ThawSTAR® Systems Automated Cell Thawing
ThawSTAR® Cell Transport and Automated Thawing System is a first-of-its-kind technology platform that combines ultra-low temperature vial transport and automated cell thawing in a convenient and compact format. The system replaces manual methods of cell thawing - such as swirling frozen vials in communal water baths and warming vials between hands - that are not standardized and potentially jeopardize the integrity and safety of the sample, and enables thawing in a cell culture hood.
How It Works
ThawSTAR® Automated Cell Thawing Instrument features intuitive operation, making integration and standardization easy. Simply insert a frozen cryogenic vial and retrieve it when the vial is gently raised at the end of the thaw cycle. The small footprint and audio/visual feedback enable use in a cell culture hood with the rest of the workflow.
Reproducibility
Six vials were thawed in a 37°C water bath. The average thaw time in the water bath was 151 s, with a 99% confidence interval of 139-164 s (range shown as dotted lines). Left: The avg. time for the same ThawSTAR® instrument to thaw > 5 vials was measured each day for 5 days. Right: The avg. thaw time for 3 different ThawSTAR instruments was measured using > 5 vials per instrument on the same day. No significant differences were identified for either scenario at p<0.05 (2-way ANOVA with post hoc Sidak test).

Mean viability is compared post cryopreservation of paired PBMCs isolated immediately after phlebotomy for cells thawed in a water bath and using ThawSTAR® CFT2 Instrument (right). With fresh isolates, duplicate samples were processed by two different technicians to compare intertechnician variability for both PBMC isolation methods (results not shown) (n = 6). For cryopreserved samples, one vial from each of three donors was thawed for each condition being analyzed (n = 3).

Water Bath ThawSTAR®
Ex-Vivo B Cells Post-Thaw Viability and Consistency Compared to Water Bath
Ex-vivo stimulated B cells: A total of 6 vials, 2 each from 3 donors, were frozen in a -80°C freezer using a CoolCell® module and then stored in liquid nitrogen for two weeks. One vial from each donor was removed from LN2, placed into a ThawSTAR® CFT Transporter and thawed in a ThawSTAR® CFT2 automated cell thawing instrument. The other vial from each donor was removed from LN2 and hand-carried to a water bath. The vials transported and thawed in the ThawSTAR transport and thawing system performed better in terms of cell viability and recovery. - Data generated by UCSF Diabetes Center and Transplant Surgery

ThawSTAR® CFT2 Transporter is comprised of an outer insulative foam lid and base, and an internal round CoolRack® CFT2 sample module that accommodates up to five 1.8 or 2.0 mL cryogenic vials. Prior to using the ThawSTAR CFT2 Transporter, a thin layer of dry ice is added to the bottom of the base. The round CoolRack CFT2 module is placed over the dry ice and allowed to equilibrate to dry-ice temperature (approximately 20 minutes). Frozen vial is then extracted from storage and quickly placed into the module. The lid is placed securely on top of the base and all samples are maintained at dry ice temperature until transported to the thawing location.
How It Works
Thermal Profile
1.8 mL cryogenic vials filled with 1.0 mL of cryopreservation medium (10% DMSO/20% FBS/70% DMEM) were fitted with an interior wall thermocouple (< 0.5 mm from internal wall surface) and a central axis thermocouple, frozen at -80°C, and maintained at -75°C in a CoolRack® on dry ice. Three vials were thawed in a 37°C water bath (left panel) or in the ThawSTAR® CFT2 Instrument (right panel). For the water bath thaw, the vials were removed from the bath when a pea-sized ice chunk remained (arrow) and then gently tapped to melt the chunk. Similarly, ThawSTAR instrument ejected the vial at the point where a pea-sized ice chunk remained (arrow) and after mixing the final vial temperature is ~5-10°C. Note that the sharp rise in temperature seen with the central thermocouple is indicative of the ice chunk breaking away from the thermocouple. The temperature profiles recorded were practically identical for both the water bath and ThawSTAR thaw.

Performance Data
ThawSTAR® CFT2 Transporter holds cryogenic vials at near dry ice temperatures prior to thawing in the ThawSTAR® CFT2 Instrument. Dry ice is placed in the foam holder below the metal CoolRack® module. To verify the holding temperature, a cryogenic vial filled with cryopreservative solution with a centrally-located thermocouple was frozen in LN2 then transferred to the pre-equilibrated CFT Transporter. The temperature profile shows that the vial equilibrates quickly (< 20 min) to the holding temperature and remains stable for hours.

The ThawSTAR Instrument thaw time is 2-3X faster than other dry thawing methods tested here.

(1) Cryobox sitting in cryostorage container with 0.5 inches of LN2 prior to insertion into CryoPod™ Carrier. Samples in cryobox <-170°C. (2) Cryobox inserted into CryoPod Carrier. (3) CryoPod Carrier transported by foot around local vicinity for 1.5 hours. Ambient temperature of 18-21°C (65-70°F). (4) Cryobox removed from CryoPod Carrier and inserted into LN2 cryostorage container.
The entire 2.5 inch working depth of the CryoPod™ Carrier remains well below -150°C for over 4 hours with one charge of LN2 and with the lid closed. Temperature readings were taken on the CryoPod Carrier interior basket floor, 2 inches and 2.5 inches from the floor, and on the top of the basket for over 4 hours. The basket is situated above the compartment containing LN2 and there is no direct contact between samples (e.g. cryobox) and the LN2.

BioT™ ULT Transporter provides a stable ultra-low temperature (< -70°C to -50°C) work zone 8.0 inches (20.3 cm) deep. Up to 8 standard 2-inch cryostorage boxes will remain safely below -50°C for over 8 hours with the lid open and over 24 hours with the lid closed with one charge of dry ice.

BioT™ ULT Transporter is a dry-ice based temperature stability system is a convenient mid-sized solution for transporting temperature-sensitive material. It also serves as a compact, portable, ultra-low freezing station for freezing samples at a collection site. The patent-pending DIR™ cooling insert technology inside the transporter harnesses cold convection for efficient, stable, and hours-long dry ice-based cooling. The base chamber accommodates up to 8 standard 2-inch cryostorage boxes. The BioT ULT Transporter LN2 Kit is an insert for the BioT™ ULT Transporter that enables the unit to be used as a transport and sorting station for samples stored at or below vapor phase liquid nitrogen temperatures. When installed in the ULT Transporter and charged with liquid nitrogen, the insert creates a cryogenic temperature zone of < -150˚C. BioT™ ULT Transporter is comprised of three parts: magnetized lid, DIR™ dry ice retainer and base chamber. To begin, insert the DIR™ retainer, flanges down, into the base chamber. Add pelletized dry ice into the cavity between the DIR retainer and the base chamber walls, filling to the top of the DIR retainer. A full load of dry ice is approximately 13 lbs (5.8 kgs). The internal chamber will reach optimal working conditions (<-70˚C to -50˚C) within approximately 10 minutes and will be ready for use.
BioT™ ULT Transporter LN2 Kit is comprised of three parts: thermo-conductive basket, thermoconductive shelf plate and absorbent/baffle pad. Simply insert the thermoconductive basket in the empty BioT ULT Transporter chamber, followed by the absorbent/baffle pad. Place the thermoconductive shelf plate into the basket on top of the absorbent/baffle pad. With the addition of LN2, (~ 2.7L) the internal chamber will reach <-150˚C within approximately 10 minutes.

BioT™ ULT Workstation is comprised of four parts: magnetized lid, DIR™ insert, base chamber and mobile cart.
The patent-pending DIR cooling insert technology inside the workstation harnesses cold convection for efficient, stable, and hours-long dry ice-based cooling. Pelletized dry ice is added to the cavity between the 3-sided DIR insert and base chamber walls filling to the top of the retainer (approx. 90 lbs, 41 kg). The ventilated thermo-conductive insert ensures uniform and consistent gas circulation to keep the entire base chamber at ultra-low temperature. This configuration also minimizes consumption of dry ice as one charge maintains lower than -50°C temperature for over 15 hours with the lid open.
The closed-cell cross-linked polyethylene foam construction of the lid and base chamber is durable, non-absorbent, and remains comfortable to the touch even when loaded with dry ice and frozen materials. The DIR insert is constructed of a thermo-conductive aluminum alloy anodized for durability. BioT ULT Workstation may be wiped clean with 10% bleach or 70% alcohol solutions. DIR insert may be autoclaved or wiped clean with 10% bleach or 70% alcohol solutions.

CoolCell® alcohol-free cell freezing containers ensure standardized controlled-rate -1oC/minute cell freezing in a -80oC freezer - without alcohol or any fluids. Proven for use with a variety of cell types including stem cells, primary cells, PBMC cell lines, insect cells, yeast and others. The patent-pending CoolCell technology utilizes a thermo-conductive alloy core and highly-insulative outer material to control the rate of heat removal and provide reproducible cell cryopreservation. CoolCell® FTS30 utilizes a solid state core and controlled micro-convection technology to evenly draw in -80°C freezer air through a bottom base vent, uniformly disperse the cold air around each vial in the central chamber and then release the thermal load from the vials through a top vent choke. The inner vial module holds 30 cryogenic vials and can be removed in one step. Each vial achieves a uniform and reproducible -1°C/minute freezing profile and thermal profiles are highly reproducible. Due to the low thermal mass of the uniform-density cross-linked polyethylene foam container, freezing can be conducted without a rise in local freezer temperature, thereby protecting nearby samples.
CoolCell® LX, in combination with a -80°C freezer, will provide the freezing rate of -1°C/minute that is ideal for cryopreservation of most cells and cell lines. Using a combination of uniform-density cross-linked polyethylene foam, a solid state core, and radial vial symmetry, freezing profiles are consistent and reproducible. It is important to fully load CoolCell containers prior to freezing. Foam is non-absorbent and will impose negligible change in the freezer environment; thereby protecting nearby frozen samples. The low heat content also ensures that CoolCell LX containers will rapidly return to room temperature when removed from freezer.

Performance test: A temperature probe was placed into a 2.0 mL cryogenic vial containing 1.0 mL of water and the tube was inserted into a room temperature CoolCell®. The CoolCell was placed into a -80°C freezer and the temperature rate and profile was recorded over a 3 hour period. The test was repeated 5 consecutive times. Conclusion: CoolCell generated identical fusion time and cooling profiles over five consecutive freeze cycles.

Human embryonic stem cells, RC-10 were frozen using the technique indicated, thawed after 2 weeks in LN2, and counted immediately (Day 1) or after 3 days of growth (Day 3).
Ag-Tregs. Effects of freezing on antigen-specific Treg (Ag-Treg) cell therapy products; Ag-Tregs (n = 6) were frozen at concentration of 1 to 10 x 106 cells/mL using the CoolCell freezing device or controlled-rate freezer (CRF) with a freezing rate of -1°C/min. Viability and absolute viable cell count of thawed Ag-Treg cell therapy products were evaluated by flow cytometry.

Choose Your CoolBox™ System
An open-platform ice-free cooler that accommodates most CoolRack® and CoolSink® modules. Low profile and small footprint make it ideal for use in the hood, keeping samples cold (0.5° to 4.0°C) up to four hours. 1°C to 8°C temperature indicator provides visual assurance of temperature performance. To extend the cooling duration, keep an additional XT Cooling Core in the freezer and rotate the Cores as needed.

CoolRack® Thermo-Conductive Tube Modules
CoolRack® thermo-conductive tube modules eliminate variability which originates from tubes placed directly into ice, dry ice, alcohol baths, water baths and other temperature sources. Place the CoolRack module directly onto a temperature source between -196°C to >100°C and it will rapidly adapt to that temperature. CoolRack modules ensure +/- 0.1°C temperature uniformity across all tubes when cooling, snap freezing, heating or thawing. Suggest-ed applications include cooling reagents such as restriction enzymes, dNTPs and antibodies, alcohol-free dry ice snap freezing of tissue, virus and bac-teria samples and bench top cryogenic tube sorting in liquid nitrogen. All CoolRack modules may be autoclaved, high heat sterilized or decontaminated with bleach, alcohol or other disinfectants or lab detergents. Certain CoolRack modules are SBS-compatible.

CoolRack in Water Bath: Heat is transferred from water bath toward relatively cooler CoolRack until equilibrium is reached.
CoolRack® and CoolSink® are precision-engineered sample modules manufactured from a novel thermo-conductive alloy material. Thermo-conductivity is the transfer of heat energy from a higher temperature region to a lower temperature region. CoolRack modules evenly distribute the temperature across all wells providing very uniform and consistent temperature to all samples (+/0.1°C).

Performance test: A temperature probe was placed into a 2.0 mL cryogenic vial containing 1.0 mL of water. The tube was inserted into a CoolRack® CF45 thermo-conductive module. The module was placed onto a ThermalTray platform in a 37°C water bath and allowed to equilibrate. The CoolRack CF45 module was then removed and placed onto dry ice and equilibrated to -78°C (0 - 15 minutes) and then returned to the water bath to re-equilibrate to 37°C (15 - 30 minutes). This experiment was repeated five consecutive times and temperature profiles were recorded.
Conclusion: The CoolRack CF45 module showed identical cooling profiles and phase transition (orange circles) over five consecutive freeze-thaw cycles.

ThermalTray™ Thermo-Conductive Platforms
ThermalTray™ thermo-conductive platforms support CoolRack® and CoolSink® sample modules in liquid temperature sources such as melting ice, water baths and liquid nitrogen. Made of the same highly conductive alloy as CoolRack and CoolSink modules, ThermalTray platforms conduct the source temperature to the CoolRack or CoolSink and, ultimately, to your samples. The stable, sturdy design makes them ideal for processing temperature-sensitive samples in melted ice baths or liquid nitrogen. All modules may be autoclaved, high heat sterilized or decontaminated with bleach, alcohol or other disinfectants or lab detergents.
ThermalTray™ platforms are compatible with all temperature sources. If using in liquid nitrogen, the ThermalTray SLP or LP is highly recommended. These low profile ThermalTray platforms allow development of a vapor layer which will help insulate samples from ambient air.

TruCool® Barcoded Cryogenic Vials TruCool leak-proof, auto-cap cryogenic vials are ideal for cell culture and biobanking. The screw cap features a co-molded thermally-fused gasket which prevents leaking, slipping and risk of contamination. The gasket is 95kPa certified to provide a leak-proof seal. The star socket on cap top is compatible with auto-decapping equipment. Each vial is individually barcoded with a unique identifier that can be read with common barcode readers. Not recommended for storage directly in liquid nitrogen. Vapor phase ok. 500 per case.
