Levels of chemical contaminants in foods and feeds can originate from natural sources such as mycotoxins, which are toxic secondary metabolites produced by fungi on agricultural commodities in the field and during storage. More than 300 mycotoxins have been identified so far with widely different chemical structures and differing modes of action. The potential health risks to animals and humans posed by food- and feed-borne mycotoxin intoxication have been recognized by national and international organisations, which have addressed this

problem by adopting regulatory limits for major mycotoxin classes and selected individual mycotoxins. The Romer Labs Guide to Mycotoxins is a timely reflection of the research progress related to the significance, sampling and determination of mycotoxins. In this respect, the Romer Labs Guide will not only provide a thorough introduction into the history of mycotoxins and mycotoxicoses, but will also discuss the chemistry and toxic effects of the most common mycotoxins. The book will also provide guidance for the development of proper sampling and sample preparation techniques to obtain representative samples for the subsequent analysis of the food and feed commodities to be checked for mycotoxins. The analytical part of the Romer Labs Guide to Mycotoxins covers proper extraction and clean-up procedures, separation and detection techniques including latest state-of-the-art rapid strip tests. Special emphasis is put on multi-toxin methods based on liquid chromatography coupled with mass spectrometry for the simultaneous determination of a wide variety of secondary fungal metabolites which allow for a comprehensive assessment of the wide range of mycotoxins humans and animals are exposed to.

AgraQuant®

ELISA Tests

The Romer Labs® AgraQuant® product family is a range of enzyme-linked immunosorbent assay (ELISA) test kits that encompasses mycotoxin, allergen, GMO and drug residue tests. ELISA tests are quantitative tests. Reactions are read by a specific ELISA reader, such as the StatFax and the Chromate ELISA reader, for quantification purposes. A set of five to six standards used for calibration is supplied with all test kits to determine a result. Creating a new calibration curve with every assay setup assures high accuracy and reproducibility. The results obtained are comparable to HPLC results.AgraQuant® kits are direct competitive ELISAs for mycotoxin and drug residue tests and sandwich ELISAs for allergen and GMO tests. All AgraQuant® ELISAs use specific antibody coated microwells and an enzyme-labeled analyte conjugate. The quality of the antibody is crucial for test-result quality. Romer Labs® has a broad range of monoclonal antibodies, which are highly specific to the respective analyte and guarantee robust and accurate results.

AgraStrip® Lateral Flow Devices

The Romer Labs® AgraStrip® product family utilizes lateral flow technology, also known as immunochromatography tests. Most lateral flow devices (LFD) come in a format that is often called a dip stick or strip test. While the use of this technology is very widespread in the clinical field, the food safety market has only seen suitable applications in the last few years. AgraStrip® LFDs use specific antibodies labeled with colloidal gold for visual color development at a test and control zone. Developed LFDs can be read visually, making instrumentation redundant. They normally produce only a yes/no answer, indicating the presence or absence of an analyte at a defined concentration. However, if instruments such as the Romer Labs® AgraVision™ are used, LFDs can be read, providing a quantitative result. A digital camera measures the intensity of the reaction bands developing at the control and the test line in this method.

RapidChek® Pathogen Detection

As a food processor, choosing the best pathogen detection method can be a very difficult decision. The method should have a meaningful impact on the total cost of a food safety program but most importantly, provide maximum assurance of reliability in the test results. your customers and essentially your business.Romer Labs RapidChek® and RapidChek® SELECT™ product lines were developed to deliver the simplicity, speed and high accuracy necessary every day. Our technology sets us apart from other rapid test methods. The RapidChek® product family utilizes highly selective enrichment media coupled to advanced lateral flow immunochemical detection of food pathogens such as Salmonella spp., Listeria spp., and Escherichia coli O157 in various foodstuffs and environmental applications. Patented phage technology used in the SELECT™ system suppresses the growth of competitive microflora which creates optimal growth conditions for Salmonella spp. Highly specific monoclonal and polyclonal antibodies are exclusive to the RapidChek® system which works without the need for expensive equipment or extensive training. RapidChek®. Simply Accurate.

FluoroQuant® Fluorometric Test

The FluoroQuant® Test Kits are the fastest, quantitative fluorometric tests based on a solid-phase or immunoaffinity column cleanup. They are USDA/GIPSA approved and AOAC Performance Tested. These tests provide a wide quantitation range without dilution steps. The FluoroQuant® tests are simple enough for field use in grain, feed and food handling facilities, but they are also appropriate for analytical laboratories. Each kit comes complete with all test and accessory materials such as columns, filter paper, pipette tips, cuvettes and reagents.The FluoroQuant® Test kits require an FQ-Reader, which reads the fluorescence signal of the test and translates it into a quantitative analytical result. The FQ-Reader comes with a touch screen and can be connected to a PC and a printer.

MycoSep® and MultiSep® Cleanup Columns

In order to shorten the sample cleanup process, Romer Labs® has developed a product portfolio of one-step-cleanup columns for different groups of mycotoxins. MycoSep® and MultiSep® columns contain packing materials made of absorbent mixes designed for all relevant food and feed commodities. MycoSep® columns allow a one-step cleanup within 30 seconds: the MycoSep® cleanup column is pushed into a test tube containing the sample extract, forcing the extract to filter upwards through the packing material of the column. The interferences adhere to the chemical packing in the column and the purified extract, containing the analytes of interest, passes through the column. Similarly to the MycoSep® column format, the MultiSep® column format allows a one-step cleanup of sample extracts, containing the

analyte of interest, within 30 seconds: the sample extract is applied on top of the MultiSep® cleanup column, allowing the extract to go through the packing material of the column by gravity, or alternatively, by forcing the extract through the column material via the application of vacuum or positive pressure. The interferences adhere to the chemical packing in the column and the purified extract, containing the analytes of interest, can be collected in a suitable vessel.

StarLine™ Immunoaffinity Columns

Romer Labs® wide range of immunoaffinity columns (IAC) for regulated mycotoxins is based on a specific antibody-analyte binding technology. StarLine™ immunoaffinity columns are designed for simple and reliable cleanups before the detection of mycotoxins in a broad range of foodstuffs and animal feed. Immunoaffinity columns contain a gel bed with toxin-specific antibodies coupled to the gel particles. These antibodies will capture mycotoxins in a sample and release them again after a washing stage. StarLine™ immunoaffinity columns have been validated in an extensive testing program and can be used for the purification and concentration of mycotoxins prior to the analysis by various techniques, such as HPLC, LC-MS, GC-MS, ELISA or direct fluorometry. All Romer Labs® IAC employ highly specific monoclonal antibodies.

Biopure Standards®, Calibrants and Reference Materials

Biopure is a Romer Labs brand which stands for high quality reference materials. All Biopure products are characterized by the appropriate independent methods, such as HPLC, NMR and MS. The selection of the right method is crucial to ensuring the identity and purity of the reference material. Biopure reference materials are accompanied by a certificate. The certificate states the uncertainty of the target analyte and documents the traceability of the certified value, expressed in SI conform units. The Certificate of Analysis is supplied in accordance to ISO Guide 31.Biopure has the broadest range of mycotoxin reference materials currently available globally. In addition to mycotoxins, Biopure supplies standards for veterinary drug residues, and other contaminants such as melamine.The stars of this product line are the fully labeled C13 internal standards, which are unique and rely on a patented technology, proprietary to Romer Labs. Laboratory Equipment

Romer Labs® aims to provide full set-up packages for its diagnostic solutions. Using the right equipment is essential to performing a method correctly. Romer Labs® provides a series of readers for its rapid test kits, such as the AgraVision (for Lateral Flow Tests) or the Chromate (for ELISA tests). For instrumental analysis, Romer Labs® has different instruments available that help improve method performance, such as the RDU (Romer® Derivatization Unit) used for Aflatoxins analysis in HPLC-FLD. Romer® laboratory mills have set the standard in sample preparation in mycotoxin analysis for over 25 years and are referenced in numerous official methods (AOAC, USDA, FAO, etc.) and scientific publications. Many multinational food and feed companies use Romer® mills in their daily routine and the products are known for their reliability and robustness.

EnzymeFast®

EnzymeFast® tests are enzymatic tests in a micotiter plate format. Enzymatic food analysis is a basic method used for the measurement of compounds such as sugars, acids, alcohols and other metabolites in foods and beverages. In general, enzymes are proteins that catalyze the conversion of one molecule into another. Certain enzymatic reactions lead to color formation or consumption that often takes place in a wavelength range invisible to the human eye. This change in intensity can be measured. The high specificity of enzyme reactions allows for the analysis of food components in complex matrices. Enzymatic methods are especially important for food production and quality assurance. Results give information on the nutrients, authenticity and hygienic status of the food. For example, levels of sugars are crucial for consumers suffering from intolerances and the presence of certain metabolites indicates a lack of hygiene during production. It is also possible to detect whether food has been tampered with using enzymatic methods.

Mycotoxins

Many species of fungi can produce a vast number of different mycotoxins. The presence of these toxins in food and feed can be of major concern in food and feed safety. Generally speaking, if elevated levels of mycotoxins are indicated, this can result in toxic, mutagenic and teratogenic effects and lead to symptoms such as skin irritation, immune suppression and neuro-toxicity, as well as to potential birth defects and death. The effects can be chronic or acute, depending on the concentration involved.

Mycotoxins are typically highly resistant to temperature and processing. Therefore, they cannot be destroyed during conventional food or feed production and can also be found in finished, processed products. Analytical procedures to find and identify mycotoxins are essential in ensuring proper risk management. Technologies and products are available for official field or on-site reference testing, depending on the intended use.

Enzymatic food analysis is a basic method used for the measurement of compounds such as sugars, acids, alcohols and other metabolites in foods and beverages. In general, enzymes are proteins that catalyze the conversion of one molecule into another. Certain enzymatic reactions lead to color formation or consumption that often takes place in a wavelength range invisible to the human eye. This change in intensity can be measured. The high specificity of enzyme reactions allows for the analysis of food components in complex matrices.

A food allergy is an immune response to a food allergen that the body mistakenly believes is harmful. Food allergies affect 1-3% of the population as a whole and 5-8% of children, making them an important health issue in modern society. Even minimal exposure to a food allergen in the milligram range can cause symptoms from mild skin rashes to fatal anaphylactic shocks.

Despite the fact that any food could provoke an allergic reaction, the vast majority of documented incidents are caused by cereals containing gluten, crustaceans, eggs, fish, peanuts, soybeans, milk and lactose, in addition to nuts. Other allergens such as mustard, sesame, celery, mollusks and lupines may be of more regional importance. Thus, legislation in many parts of the world requires that these ingredients be labeled on food products independent of their concentration.

Allergens might be present as food ingredients, but what is more dangerous is their unexpected appearance via cross-contaminations during the production process. This means testing needs to be done not only by the authorities for labeling compliance, but also by the manufacturer itself for the on-site control of ingredients and finished products.

Foodborne illness is a serious public health concern around the world. Bovine spongiform encephalopathy (BSE), or “mad cow disease”, is one of several transmissible spongiform encephalopathies (TSEs) or degenerative neurological diseases caused by abnormal prion protein found in the nervous tissues of infected animals that is known to affect various mammalian species including humans. It is spread through the ingestion of infected animal tissue. Most animal health experts believe that the inadvertent contamination of meat and bone meal (MBM) with infected animal tissue and the subsequent use of this material as a feed supplement contribute to the spread of the disease. As a result, the use of processed animal proteins (PAPs) in animal feeds is regulated in many countries around the world.

While feed testing is the only definitive means to certify compliance, regulatory compliance often relies solely on paper certification. Recently, rapid methods have become available for use by regulators to determine compliance during routine inspections. According to the UN, the global area of GM crop plantation has grown 80-fold between 1996 and 2009.”

“Plants of vital importance to agricul-ture are often genetically modified by the insertion of foreign DNA material into their DNA sequence, resulting in expressed novel traits that normally would not appear in nature -- typically herbicide tolerance or insect resistance. Seeds harvested from GMO plants will also contain the modification.

Current GMO production basically cent-ers on four main crops: soybeans, corn, cotton and oilseed rape. On a global level, GMO crops are cultivated on sub-stantial areas of arable land. Approxi-mately 90% of all soy available com-mercially is GMO soy (ca. 80% of corn, ca. 70% oilseed rape and almost 50% of cottonseed).

When testing for GMOs, detection is achieved either through the inserted DNA or expressed protein translated from the novel DNA. The inserted DNA and its corresponding protein are found in all the cells of the plant. Thus, the seeds and/or plant tissue material (e.g. leaves) can be used for GMO testing.

Testing for the expressed protein can be performed using rapid on-site test kits, such as lateral flow devices (LFD) or ELISA, while the reference method for detecting the DNA itself -- using poly-merase chain reaction (PCR) methods -- needs to be done by specialized labs. Romer Labs® offers a wide range of rap-id tests, based on LFD and ELISA tech-nologies, covering the majority of reg-istered GMO traits found on the market, in cooperation with SDIX.

Residues in food and feed are a very heterogeneous group of substances ranging from process contaminants to pesticides, veterinary drugs or even substances added illegally, such as melamine.

Veterinary drugs are used in animal production to treat disease, maintain herd and flock health, promote growth, improve meat or otherwise reduce production costs. There is a possibility that residues or their metabolites may appear in the final animal-based foods with their use. Government authorities control the use of veterinary drugs and monitor food for unsafe or prohibited residues. There are a wide variety of regulations in place, with some veterinary drugs allowed in animal products for food production under certain conditions and others banned from use completely.

Melamine, widely used in combination with formaldehyde to produce melamine resin and melamine foam for countertops, glues, household goods and flame retardants, has been added to raw milk and feed illegally to give the appearance of higher protein content than the actual level. Melamine itself is non-toxic in low doses, but can cause fatal bladder cancer and kidney stones in combination with other chemicals.

Pyrrolizidine alkaloids, plant toxins widely found in the Asteraceae, Boraginaceae and Fabaceae families, can form toxic compounds when metabolized (e.g. in the liver). Several food products such as milk, honey, eggs and herbal teas, as well as medicinal products and animal feed, have been found contaminated with pyrrolizidine alkaloids. Analyzing samples in your own laboratory often raises the question of comparability. How do the results generated compare to other labs using identical samples? The most common way to find out is by taking part in a proficiency testing program or a check sample survey. In such program, participants receive homogenized sample material to be analyzed using in-house laboratory methods. A description of the procedure and the results will be then forwarded to the organizer, following which the results will be compiled and analyzed statistically. Every participant will get a final report, accompanied by an anonymous summary of all results, together with their own individual results. Romer Labs® organizes such programs four times a year for mycotoxins and anyone may take part on a worldwide basis. Naturally contaminated material, homogenized and characterized, will be used as samples for the check sample survey.

When performing an analytical test, strong focus has to be put on the equipment used to obtain accurate and reproducible test results. The first step of any analysis is the use of a well-established sampling plan to obtain a representative sample. After taking the representative sample – for instance, consisting mostly of several aggregated subsamples taken out of a large storage tank – appropriate sample preparation is vital.

This lab sample needs to contain the same concentration of the analyte as the sample from which it was taken. To achieve this goal, two steps are required. First, the representative sample needs to be ground and then a small analytical sample needs to be taken. This is done by using equipment such as mills and/or subsampling devices.

Romer Labs® offers a range of equipment for such analytical purposes, including Romer Mills for grinding and sample division and the Romer Evap® System for the rapid concentration of sample extracts.

Furthermore, quantitative and/or semi-quantitative analyses often require additional equipment for result interpretation. For instance, Romer Labs® offers the AgraVision for lateral flow test interpretation and StatFax or ChroMate from Awareness Technology, Inc. for ELISA quantification purposes.

Last but not least, aflatoxin analysis by HPLC-FLD requires derivatization of the analytes for higher sensitivity; a task made possible by Romer Labs® photo chemical or electrochemical derivatization units.

RDU™ EQOLE1480 Romer® Derivatization Unit The Romer® Derivatization Unit (RDU™) is a photochemical post-column

derivatization for the determination of aflatoxins by HPLC-FLD. For sensitive determination of aflatoxins, the aflatoxins B1 and G1 must be derivatized in order to enhance their natural fluorescence. This may be done with ease photochemically by irradiation with UV light at 254 nm with the RDU™. The method performance is equivalent to an electrochemical derivatization (e.g. Kobra Cell) as shown in the literature. Addtionally, by using the RDU™ there is no necessity for additional purging and lifetime of the HPLC system is increased due to the fact the RDU™ doesn’t require any aggressive reagents to be added to the mobile phase, as this is the case for electrochemical methods.
