Why Substitute Estrogen for Why Substitute Estrogen for GnRHGnRH??

• Estrogen induces:

– Sexual behavior--estrus

– Uterine tone

– Secretion of mucus

• Cows may be somewhat easier toinseminate at timed AI.

• Increased estrus activity has positivepsychological effects on thoseinseminating cows.

• Estrogen is significantly less costly.

How Do Estrogen and How Do Estrogen and GnRHGnRH Work? Work?

GnRH is secreted by thehypothalamus andinduces release of LHand FSH from the AP.

Extra Label Use of DrugsExtra Label Use of Drugs

• Extra label use means a drug is used forpurposes NOT listed as one of its Indications onthe bottle label or bottle insert.

• For example, the label for eachGnRH product indicates that itsapproved use is for the treatment ofovarian follicular cysts.

• Use of GnRH in any estrus-synchronization orovulation control program is considered to be anextra labelextra label use.

Extra Label Use of DrugsExtra Label Use of Drugs

• GnRH products have therapuetic approvals foruse in cattle in the U.S.

• Strict interpretation of Animal Medicinal Drug UseClarification Act (AMDUCA) is that GnRH productscannot be used for production purposes in cattle.

• However, GnRH products are being usedextensively for estrus-synchronization programsby veterinarians and academic researchers whohave published their results in scientific journalsand producer press.

Extra Label Use of DrugsExtra Label Use of Drugs

• GnRH is a peptide (very small protein with a shortblood half life) with no known health concerns.

• FDA must have minimal concerns regarding useof GnRH products in estrus-synchronizationprograms because no known prosecutions havebeen initiated.

Page 8: (ECP®) Using vs GnRH - BIF · PDF fileUsing Estradiol Cypionate (ECP®) vs. GnRH in Controlled A.I.-Breeding Programs Jeffrey S. Stevenson, Ph.D. Kansas State University

• Estradiol benzoate (EB) has no human or animalapproval in the U.S.

• Strict interpretation of AMDUCA is that EB cannot beused for production purposes in cattle.

• Therefore, use of EB in cattle for estrus-synchronization programs is illegal.

• Use of EB also is illegal when compounded with anyother approved product.

• Use of the Eazi-Breed™ CIDR® Cattle insert plusLutalyse® is an approvedapproved compounding of products.

What Estrogen is Approved? What Estrogen is Approved?

• Estradiol cypionate (ECP)has a therapeutic label foruse in cattle in the U.S.

• It is the only estrogenapproved for use in cattle isECP® (Pharmacia)

• ECP has multiple label indications including “tocorrect anestrus [absence of heat period] in theabsence of follicular cysts” at 3 to 5 mg doses.

Page 10: (ECP®) Using vs GnRH - BIF · PDF fileUsing Estradiol Cypionate (ECP®) vs. GnRH in Controlled A.I.-Breeding Programs Jeffrey S. Stevenson, Ph.D. Kansas State University

Use of ECP in Breeding ProgramsUse of ECP in Breeding Programs

• Strict interpretation ofAMDUCA is that ECP cannotbe used for productionpurposes in cattle.

• Because ECP is an estrogen,it is of concern to the U.S.Food and DrugAdministration-Center forVeterinary Medicine relativeto human health and safety.

Use of ECP in Breeding ProgramsUse of ECP in Breeding Programs

• ECP is being used extensively forestrus-synchronization programsby veterinarians and academicresearchers who have publishedtheir results in scientific journalsand producer press.

• FDA has not initiated prosecutionsof either researchers orveterinarians using ECP in cattleestrus-synchronization programs.

What Must Estrogen Do To ReplaceWhat Must Estrogen Do To ReplaceGnRHGnRH in Breeding Programs? in Breeding Programs?

• Estrogen must induceupfront follicle turnoverin a synchronizationprogram in cycling cows.

• Estrogen must induceupfront ovulation inanestrous cows.

• Estrogen must induceovulation after PGF.

• Estrogen must notproduce “hyper-estrus”activity to preventinjury of cows causedby excessive ridingand standing behavior.

• Estrogen must be easyto administer.

• Use of EB at 0.5 or 1.0 mg dose at the time ofCIDR insertion did not induce ovulation effectivelyin seasonally anestrous dairy cattle (Verkerk et al.,1998). Beef cattle?

• EB + CIDR reduced formation of persistentfollicles in lactating anestrous dairy cows, butdelayed follicular development in some anestrouscows (Rhodes et al., 2002). Beef cattle?

• Immature dominant follicles in suckled anestrouscows were less likely to ovulate after EB (Burke etal., 2001).

• ECP is dosed at 2 mg per mL.

• A small syringe is required todeliver 1 mg of ECP i.m. in avolume of 0.5 mL (0.5 cc).

• When injecting cows, followBeef Quality Assurance (BQA)guidelines to reduce carcassbruising and injection sitelesions (i.e., use neck injectionsites).

SummarySummary

• ECP is an alternative to GnRH for upfrontfollicle control, but may not be as effectiveas GnRH for anestrous cows.

• If ECP is used upfront at CIDR insertion, theCIDR must be in place for 9 days, ratherthan 7 days when using GnRH.

• After CIDR removal, ECP is an alternative toGnRH after luteolysis for TAI systems.

• Pregnancy rates to TAI tended to be greaterin suckled cows when treated after PGF withECP than GnRH.

Resynchronization of repeat estrus:• Had no negative effect on established

pregnancies.

• Increased synchrony of repeat estrus.

• Tended to reduce resynchronized conceptionrates after resynchronization in dairy and beefheifers .

• Produced normal conception rates at theresynchronized estrus in suckled beef cowswhen ECP + CIDR were used.
