Bipolar and abnormal myo-Inositol levels
In manic patients the findings suggest an increase in Inositol levels in the frontal cortexduring the manic phase and and a decrease during the depressive phase

Common Mechanism of Action
Increased growth cone spreading from rat dorsal root ganglia via Inositol depletion mechanism
Williams R, et al, Nature, 2002
![Page 6: Cardiff Presentation 091006](https://reader033.fdocuments.net/reader033/viewer/2022060121/559464dc1a28ab554a8b4573/html5/thumbnails/6.jpg)
All neurites elongate by growth at their distal end in a morphological distinct region called the growth cone
The growth cone integrates external signals acting on the growing neurite and translate them into changes in the rate and direction of growth
The Growth Cone
Harwood AJ, et al., 2004
![Page 7: Cardiff Presentation 091006](https://reader033.fdocuments.net/reader033/viewer/2022060121/559464dc1a28ab554a8b4573/html5/thumbnails/7.jpg)
Enlarged growth cones are often found at sites where the neurite is translocating slowly and making choices about which direction to take
Growth cones have a more narrowed, bullet-like appearance when they move more rapidly
![Page 8: Cardiff Presentation 091006](https://reader033.fdocuments.net/reader033/viewer/2022060121/559464dc1a28ab554a8b4573/html5/thumbnails/8.jpg)
Dorsal Root Ganglion (DRG)
DRG are composed of several thousand cell bodies of somatosensory neurons, involved in perception of pain and other stimuli

![Page 21: Cardiff Presentation 091006](https://reader033.fdocuments.net/reader033/viewer/2022060121/559464dc1a28ab554a8b4573/html5/thumbnails/21.jpg)
Conclusions
• Branching at C-2 of VPA and the derivatization by methylamine of the COOH group of TMCA intensified the growth cone spreading effect
• The potent VPA derivatives and analogs, PIA, DIA and MTMCD, do not stimulate TCF promoter activity, and therefore may regarded as non teratogenic
• The growth cone spreading effect of PIA, DIA and MTMCD is GSK3 independent
