Front-end liquid handlers and autosamplers are available in a range of bed capacities and optimized bench space footprints . All models accommodate innovative injection and fraction collection capabilities within a single platform . Trilution® LC Software completely controls the entire HPLC system, allowing focus to remain on the science rather than the software .

The GX-271 Liquid Handler, is a mid-size capacity and throughput instrument fulfilling numerous laboratory needs at an economical price . (see page 43)

The 215 Series Liquid Handlers, known as the workhorses of the industry, are available in single, multiple and variable spanning probe models . (see page 48)

Gilson GX-281 Liquid Handler is the premier front-end instrument featuring a large bed capacity and a wide array of components such as automatic reinjection of fractions using innovative GX Z-Injection technology . (see page 46)

allows for injection, fraction collection and reinjection . The system is also ideal for liquid handling techniques such as sample mixing, dilutions and sample extract transfers .

The GX-281 is a multi-functional instrument that can perform injection, fraction collection and reinjection .

GX Direct Injection technology features an injection port that connects directly to the valve, minimizing volume associated with connective tubing (located behind the racks) .The GX Z-Injection Module

allows sample aspiration directly into the valve without a separate injection port, eliminating a source of potential carryover .

The prep-scale HPLC pumps feature low-dwell volumes which allow for fast, reproducible gradients . (see page 21)

The GX Prep Solvent System features a dynamic range of volumes and flow rates using syringeless technology within a solvent delivery pump capable of accessing five off-bed solvents .

The GX Z-Injection Module allows fast injections with low carryover and low maintenance by aspirating the sample directly into the valve .

The GX-281 features a Jet Wash Rinse station design that delivers individual streams of solvent for each separate rinse station to actively wash the outside of probes . Four rinse solvents are available for selection .

Separate injection and fraction collection paths reduce cross-contamination of fractions from sample injection . The cycle time of the injection is minimized by eliminating rinse steps which are required with single fluid path configurations .

Detection solutions are available for DAD, fixed or variable wavelength UV/VIS, UV, MS and ELS detection . (see page 25)

The Fraction Trapping System is ideally suited for processing fractions up to 20 mL in volume and concentrations up to 500 mg that have been purified on virtually any preparative HPLC system . Washing steps can be performed for removal of salts, buffers and mobile phase modifiers such as TFA that can hinder further LC/MS analysis . Fractions containing greater than even 50% water content can be processed and collected in 100% of organic solvent . Preservation and Quantification of Hormones in Surface Water

Using the Gilson GX-271 ASPEC™ System

The contamination of aquatic environments with hormones can disrupt the reproductive and developmental function of aquatic organisms such as fish (Jensen, K .M . et al ., 2006; Orlando, E .F . et al ., 2004) . One source of these hormones is the natural and synthetic hormones that are present in manure from large-scale livestock operations . Approximately 130 billion pounds of manure are produced annually in the United States (USEPA, 2000) . The hormones that are present in manure and in crop fields that have been amended with manure can be readily transported to surface and shallow ground water with events such as rain or snowmelt . There is a great deal of interest in studying the distribution and fate of these hormones in surface water . This application note describes the use of different preservatives (sodium azide, hydrochloric acid and sulfuric acid) to inhibit the degradation of hormones in samples of surface water runoff from cattle manure-amended fields during storage at 4° C . The Gilson GX-271 ASPEC™ System was used for this application .

An Automated Method for the Fractionation of Extractable Petroleum Hydrocarbons (EPH) from Water and Soil Extracts

Crude and refined petroleum products contain a complex mixture of aliphatic and aromatic hydrocarbons as well as a variety of other compounds . The concentration of these different hydrocarbons will vary in different products such as crude oil, refined products and other residual oil products . Many of these hydrocarbons have been shown to pose a risk to human health or to aquatic life . Leaking underground storage tanks are a common source of groundwater and soil contamination . It is

important to determine the types of hydrocarbons that may be present in contaminated soil and water . Government agencies and other regulatory bodies have developed several methods for determining the types of aliphatic and aromatic hydrocarbons that may be found in contaminated soil or water . The Gilson GX-274 ASPEC was used for this application .

Evaluation of Several Columns and Solvents for Post-Extraction Gel Permeation Chromatography (GPC) Clean-up of Fish Tissue Prior to PCB Analysis

The determination of PCBs, as well as other environmental contaminants, in fish tissue requires extensive sample cleanup prior to analysis by gas chromatography with an electron capture detector (GC-ECD) . The high fat content of fish tissue can cause buildup of nonvolatile materials on the GC injection port and the analytical column resulting in poor analytical results and high instrument maintenance costs . Gel Permeation Chromatography (GPC) is a size-exclusion clean-up procedure and it is a highly effective method for the removal of high molecular interferences such as lipids, proteins, pigments and cellular components from animal tissue . The Gilson GPC Clean-up System was used for this application .

An Automated Method for the Selective Solid Phase Extraction of Zearalenone from Wheat Using Molecularly Imprinted Polymers

Zearalenone [6-(10-hydroxy-6-oxo-trans-1-undecenyl)-ß-resorcyclic acid lactone] is a mycotoxin produced as a secondary metabolite of various Fusarium fungi . ZON is also known as RAL, F-2 toxin or ZAR . Zearalenone (ZON) has been detected in a variety of cereal products such as wheat, maize, barley, oats, rice and sorghum as well as beer that has been produced with contaminated grains . ZON can be excreted in cow’s milk after lactating cows are fed ZON in high doses (JECFA, 2000) . Exposure to ZON, in animals such as pigs and dairy cattle, has been known to cause estrogenic effects including infertility, reduced serum testosterone levels and sperm counts, reduced incidence of pregnancy and changes in progesterone levels . In addition, ZON can delay the breeding process and cost the producer significant economic and physical losses . Molecularly Imprinted Polymers (MIPs) have been demonstrated to be very effective tools for the selective extraction of an analyte from a complex matrix such as a food product (Haginaka, 2009; Wei et al ., 2007) . The Gilson GX-271 ASPEC™ System was used for this application .

Analysis of Sucralose in Water by Liquid Chromatography/ Time-of-Flight Mass Spectrometry (LC/TOF-MS) after Automated Solid Phase Extraction

Non-nutritive sweeteners are steadily increasing in importance with increased awareness of diabetes and its special dietary requirements and with more consumers becoming concerned about obesity and dental caries . Some of the most widely used non-nutritive sweeteners include saccharin, aspartame and sucralose . Sucralose is a water soluble tri-chlorinated disaccharide that is 600 times sweeter than sugar . Sucralose has a low human toxicity and is not metabolized by the human body . There has been a great deal of interest in monitoring levels of sucralose in water samples (Dye, 2007; Brorström-Lundén, 2007; Mead et al ., 2009) .Recently, some methods employing liquid chromatography-mass spectrometry (LC/MS) have been published for the analysis of sucralose (Loos et al ., 2009) and other artificial sweeteners in water samples (Scheurer et al ., 2009) . The Gilson GX-271 ASPEC System was used for this application .

An Automated Liquid Handling Strategy for Complex Matrices Based on Sample Viscosity

Performing manual liquid handling procedures in the laboratory is commonplace . When a laboratory faces an increase in sample volumes and throughput, it is often necessary to automate these liquid handling procedures . Automation of the liquid handling steps should improve day-to-day reproducibility of results . The viscosity of a liquid, defined as the resistance to a fluid to shear motion, can be especially challenging when developing an automated liquid handling protocol . Liquids with a higher or lower viscosity compared to water may require a different set of parameters to optimize the method . This note describes an automation strategy for the scientist who is transitioning from a manual liquid method to an automated method that is based on the viscosity of the liquid matrix . The Gilson GX-271 ASPEC System was used for this application .

Glycans consist of polymers of monosaccharides joined together by glycosidic linkages . Glycans are complex in structure and can be either branched or linear . Glycans are commonly attached to proteins to form glycoproteins . N-linked glycans (N-glycans) are the most common glycans bound to proteins in eukaryotic cells (Apweiler, R . et al ., 1999; Kronewitter, S .R ., 2010) . N-glycans play a major role in regulating biochemical pathways and in the communication at the cellular level between cells, tissue and organs (Shriver at al ., 2004) . As such, there is a great deal of interest in the study of glycosylation and N-glycans as biomarkers for a variety of diseases (Lebrilla, C .B ., 2009; deLeoz, M .L .A . et al ., 2008; Turnbull, J .E . et al ., 2007) . This application note describes a high throughput sample preparation procedure for global glycan massprofiling . The Gilson GX-271 ASPEC™ System was used for this application .

Identification and Quantification of Illegal Drugs and Benzodiazepines in Human Plasma by LC/MS-MS Analysis after Solid Phase Extraction (SPE)

The identification and quantification of drugs of abuse in blood (plasma, serum, whole blood) has become very common in the forensic medicine laboratory . A variety of methods for the extraction and analysis of these drugs have been described in the literature (Sadeg and Dumontet, 2001; Moeller and Kraemer, 2002), with particular focus on the use of GC/MS or LC/MS for analysis . Liquid/liquid extraction (LLE) has been traditionally used at a basic pH (pH = 9) for the extraction of amphetamines, cocaine and its metabolites and opiates from blood . This is sometimes followed by an additional clean-up step . This study describes an automated SPE protocol using the Gilson GX-271 ASPEC System .

Automated Solid Phase Extraction of Allantoin from Cosmetics and Topical Pharmaceuticals Prior to Analysis by HPLC

Allantoin is a heterocyclic organic compound derived from purine . It is also referred to as Glyoxylic Acid Diureide or 5-Ureidhyantoin . Allantoin is a metabolic end product of purine degradation in mammals (with the exception of humans and higher apes), as well as a metabolic intermediate in plants and some bacteria (Young et al ., 1944; Fujiwara et al ., 1995) . Allantoin can be found in many plant species . Comfrey (Symphytum officinale) has particularly high levels of allantoin . Allantoin has a long history of use in a variety of topical pharmaceuticals and cosmetics for skin care . It has keratolytic, moisturizing, soothing and anti-irritant properties . Allantoin promotes the renewal of epidermal cells and accelerates wound healing (AKEMA, 2008) . Allantoin is used in pharmaceuticals and dermatologic products in the treatment of ulcers, slow-healing wounds, burns, psoriasis and dry skin . There is a great deal of interest from pharmaceutical and cosmetic manufacturers in determining the amount of allantoin in a variety of products . This application note describes an automated SPE method using the Gilson GX-271 ASPEC™ System for the extraction of allantoin from a cosmetic product .

The vinegar fly or fruit fly, Drosophila melanogaster, has been utilized in medical and biological research laboratories for over 100 years . It was one of the first organisms used for genetic research and is the genetically best-known of all eukaryotic organisms . Research in the past twenty years has demonstrated that humans and fruit flies are very similar at the molecular level . About 75% of known human disease genes have a recognizable match in the genetic code of Drosophila melanogaster (Reiter, et al ., 2001) and 50% of fly protein sequences have mammalian analogs . Thus, D . melanogaster is an excellent animal model for studying a variety of human diseases . This application note describes the use of an automated instrument for filling large numbers of culture vials with fly food or fly culture media using the Gilson 215 Liquid Handler .

Using the Gilson GX-271 ASPEC System

Solid Phase Extraction has become a technique of choice for sample cleanup and trace Enrichment . Some of the benefits of SPE are: Versatility, Selectivity, Speed and Low Solvent usage . There are other methods for sample cleanup, such as Liquid-Liquid Extraction, Protein Precipitation, Turbulent Flow and Dialysis . Most automated methods find their roots in a manual method, but most manual methods don’t optimize the extraction mechanism due to the so called “time” believed to be involved in method development . Automation in its most general term removes the repetitive/monotonous nature associated with method development and the drain on human capital, while increasing recovery and optimizing precision . The Gilson GX-271 ASPEC System was used for this application .
