Controllability of PCRTo be able to obtain an accurate and reproducible result in PCR, all variables of this process,being reagents, oligonucleotides, template DNA, plastics and thermocycler, need to be controlled.

The BIOzym group, through CYCLERtest BV and BIOplastics BV, is focusing on the two variableswhich are most often overseen: the PCR plastics and the thermal performance of the thermocycler.

Extreme Uniform plasticsTo control your PCR in the best possible way, so you can obtain the most reproducible andaccurate results, we have developed a state-of-the-art range of PCR disposables called: Extreme Uniform (q)PCR plastics.

These plastics are characterized by Extreme Uniformity in

How is extreme uniformity achieved?Extreme Uniform mouldsIn the designing and developing stage, BIOplastics pays maximum attention to the accuracy of manufacturing the moulds. We allow a minimalspread in the dimensions of the cavities, leading to an average wall thickness of 0.30 mm and a tolerance of 0.05 mm.

Low-cavity moulds If you want a certain tip, tube or plate to have the highest uniformity available, a low-cavity mould should be used in its production. This implies that only a few tips, tubes or plates are moulded at a time, and after that the next few are moulded. On top of that, during releasefrom the mould, each injected product is collected in one bag, resulting in batches with exactly the same specifications. This takes more time, and is more expensive than using multi-cavity moulds from which 32, 64 or even 128 tips or tubes are released in one step. However, these tipsand tubes differ heavily in specification and quality because a multi-cavity mould is less precise than a low-cavity one. To control the process ofinjecting the polymer into the a multi-cavity mould (pressure/temperature) is much more difficult. It is nearly impossible to balance such a mould.This can result in batches of tips, tubes or plates with differences in wall-thickness, orifice and fit, influencing the reproducibility of yourexperiment. BIOplastics products are manufactured using Low- and Semi-Low cavity moulds.

Extreme Uniform manufacturing conditionsDuring the injection molding process all physical, chemical and mechanical parameters are fine tuned to obtain an Extreme Uniform plastic. Our superior in house expertise allows us to determine the optimum and balance betweeninjection temperature of the polypropylene, the temperature of the mold itself during the solidification process, the injection time and theshrinkage time. The injection molding process takes place in a dedicated room under GMP and no-hands-on conditions to ensure the absence of detectable levels of DNA, RNA, DNase, RNase, pyrogens and ATP.

Extreme Uniform raw materialThe polypropylenes used are of the highest medical grade virgin polypropylene, which ensures no release of metals or other contaminants intothe reaction mix. By careful selection of different types of polypropylenes and mixing these into well defined proprietary ratios we, refrain from theuse of softeners, coatings and mould releasing agents.

Wall uniformityExtreme Uniform wall thickness leads to lower evaporation rates and therefore more consistentPCR results. Extreme Uniform wall thickness also leads to even heating of the sample and morehomogenous reaction conditions, leading to more reproducible results.

Wall thicknessPolypropylene is not completely gas tight and evaporation through the walls is a given inevitable.BIOplastics BV designs its tubes with thin walls at the bottom, to allow maximal heat transfer by the thermocycler, and thicker walls at the top to minimize evaporation through the polypropylene,leading to more reproducible results.

Thin Wall Thickness of BIOplastics products are defined as, and measured, 3 mm from thebottom, where as:

Single tubes and strips : 0.30 mm +/- 0.05 mmPlates: 0.35 mm +/- 0.05 mm

High versus low profileIn low profile tubes there is a smaller volume above the reaction mix than in high profile tubes.This allows less reaction mix to go into gas phase, leading to less concentration of the samples,less change in reaction conditions and therefore to more reproducible results.

Lid designCleaver design and sealing rims around the tube closure points provide a leak free sealing,reducing evaporation and leading to more reproducible results. This very tight sealing allows you to run PCR reactions with volumes as low as 5 µl.

Tube angleDifferent thermocyclers brands have also different block well angles. BIOplastics BV has designedits tubes with an angle that fits all main brand thermocyclers to allow maximal heat transfer toobtain optimal results.

Type and blend of polypropyleneAlthough polypropylenes are chemically inert, they still exhibit charged groups, static propertiesand hydrophobic areas. Different types and blends of polypropylene therefore also showdifferences in binding of ions like magnesium, leading to different results. BIOplastics BV offers 3different blends of polypropylene, being O-type, A-type material and M-type material. The O-type material is optimal material used in PCR grade reaction vessels.A-type material resembles the classic polypropylene mix and is available for most of the PCR and qPCR reaction vessels. M-type material is a more robust material which is mainly used for0,5 ml, 1,5 ml tubes and pipet tips. We recommend O-type material for optimal results, as it is the least sticky and shows the highest thermal conductivity.

The influence of tube characteristics on PCR results

SW products: Undefined Swirling productsIt looks like ART, it is never the same in color composition, it is interesting to see and it bringslife to your laboratory! So what are they? They are BIOplastics products which were collected,tested and packed during a change of color of raw materials in our injection mouldingmachines. It results in the same product qualification but what is not defined is the color it self.It may look like swirling from one colour to another, gradient colored, fancy or just strangecolored. Different but with a character. The products can be recognized by SW in the ordernumber.

Trying to crack an EU tube…Cracking of a PCR tube by applying too much pressure is often a major cause of volume loss.The small, microscopic hairline cracks are not visible with the bare eye. We ask you to take anEU tube of any kind between your fingers, squeeze it, and see what happens….. Right, nothing!! Because of the different material composition and the extreme uniform wallthickness EU products are strong and flexible at the same time. No cracking, but easy handling!


High Thermal Conductive PolypropyleneDifferent brands of PCR tubes, strips and plates are made of a wide variety of materials. Our special raw material blend composition selection isbased on optical clarity, impact resistance, softness, gas-tightness, and gamma irradiation withstanding, as well as the new and selectable“binding” characteristics. In general BIOplastics BV has modified the basic mixture in a way that, products have a superior thermal conductivityover other standard materials. The choice of this new HCT (high thermal conductivity) material results in faster thermal transfer from block toreaction mixture. The fact that our products are extreme uniform enables not only fast but also even better thermal transfer and by having thesecharacteristics in place, BIOplastics PCR products can be used in any normal, fast and extreme fast PCR and Real-Time PCR procedure. Generally the faster PCR protocol you run the higher demand you require to use our High Thermal Conductive material.

Colorful world of BIOplasticsBIOplastics’ colorful products brighten up your world and your laboratory. Besides transparent,natural and clear, we offer many of our products in exciting tones and shades. They will not onlyinfluence your mood, but can also be very helpful for coding purposes.

Colored products are manufactured by adding trace amounts of dye to the raw injectionmaterial. Based on extensive testing, we can guarantee that the tint of a tube, strip, plate or pipettip will not affect your experiment in anyway. So, why not cheer up your experiments by usingthem?

To provide you with a rough idea on how our colors really show off, please take a look at thecolor ranges on the right. It might assist you to make a positive, colorful choice!

Besides regular PCR, every EU product, from single tube to plate, is tested for use in Real-TimePCR applications. The caps of EU tubes and strips are designed to enable fluorescent signals topass through to the optical detection unit of the thermal cycler.

For the closure of EU strips and plates, three different versions of cap-strips can be applied. The semi-domed 8-cap strip has a small, optically flat, parallel area. The best choice for Real-Time PCR, however, is closure with the EU optical flat 8-cap strip (B79701 or F79701). Its extremely clear glass-grade area equals the performance properties of optical seals. In contrast to seals, they do not have the disadvantage of adhesives that can interfere with thesignals (higher signal/noise ratios), and exhibit better closure, thus less evaporation. The 8-tubestrips with single attached caps (B79201) have optical, glass grade areas of their own. Most EUproducts are available in white or black and some in “frosted” versions. Tests show significantincreases in signal/noise ratio when using the non-transparent white version and/or frostedversions, compared to natural transparent ones.

Use of EU products in Real-Time PCR

White and Frosted Tubes...why ??Often customers are (still) using regular clear and transparent vessel products in their qPCRapplications. Be aware that you can increase your signal/noise ratio significantly by using whitePCR tubes and plates. Although these higher signal/noise ratios most customers still prefer touse the transparent product since it enables them to view the content of the vessel easily duringthe handling process. By using frosted product both content view ability and high signal/noiseratio are achieved. Frosted products are therefore the most optimal choice for any qPCR applications. BIOplastics BV has a wide variety of frosted PCR plastics available.

Purity and CertificationsProduct purity is, next to excellent design and functionality of products, an important and solidbase that enables customers to generate reliable, reproducible and trustworthy results.

BIOplastics BV products are frequently tested on quality. BIOplastics products comply to thehighest standards and requirements. Depending on product type we can provide batchcertificates as well as individual certificates.

What is sterility?

A sterile product is a product that is free of any living micro-organisms. Sterilization ofBIOplastics products is, in general, done by means of 60Co irradiation. However, irradiatedproducts age faster than non-irradiated products. They are less flexible and break easier,because 60Co irradiation slightly influences the characteristics of the polymer material.Autoclaving the product yourself is much “softer” for the plastic, but when done several times itwill also harden the polymer. One could argue whether it is necessary to autoclave or irradiatePCR disposables at all; BIOplastics products are produced under clean room and “no-hands-on” conditions.

What is DNa(se), RNa(se) and Pyrogen-free?

Producing products under strict clean-room and “no-hands-on” conditions ensures the absenceof DNA, RNA, and their breakdown enzymes. A sterile product is not necessarily DNA(se),RNA(se) and Pyrogen-free. Only regular testing during and after the production processguarantees a DNA(se), RNA(se) and Pyrogen-free product. You can recognize the DNA(se),RNA(se) and Pyrogen-free plastics in this catalogue when you see the thumbs-up icon.

Human DNA and ATP absence?

Producing products under strict clean and “no-hands-on” conditions ensures the absence ofhuman DNA and ATP. Regular testing during and after the production process is supporting the statements.

What is metal-free?

The absence of metals in polymer products like tips and tubes is essential for laboratory use.BIOplastics tips and tubes are made of PP (Polypropylene) of the highest quality available, whichare also guaranteed to be metal free. No mould releasing agents are used during this process.This is possible because the moulds (the template shape in which the molten PP is injected) aremanufactured precisely and with correct angles, enabling an easy release when the product issolidified.

High vs. Low ProfileThe difference between high and low profile products is illustrated in these photographs. More than half of a high-profile strip remains above thethermal block level. Low profile products have a much better ratio between gas and liquid phase (less dead space), thus bringing the chances ofwall-leakage through the gas phase down. However, some thermal cyclers do not have height-adjustable heated lids, so an adaptor is needed incombination with low-profile products in these instruments (see tables on pages 14 - 17). EU products are available in both high and low profile versions.

Tube support grid, make your own plate

Tube support grid holds plates, strips or individual tubes. The tube support grids are availablein a wide range of colors, are alphanumeric marked and have knobs to fix EU tubes strips andplates. Once the tube strips or plates are positioned, and after pipetting the required reagents,the Tube support grid is used as a carrier which can be placed completely in the thermal cycler,during pre- and post the cycling process. These EU 96 well plates are the quality and price alternative, meant for use in any Applied Biosystems regular or Real-time PCR thermal cycler (see tables on pages 14 - 17). They have a very rigid, extra stabilized frame, and an elevated skirt. They are suited for both automatic loading as well as robotic handling. To improve Real-Time PCR signal yields, the tubes in this 96-well plate are frosted.

Closure can be accomplished with either EU optical, strip caps, mats or seals (see PCR strip caps and seals page 35 - 38). When closure is accomplished with flat cap strips, or with one of the EU Seals, the use of EU adaptor B77500 or an ABI expansion or compression mat isrecommended. EU Adaptors

This EU adaptor (96) should be used whenever low profile versions of EU 0.2 ml thin-wall 8-tube strips or 0.2 ml single tubes are used in ABI/PEor Bio-Rad PCR and Real-Time PCR thermal cyclers (see table, page 14 - 17). How to open and close EU PCR products….EU products can be opened and closed many times without breaking, or losing their shape and sealing properties. When using single EU tubes, or strips with single attached caps, closure of the caps should be accomplished by pressing the front face end of the cap slightlybackwards towards the hinge, and not by pushing it from the backside. Cap-strips should be applied in a parallel manner, and in an angle onto the strips and plates, preferably closing all 8 wells simultaneously. No special closing tools are required.

The Importance of Filter MaterialFiltertips are generally accepted as the solution in the prevention of cross-contamination in PCRreactions. A main source of this contamination is the formation of aerosols in the shaft of thepipette used during the PCR liquid handling. Carry-over of previously pipetted DNA can causefalse positive signals.

Filter materials

The use of a filter in a filter tip eliminates the formation of aerosols in the shaft of the pipette,because it blocks the air-to-liquid interface between the sample and the shaft. This simple ideahas evolved in a numerous types and brands of filtertips and filter materials.

In earlier days filters were made of cellulose. Not being inert, this material was not suitable toprevent cross-contamination in sensitive pipetting procedures. Nowadays most filters are madeof 3-dimensional cross-linked PE (Polyethylene), which is totally inert.

Self Sealing

The most important parameter for a filter tip is the pore size of the applied filter material. The pore size must be smaller than 25 µm to protect against aerosols at all. If the pore size is between 20 and 25 µm, a filter will give reasonable protection, however liquid will pass when overloaded. Below 20 µm, a filter will give superior protection. When pore sizes get below 14 µm, it becomes too difficult to pipet liquids in a reproduciblemanner, because of the airflow barrier. Therefore, it makes no sense to use filters with this small pore size.

The costs and investments to manufacture a filter material with an average pore size of 18 µmare high. Only the BIOplastics Self Sealing Non Collapsing (SSNC) filter tip meets thisspecification and is build up with a density gradient from top to bottom. It is the perfect balancebetween superior protection and accurate pipetting.

Filtering Capacity

The filtering capacity of a certain filter is defined as the ratio of filter length and pore size. The longer a filter is, the better it filters. This relation is almost linear. BIOplastics filters have the longest length available in the market when compared to other brands. On the other hand,the smaller the pore size is (down to 14 µm), the better it protects. When these two factors arecombined, the result is a superior filter tip.

3. PIPETTE TIPSThe accuracy of pipetting procedures greatly depends on the quality of the tip used. Buying an expensive accurate pipet makes no sense at all, when the tips used are of inferiorquality. Tips should be made of the highest quality virgin PP (Polypropylene), so they are flexibleand soft to secure a good, leak-free fit around the shaft of the pipette. Fitting bands areessential for the same reason.

All our tips are designed to have a very fine orifice for complete, reproducible pipetting. Some tips, marked with the beveled orifice icon, have a special 45° beveled orifice to guide the rejection of the fluids even better. Tips with special features can be found in the relevantchapters.

3.1 Regular TipsPipette tips for PCR, Low Adhesion, Non-binding, High recoveryThe accuracy of pipetting tips particularly depends on the quality of the tips used. Due to the fact that PCR methods are more and more beingused in diagnostic applications, BIOplastics BV has developed a superior pipette tip for use in any PCR or related high performance technique.BIOplastics BV has used the existing knowledge of superior design and manufacturing capabilities to generate this new highly accurate pipettetips.

Low adhesion and non-bindingBIOplastics BV is using a similar high performance blend of polypropylene as used in EU PCR tubes and plates. By using this material a perfectseal to the pipette is guaranteed, and the inert, and no surface charged PP assures no binding of any charged molecules like DNA, RNA, proteinsetc.. Furthermore, high recovery is achieved due to the mould polish and material characteristics. It means that "all" liquid is pipetted and no orvery limited liquid films will remain in the tip enabling the highest possible sample recovery.

Tips in Multi Purpose Racks (MPR)BIOplastics BV has designed a new type multi purpose racks in which tips are packed. 4. TUBESTubes are the key components in experiments. Tubes are used to prepare and performreactions, and to store the final reaction product. Tubes should be of a trustworthy quality,durable, consistent and stable. BIOplastics tubes are manufactured under strict qualitycontrolled conditions. The design is ensuring smooth inner surfaces, easy closure and properrepeatable results. Most tubes are made of PP unless otherwise indicated. Used separately or together, pre- and post-PCR sampling is made easy with the sampling BoxRack. A BoxRack consist of a Station Box, Station support insert and a Tube support grid. The Station support insert (96 positions, 8 x 12) easily slips in and out of the BoxRack. The Tube support grid can be “clicked” onto the Station support insert. Once placed the Tube support grid holds plates, strips or individual tubes. The Tube support grids are available in a wide range of colors, the grids are alphanumeric marked and have knobs to fix EU tubes stripsand plates. Once the tube strips or plates are positioned, and after pipetting the required reagents, the Tube support grid is used as a carrierwhich can be placed completely in the thermal cycler, during pre- and post the cycling process. After cycling the grid with vessels is removed from the cycler and re-placed in the Station support insert for post PCR work. Stacking aids on both the top and bottom of the BoxRack and the Station support insert provide extra stability for placing them on top of each other. The Station boxes are manufactured of durable and impactresistant polypropylene, which is autoclavable. Boxes can be separated by color in any combination based on e.g. pre- or post PCR separation.
