Why choose Mast Isoplex®?

Mast Isoplex utilises the advantages of loop mediated isothermal amplification (LAMP*) to create effective and convenient diagnostic assays. LAMP is a well published and proven alternative gene amplification method, overcoming many of the practical limitations of working with conventional PCR.

RapidMast Isoplex® produces up to 1000 times more nucleic acid than PCR in an extremely short reaction time of 15-60 minutes.

RobustMast Isoplex® is less prone to substance inhibition than PCR. Accordingly, the assay is ideal for use in blood and faecal samples where inhibition1 can cause difficulties.

SimpleMast Isoplex® achieves excellent results without the need for extensive sample extraction and concentration (a requirement for PCR) with selected assays2.

ConvenienceDNA and RNA amplification kits afford a reduction in the time and complexity of assay development whilst dramatically simplifying the Quality Assurance process required for in-house assays.

Isothermal Amplification Simple temperature management at 60-65°C, allows continuous amplification of target gene sequences, removing the restriction of short periods of extension seen in PCR. Equipment requirements are minimal – a hot block and micro-centrifuge is all that is needed to run the assay. Simple Assay DesignPrimers can be designed using free web-based design tools or from commerciallyavailable software packages.

Single Tube Assay Reverse transcription, double looped template formulation, exponential amplification and detection of all reactions simultaneously in a single tube. As a result, the risk of contamination is significantly reduced.

Benefits of LAMP technology

LAMP technology can be applied to a variety of fields, by designing specific primers for targeting anyDNA or RNA sequence, rendering the assay especially effective for detecting target sequences fromviruses3, protozoa4, bacteria and fungi.

Highly Sensitive and SpecificSignificantly higher sensitivity when compared with real-time PCR, without compromising specificity.

Inhibitory substances have significantly less impact with LAMP, than PCR due to the reaction chemistry and isothermal amplification conditions.

Detection Methods

Two different detection dye options are included within all DNA and RNA amplification kits to aid differentiation:

Visual Detection Dye for visual end point reading • Hydroxynaphtholblue(HNB)isametal indicator, which enables the visual detection of the Mg2+ concentration and its decrease by pyrophosphate precipitation during positive LAMPamplification.TheadditionofHNBto the reaction mix, changes the colour from purple to blue in positive samples during DNA or RNA amplification.
