1.0 PRODUCT INTRODUCTION AND DESCRIPTION

The BioCapture® 650 portable air sampler collects particles into a buffer solution, maximizing the viability of airborne microorganisms and enabling users to quickly collect samples for subsequent analysis.

The collector uses patented rotating impeller arm technology to impact airborne particles, and then automatically wash the surface with a fluid. Manifold geometries that allow the air to be exhausted separately from the particle laden liquid have been optimized for minimum evaporation and maximum flow rate. The motor and control systems have been optimized for high impeller revolution rate and low power consumption while maintaining excellent reliability.

The BioCapture is deployment ready out of the box. The disposable fluidics cartridge, and integrated pumps, and control logic eliminate sample preparation. Users simply press a button to initiate the sampling process. The collected sample may be used with various detection methods including PCR, hand-held assays, and traditional culturing.

Time savings is also realized in the decontamination process. Upon completion of the sampling mission, the disposable BioCapture cartridge eliminates carry-over contamination risks. There is no possibility of introducing contaminants from a prior mission into a future sample when the entire mechanism is replaced. Users wipe the instrument with a bleach solution, air dry, replace the cartridge, and shelve for future use.

5.0 SAFETY PRECAUTIONS

Read all instructions before operating the unit

Use only the included batteries and charger

Do not touch the impeller shaft when power is on

Use caution when handling all electrical equipment

Do not attempt to disassemble, repair or modify the unit

Do not expose unit to temperatures above 60°C (140°F)

Do not handle damaged or leaking batteries

Do not submerge unit in liquid

Do not use unit in direct rain as cartridge may overflow

6.0 BATTERY INSTALLATION

Ensure the battery is fully charged before installation (Section 12.0 BATTERY AND CHARGER). A fully charged battery provides approximately two hours of sampling time.

CAUTION: Leave the Training Cartridge attached to the unit while installing the battery to protect theAttachment Latches and Impeller Shaft from damage.

Step 1: Hold the BioCapture upside-down in one arm.

Step 2: Turn the attachment ring counter-clockwise ¼ turn to unlock the battery door (Figure 6).

Step 3: Fully open the battery door to access the battery compartment.

Step 4: Slide the battery label-side up into place inside the compartment.

Step 5: Close and lock the battery door by turning the attachment ring clockwise ¼ turn.

Step 5: Proceed to Section 7.0 COLLECTION CARTRIDGE ATTACHMENT.

7.0 COLLECTION CARTRIDGE ATTACHMENT

The Collection Cartridge is a single-use, disposable cartridge with a removable Sample Vial forsubsequent analysis. Both the cartridge and vial have stickers with matching numbers to facilitate identification (Figure 5). Always begin sample collection with a new, unwrapped cartridge. Collection Cartridges have a one year shelf-life. Check the expiration date printed on the cartridge sticker before attachment.

WARNNG: Do not reattach a previously used cartridge after it has been removed from the unit.

Step 1: Place the BioCapture onto a flat, secure surface.

Step 2: Remove the Training Cartridge. Place index fingers on each Attachment Latch and hook the remaining fingers underneath the edge of the cartridge (Figure 7, left).

Step 3: Press in the latch while gently pushing out on the cartridge to detach the cartridge from the unit (Figure 7, right).

Step 4: Lift the cartridge up and away from the unit.

Step 5: Store the cartridge in the carrying case.

Step 6: Press and release the Power Button. The Start/Stop LED will illuminate green. The Mode LED will illuminate green indicating the time used during the previous sample collection. The LCD will briefly display in sequence: “Firmware v5.01J”, “MesoSystems BioCapture 650”, before displaying “Attach Cartridge”. A battery bar and the previous sampling time used will also be displayed.

Step 7: Unwrap a new Collection Cartridge; discard all packaging.

WARNING: Always press the Power Button before attaching a cartridge. The instrument has been designed with an automatic alignment feature to ensure the pump actuators are properly aligned with the cartridge. If a cartridge does not attach it is usually because the actuators are not aligned.

Step 8: Place the new cartridge over the attachment latches (Figure 8, left).

Step 9: Press the cartridge down evenly until it snaps into place.

Step 10: Check both sides of the cartridge to ensure the cartridge is securely attached (Figure 8, right).

The LCD will change from “Attach Cartridge” to “Ready” after the cartridge is attached (Figure 9).

8.0 SAMPLE COLLECTION

Airborne particles are drawn through the air intake into the Collection Cartridge during SAMPLE COLLECTION. The user has a choice of three sampling times—5, 15 and 30 minutes. A 60 minutesampling time is also available when all four MODE LEDs are illuminated. Collected particles are automatically rinsed from the cartridge into the Sample Vial when 02:30 is left in the cycle.Approximately 2-7 mL of sample fluid will remain in the vial for subsequent analysis.

WARNING: The unit may emit aerosols and small water droplets from the air exhaust during sample collection due to the rapid rotations of the motor and pumps. This is expected and is non-hazardous.

Step 1: Press and release the Mode Button until the LED above the desired sampling time illuminates green. Each LED will illuminate once then all four LEDs will illuminate to indicate the 60 minute sampling time before cycling through again. The LCD will display the selected time.

WARNING: If the battery level is too low to complete the time selected, the Mode LED will turn red,the LCD will display “Insuff Battery”, and the Start/Stop LED will flash red. Replace the battery or select a shorter sampling time to proceed.

Step 2: Press and release the Start/Stop Button. The Start/Stop LED will blink green. The motor, fan and pump will turn on and the LCD will display “Initiating…” for 10 seconds.

A pre-sampling diagnostic cycle verifies the power level, cartridge installation and motor function before sampling. The LCD will display a diagnostic message if the unit does not pass the initial diagnostics (Section 16.0 TROUBLESHOOTING).

The unit automatically begins sampling after the pre-sampling diagnostics are complete. The Start/Stop LED will blink green; the LCD will display “Sampling…” and count down the collection time remaining.

Particles will be rinsed from the cartridge with 02:30 remaining. At 00:00, the LCD will change to “Recovery…” the Start/Stop LED will blink rapidly; the time will change to 01:19 and begin a new count down.

SAMPLE COLLECTION is complete when the LCD changes to “Sample In Vial” and the Start/Stop LED remains solid green. The Dispense LED will illuminate solid green. The following options are now available:

WARNING: Do not remove the cartridge before performing the TEST STRIP CYCLE.

• Remove the cartridge to access the Sample Vial for sample analysis (Section 10.0 COLLECTION CARTRIDGE REMOVAL)

• Replace the cartridge to perform another SAMPLE COLLECTION

9.0 TEST STRIP CYCLE

The BioCapture sample fluid is compatible for use with hand-held assays. The unit has also been designed with a Test Strip Port to accommodate commercially available strips for automated dispense. Contact Customer Support for a list of test strip manufacturers.

CAUTION: Leave the Collection Cartridge in place following SAMPLE COLLECTION to perform this cycle. Visually confirm the Sample Vial is at least half-full before dispensing. If the vial is less than half-full it may be possible to drain the sample vial such that there is not enough sample remaining in the vial for laboratory analysis.

During the TEST STRIP CYCLE fluid lines are first primed. After the lines are primed, a test strip is inserted into the Test Strip Port located on the side of the Collection Cartridge. Sample fluid is then drawn from the Sample Vial and deposited directly onto the strip for in-field analysis. Up to eight (8) test strips may be tested. Always use new, unwrapped test strips and refer to the test strip manufacturer’s instructions when performing this cycle.

NOTE: Sample fluid will remain in the lines after priming. This fluid is not recoverable back to the vial.

Finish SAMPLE COLLECTION (Section 8.0) before starting. Both the Start/Stop LED and the Dispense LED will be illuminated green and the LCD will display “Sample In Vial”.

NOTE: If the Sample Collection is stopped mid-cycle the LCD will display “Partial Sample” and the Start/Stop LED will blink alternately green then red.

Step 1: Press and release the Dispense Button. The Dispense LED and the Start/Stop LEDs will remain green, the LCD will display “Flushing…”, the pump will turn on and the timer will count down from 00:35. When flushing is complete the LED will begin to blink green slowly and the LCD will change to “Ready” and the pump will turn off.

WARNING: Do not insert the test strip until priming is complete and the LCD displays “Ready”. Inserting the test strip before this point will result in the test strip overflowing.

Step 2: Unwrap a new test strip and discard packaging.

Step 3: Hold the test strip right-side up as per the manufacturer’s instructions and insert into the Test Strip Port (Figure 10).

Step 4: Press and release the Dispense Button. The Dispense LED will blink green rapidly, the Start/Stop LED will remain solid, the LCD will display “Dispensing…” and the timer will count down from 01:00. The pump will turn on and automatically meter the appropriate amount of sample directly onto the test strip for three seconds. The LCD will continue to count down to zero (00:00).

WARNING: Do not move the unit or remove the test strip while the Dispense LED is blinking and the LCD displays “Dispensing...” to ensure the fluid thoroughly soaks the strip.

The TEST STRIP CYCLE is complete when the Dispense LED returns to a slow blink, the LCD displays “Disp. Complete” and the time is at 00:00.

Step 5: Carefully remove the test strip.

Step 6: Label as appropriate.

Step 7: Repeat steps 3-7 to run additional test strips.

NOTE: The LCD will not display “Flushing…” during subsequent test strip cycles and the time will count down from 01:00.

10.0 COLLECTION CARTRIDGE REMOVAL

The Collection Cartridge should be removed only after sample collection is complete. Once removed, the cartridge may not be reused. Follow your Standard Operating Procedures (SOPs) when handling sample.

Step 1: Place index fingers on the Attachment Latch and hook fingers underneath cartridge.

Step 2: Press in the latches while gently pushing out the cartridge to release the side of the cartridge from the unit.

Step 3: Lift the cartridge up and away from the unit.

Step 4: Support vial with thumb and, bend the vial support down then fully backwards to access the Sample Vial (Figure 11, left).

Step 5: Gently remove the Sample Vial ensuring the sample does not spill (Figure 11, right).

Step 6: Snap the attached vial cap firmly in place. Discard the cartridge as appropriate.

11.0 TRAINING CARTRIDGE

The Training Cartridge has been provided to help familiarize the user with the BioCapture 650-01 by simulating the unit’s operations, LCDs and LED indications. Labeled to distinguish it from the Collection Cartridge, the Training Cartridge does not have liquid or rotating impactors and may be used repeatedly. It is also used as a protective cover for the impeller and sensors when the unit is in storage (Section 14.0 STORAGE).

Note: The Training Cartridge sounds significantly different from the Collection Cartridge duringoperations.

12.0 BATTERY AND CHARGER

The BioCapture is powered by a specially designed, rechargeable lithium-ion battery. The battery has a fuel gauge check feature on the bottom right side (Figure 12). Press the Check Button on the battery to determine the charge level.

Approximately three hours are required to fully recharge a battery. A fully charged battery will operate the unit for approximately two hours.

Recharge batteries at room temperature (10ºC-30°C, 32°F-86ºF) for best results.

CAUTION: Always remove the battery from the BioCapture when not in use. A battery with a low charge left in the instrument will lose its charge over the course of several days.

Step 1: Insert the plug end of the power cable into the DC power jack on the charger and the AC plug into a standard 110V wall outlet.

Two batteries may be placed in the charger at the same time. Depending on the charge level of the batteries either both will charge at the same time or first one battery will charge while the other is on standby. The second battery automatically begins recharging after the first battery has been charged.

WARNING: Always remove the battery from the unit before transporting. Failure to remove the battery will invalidate the warranty and may cause catastrophic damage to the internal parts requiring unit’s return to the factory for service and repair.

NOTE: In order to keep the battery fuel gauge as accurate as possible it is necessary to occasionally run the battery through a recalibration cycle. To do this, place the battery in the left bay of the charger and press the button on the front label. Recalibration may take up to nine (9) hours.

13.0 DECONTAMINATION

Wipe down the outside of the BioCapture with a cloth dampened with a 3% solution of household bleach to decontaminate. Clean the area around the pumps with a cotton swab dampened with a bleach solution. Always wear appropriate protective clothing when decontaminating the unit.

CAUTION: Never submerge the BioCapture in liquid or leave liquid droplets on the unit’s surfaces to ensure liquid does not reach the motor. If liquid reaches the motor the unit may shut down and require repair. The cloth and cotton swab should be damp but not saturated when wiping down the unit.

Step 1: Dilute 3 mL of household bleach with water to form 100 mL of 3% bleach.

Step 2: Remove and discard the Collection Cartridge (if attached).

Step 3: Wipe down the unit with a cloth dampened with the bleach solution.

Step 4: Carefully wipe the pump, valves and impeller shaft with a cotton swab dampened with the bleach solution.

Step 5: Wipe all areas of the unit again with a cloth or cotton swab dampened with sterile water to remove bleach residue.

Step 6: Allow the unit to air dry thoroughly.

Step 7: Attach the Training Cartridge.

Step 8: Remove the battery.

Step 9: Put the BioCapture into its case.

Store the BioCapture and accessories as recommended below for best results.

BioCapture 650

Store the unit in its case with the Training Cartridge attached in temperatures –46°C to 60°C (-51°F to 140°F).

Lithium-Ion Battery

Remove the battery from the BioCapture before storing. Store the battery in temperatures 0°C to 35°C (32°F to 96ºF) for longest shelf life. When transporting, protect from freezing.

Collection Cartridge

Store the cartridge, wrapped, at room temperature. When transporting, protect from freezing.

17.0 ATTACHMENT LATCH REPLACEMENT

The cartridge attachment latches have been designed for field replacement. An Attachment Latch Repair Kit with two (2) replacement latches and two (2) screws (Figure 13) are included with the unit.

Step 1: Remove the screw holding the broken attachment latch in place (Figure 14, circled left) using the 2.5mm hex head Allen wrench provided in the kit. Discard broken attachment. Set screw aside to use with new latch or discard.

Step 2: Inspect the area around the removed latch and alignment boss (Figure 15, circled) for cracks or damage.

Step 3: Remove a new latch from the replacement kit.

Step 4: Place new latch on handle, fitting it over the alignment boss (Figure 16).

Step 6: Replace the screw removed in Step 1 with either the removed screw from the broken latch or a new one out of the attachment latch kit.

Step 7: Tighten the screw with the Allen wrench to secure new latch in place (Figure 17).

19.0 WARRANTY

Limited Warranty and Remedy. ICx Technologies warrants that, upon delivery, each Product will be free from defects in materials and workmanship and will operate in all material respects in accordance with applicable instructions and manuals provided by ICx. Coverage under this Limited Warranty shall commence immediately upon delivery and is valid for a period of one (1) year from the date of the invoice. This warranty will not apply to any Product that (a) has not been operated and maintained in accordance with applicable instructions and manuals, (b) has been repaired or altered by unauthorized personnel, or (c) has been misused, abused, damaged or subjected to operation for which it was not intended. ICx will use commercially reasonable efforts to correct any failure of any Product that does not substantially conform to the foregoing warranty (e.g., by modification or repair of the noncomplying Product) provided that Customer gives ICx written notice of the noncompliance within thirty (30) days after delivery. If Customer gives ICx notice of any such noncompliance in accordance with the foregoing and ICx fails to correct the noncompliance within a reasonable period of time after receipt of such notice, then Customer may return the Product and elect to either (i) receive a full refund of any amount paid by Customer for the Product or (ii) require ICx to promptly deliver a replacement Product, in either case without cost to Customer.
