Introduction Bread wheat (Triticum aestivum L.) is the first important and
strategic cereal crop for the majority of world's populations and
Egypt as well. Stripe or yellow rust, caused by Puccinia
striiformis f. sp. tritici, is an important wheat disease and causes
considerable yield losses in wheat growing areas worldwide.
The objectives of the present study were to evaluate the
monogenic lines (Yr's) and candidate wheat genotypes under
greenhouse and field condition. In addition to identify of
resistance genes by three DNA markers.
Materials and Methods The present work was carried out at Sakha Agricultural
Research Station under greenhouse and field condition.
Fourteen monogenic lines and thirteen wheat genotypes
(nine Egyptian wheat cultivars and four entries from
CIMMYT-8STEMRRS) were evaluated at the seedling
stage under greenhouse conditions and the same germplasm
was also evaluated under field conditions against stripe rust.
In addition, Yr9, Yr17 and Yr18 resistance genes were
identified by molecular marker (Robert et al. 1999).
Results and Discussion Data showed that Yr1, Yr5, Yr10, Yr15, Yr17, Yr32 and
YrSP monogenic lines exhibited high resistance against
stripe rust reaction at seedling response, where the infection
type ranged from 0 to 2 with two races i.e. 198E56 and
128E28 (Table 1). Data also showed that at adult stage only
Yr1, Yr5, Yr10, Yr15, and Yr32 gave disease severity with
complete resistant reaction to stripe rust followed Yr17,
YrSP and YrSk, where their disease severity were 5MR-
10MR. Nine Egyptian cultivars and four lines from CIMYT
tested at seedling stage were susceptible (IT 6-8). Misr-1,
Misr-2, Gemmezia-10, Line 6043, Line 6085. Line 6086
and Line 6107 showed a ranged of adult plant responses
TrR to 10MR and the remainder had variable degree
susceptibility 5MS to 30S. Thirteen genotypes were
screened with three DNA markers aimed at detecting the
presence of Yr9, Yr17, and Yr18. The 1100bp diagnostic
for Yr9 was present in 7 genotypes i.e. Misr-1, Misr-2,
Sids-12, Sids-13, Gemmeiza-9, Gemmeiza-10 and
Gemmeiza-11 a 252bp fragment in Line6043 (8STEMRRS)
from CIMMYT was indicative of Yr17, (Fig. 1), and Yr18
was present in most Egyptian and CIMMYT genotypes
tested.
