, spilling or splattering.3. Always flush the outside of acid bottle withwater before opening. Do not put the stopperon the counter top where someone else maycome in contact with acid residue.4. Special care is needed when dealing withmercury. Even a small amount of mercury in thebottom of a drawer can poison the roomatmosphere. Mercury toxicity is cumulative andthe element’s ability to amalgamate with anumber of metals is wel l known. After anaccident involving mercury, the area should becleaned carefully until there are no globulesremaining.All mercury containers should be kept well-stoppered.5. Never drink from a beaker. A beaker leftspecifically for drinking is a menace to thelaboratory. Do not taste chemicals foridentif ication. Smell chemicals only whennecessary and by waiting a small amount ofvapour towards the nose.6. Avoid pipeting by mouth, particularly whenusing concentrated acids, alkalis or potentiallybiohazardous materials. Use m e c h a n i c a lmeans such as a rubber bulb or an automaticdispenser.7. Never fill receptacle with material otherthan that called for by the label. Label allcontainers before filling. Throw away contentsof unlabelled containers.8. To avoid breakage when clampingglassware, do not permit glass-to-metal contactand do not use excessive force to tighten the

clamps.9. Do not look down into a test tube beingheated or containing chemicals and do notpoint its open end at another person. A reactionmight cause the contents to be ejected,resulting in injury.10. Spattering from acids, caustic materials andstrong oxidizing solutions on the skin or clothingshould be washed off immediately with largequantities of water.11. When working with chlorine, hydrogensulphide, carbon monoxide, hydrogen cyanideand other very toxic substances, always use aprotective mask or perform these experimentsunder a fume hood on a well ventilated area.12. In working with volatile materials, rememberthat heat causes expansion and confinementof expansion results in explosion. Remember alsothat danger exists even though external heat isnot applied.13. Perchloric acid is especially dangerousbecause it explodes on contact with organicmaterials. Do not use perchloric acid aroundwooden benches or tables. Keep perchloricacid, wear protective clothing.14. When using hot plates and other electricalequipments, ensure the wire and plugs are ingood condit ion. Never handle electr icalconnection with damp hands.

CLEANINGSuccessful experimental results can only be

achieved by using a clean apparatus. In allinstances laboratory glassware must bephysically clean, in nearly all cases it must bechemically clean and in specific cases it mustbe bacteriologically clean or sterile. There mustbe no trace of grease and the safest criteria ofcleanliness is the uniform wetting of the glasssurface by distilled water-this being of the utmostimportance for glassware used for volumetricmethods. Any prevention of uniform wetting ofthe surface wil l introduce errors such asdistortion of the meniscus and accuracy ofvolume.

GENERAL CLEANING1. Cleaning of glassware which has containedhazardous materials must be solely undertakenby experienced personal.2. Most new glassware is slightly alkaline inreaction. For precision chemical tests, newglassware should be soaked several hours inacid water (1% solution hydrochloric acid ornitric acid) before washing.3. Glassware which is contaminated withblood clots, culture media, etc. must besterilized before cleaning.4. If glassware become induly clouded or dirtyor contains coagulated organic matter, it must

be cleaned with chromic acid cleaning solution.The dichromates should be handle with extremecare because it is a powerful corrosive.5. Wash glassware as quickly as possible afteruse but if delays are unavoidable, the articlesshould be allowed to soak in water.6. Grease is removed by weak sodiumcarbonate solution or acetone or fat solvents.Never use strong alkalis.7. Hot water with recommended detergentsshould be used and if glass is exceptionally dirtya cleaning power with a mild abrasive actioncan be applied, provided the surface is notscratched.8. During washing all parts of the article shouldbe throughly scrubbed with a brush selected forthe shape and size of the glassware. Brushesshould always be in good condition to avoid anyabrasion of glassware.9. When chromic acid solution is used, the itemmay be rinsed with the cleaning solution or itmay be filled and allowed to stand. The amountof t ime should depend on amount ofcontamination on the glassware.10. Special type of precipitate material mayrequired removal with nitric acid, aqua regia orfuming sulphuric acid. These are very corrosivesubstances and should be used only whenrequired.11. It is imperative that all soap detergents andother cleaning fluids be removed from glasswarebefore use. This is especially important with thedetergents, slight traces of which will interferewith serologic and culture reactions. Aftercleaning, thoroughly r inse with tap waterensuring that containers are partly filled withwater. shaken and emptied several times. Finallyrinse with deionised or distilled water.12. Drying can be undertaken either in basketsor on pages in air or at a temperature notexceeding 120°C.13. Always protect clean glassware from dustby use of temporary closures or by placing in adust free cabinet. For cleaning specific type ofglassware, please refer the following pages.

Cleaning Specific Types of Glassware Pipettes1. Place pipettes tips down, in a cylinder or talljar of water immediately after use. Do not dropthem into the jar, since this may break or chipthe tips and render the pipettes useless foraccurate measurements. A pad of cotton orglass wool at the bottom of the jar will help toprevent breaking of the tips. Be certain that thewater level is high enough to immerse thegreater portion or all or each pipette. At aconvenient time, the pipettes may then bedrained and placed and in a cylinder or jar ofdissolved detergent or, if exceptionally dirty, ina jar of chromic acid cleaning solution. Aftersoaking for several hours, or overnight, drain the

pipettes and run tap water over and throughthen until all traces of dirt are removed. Soakthe pipettes in distilled water for at least onehour. Remove from the distilled water, dry theoutside with a cloth, shake out the water anddry.

Burettes (with glass stopcock)1. Remove the stopcock key and wash theburette with detergent and water.2. Rinse with tap water unti l al l the dirt isremoved. Then rinse with distilled water and dry.3. Wash the stopcock key separately. Beforethe stopcock key is replaced in the burettsstopcock key are not interchangeable 4. Always cover burettes when not in use.

Culture Tubes1. Culture tubes which have been usedpreviously must be sterilized before cleaning.The best general method for sterilising culturetubes is by autoclaving for 30 minutes at 121°C(15ib. pressure). Media which solidify on coolingshould be poured out while the tubes areemptied, brush with detergent and water, rinsethroughly with tap water, rinse with distilledwater, place in a basket and dry.2. If tubes are to be filled with a medium whichis sterilized by autoclaving, do not plug until themedium is added .Both medium & tubes are thussterilized with one autoclave.3. If the tubes are to be filled with a sterilemedium or if they are to be sterilized by thefractional method then sterilize the tubes in theautoclave or dry air sterilizer before adding themedium.

Serological Tube1. Serological Tubes should be chemically cleanbut need not be sterile. However, specimens ofblood which are to be kept for some time atroom temperature should be collected in asteri le container. It may be expendient tosterilize all tubes as routine.2. To clean and sterilize tubes containing blood,discard the clots in a waste container and placethe tubes in a large basket. Put the basket. withothers, in a large bucket or boiler. Cover withwater, add a fair quantity of soap or detergentand boil for 30 minutes. Rinse the tubes and cleanwith brush, r inse and dry with the usualprecautions.3. It is imperative when washing serologicalglassware that all acid, alkali and detergent becompletely removed, Both acid and alkali insmall amounts destroy complement and in largeamounts produce hemolysis. Detergentsinterfere with serologic reactions.4. Serological tubes and glassware should bekept separate from all other glassware and usedfor nothing except serologic procedures.

BURETTES

When you use a burette you expect accuracy, reliability and durability .Only Loba life-Glassco brandburettes offer all three even after repeated usage. A combination of the best material andworkmanship goes into Loba life-Glassco brand burettes.The highest quality accurate bore tubing isselected to provide uniform and accurate graduations.Each Burette is individually calibrated for precisionand high accuracy.All graduations are in durable white enamel. The burette stopcocks are speciallyground so that they provide a leak proof operation.It is advantageous to use different types of greasesdepending on the reagents used to get leak proof operations.Stop-cock plugs are solid pressed andground.These are not interchangeable. In addition to Burettes with glass stop-cocks we have buretteswith straight bore PTFE STOPCOCKS and a range of Boroflo Burette fitted with Boroflo (GP) screw threadstop-cock with PTFE Keys.* Remove stopcock key and wash the burette with detergent and water,* Rinse with tap water until all the dirt is removed. Then rinse with distilled water and dry.* Wash the stopcock key separately. Before the stopcock key is replaced in the burette, lubricate thejoint with a small amount of lubricant, Remember that burette stopcock keys are not interchangeable.* Always cover burettes when not in use.

VOLUMETRIC WARE

Volumetric measurement is a routine task in the laboratory. The basic equipments used are Pipette,Volumetric Flasks and Measuring Cylinders.a) calibration of Measuring Equipment Proper use of calibrated volumetric glassware requires that theuser be informed whether the ware is a "to contain" or "to deliver" vessel. Volumetric ware is Calibratedto deliver (Ex or TD) or to contain (In or TC) the stated volume. (The International standards Organisationfor "to containis "In" and "todeliver,is"Ex") "Ex" Calibrated to deliver. The quantity of liquid deliverd corresponds to the volume indicated on theware. E.g..pipette. "In" Calibrated to contain. The quantity of liquid contained corresponds to the volume printed on theware. E.g..volumetric flasks, measuring cylinder.b) Accuracy Class "A" is the most accurate class. These items are marked with unique number and thecertificate is issued. Class "B" volumetric glassware is intended to use for general purpose in laboratory.i) In case of water the edges are drawn slightly upward, forming a concave meniscus should just touchthe graduation lineii) Incase of mercury a convex meniscus is formed. According to the International standards the volumeshould be read at the highest point of the liquid surface.i.e.the highest point of the meniscus should justtouch graduation line.iii) When a Schell Bach strip is provided, the contracts into a an arrow shape at the meniscus. Due to thelens effect of the water column the line appears broad inside the water column.Both the narrow and board line meet at the meniscus in arrow shape. The meeting point of both thearrows is the read off point of the graduation mark.

PIPETTES

LOBA Life Glassco-PipettesPipettes are used for accurate measurements and transfer of liquids. Volumetric pipettes will onlydeliver specific quantities. Graduated pipettes allow take - up of different liquid volumes and deliveryof the same or different quantities. Graduated and volumetric pipettes are of Loba life brand. They arecalibrated to deliver ( "EX") at the reference temperature of 20oC.In the case of calibrated pipettesand Class 'AS' pipettes, the tolerences on volume content corresponds to the accuracy limits of ISO -648 standards. The accuracy limits for Loba life Class 'B' type are about one and half times that of Class'AS'. This means that they are better than required by ISO 648 standard. Particularly in clinical laboratoriesis carried out on water or on dilute aqueous solutions. Thus, apparatus with the same accuracy limits,but with run-out time considerably shorter than hitherto demanded, are now admitted for calibration.
