1. 3300 ELSD CHEMSTATION DRIVER INSTRUCTIONS

1.1 REQUIREMENTS

• Minimum PC requirements: Operating System: Microsoft® Windows® XP (Service Pack 2) or Windows® 2000 (Service Pack 4) Processor Speed: 1.5GHz Memory: 512 MB RAM Hard Disk: 40GB Ethernet Port

• Agilent ChemStation software, version B.02.01 SR 1 or above. Earlier ChemStation versions may require upgrade for compatibility; contact your Agilent representative for details.

• Microsoft® Installer 3.1 Software. To determine whether your PC already has the software installed, select “Add-Remove Programs” from the Windows® Start menu and view the list under “Currently installed programs”. This software can be downloaded from the Microsoft® website (Download Center) by clicking here

• Microsoft® .Net Framework Version 2.0 Software. To determine whether your PC already has the software installed, select “Add-Remove Programs” from the Windows® Start menu and view the list under “Currently installed programs”. This software can be downloaded from the “3300 ELSD Driver for Agilent ChemStation” CD (by clicking on “.NET Framework 2.0”) or from the Microsoft® website (Download Center) by clicking here.

• Alltech® 3300 ELSD ChemStation Driver Software. This software can be downloaded from the “3300 ELSD Driver for Agilent ChemStation CD” (by clicking on “3300 ELSD ChemStation Driver”) or from the Grace Davison Discovery Sciences www.elsd.com website (by selecting “ELSD Software Update”) by clicking here.

• Alltech® 3300 ELSD with Application software version 1.0.20 or above and OS version 1.0.0 or above. To check the current software versions on the 3300 ELSD, select “Maintenance/About” on the front panel using the keypad. If an upgrade is required, it can be downloaded from the Grace Davison Discovery Sciences www.elsd.com website (by selecting “ELSD Software Update”) by clicking here.

• Ethernet Cable(s). The type of ethernet cable(s) required will depend on the system configuration. If the system includes a 3300 ELSD with ChemStation-controlled Agilent instrumentation, then standard ethernet cables should be used to make the following connections: from the 3300 ELSD to a hub/switch, from the Agilent system to a hub/switch, and from the hub/switch to the PC. If the system includes a 3300 ELSD without other ChemStation-controlled instrumentation, then a crossover ethernet cable can be connected directly from the 3300 ELSD to the PC. NOTE: It is NOT possible to piggyback the cabling of the 3300 ELSD directly to the Agilent instrumentation; a hub or switch must be used instead.

• Ethernet Hub or Switch, ONLY if the system includes a 3300 ELSD with ChemStation-controlled Agilent instrumentation. A hub or switch is NOT required for a system that includes a 3300 ELSD with no other ChemStation-controlled instrumentation.

1.2 INSTRUCTIONS

1. First, confirm that the 3300 ELSD has the proper software versions installed by selecting “Maintenance/About” from the front panel keypad. The 3300 ELSD must have Application software version 1.0.20 or above, and OS software version 1.0.0 or above to be compatible with the ChemStation driver. If the unit requires a software upgrade, the upgrade must be downloaded from the Grace Davison Discovery Sciences www.elsd.com website (by selecting “ELSD Software Update”) and installed onto the unit before continuing with the driver installation. This software can be downloaded directly from the website by clicking here. Follow the instructions included with the download to install the software onto the 3300 ELSD. Once the 3300 ELSD has the proper software versions installed, continue on to step 2.

2. Confirm that the PC has the minimum requirements listed in Section 1.1 above.

3. Determine if Microsoft® Installer 3.1 Software is already installed on your PC by selecting “Add-Remove Programs” from the Windows® Start menu and viewing the list under “Currently installed programs”. If the software is not already installed on the PC, download and install the software program from the Microsoft® website (Download Center) by clicking here.

4. Determine if Microsoft® .NET Framework Version 2.0 is already installed on your PC by selecting “Add-Remove Programs” from the Windows® Start menu and viewing the list under “Currently installed programs”. If the software is not already installed on the PC, download and install the software program from the “3300 ELSD Driver for Agilent ChemStation” CD (by clicking on “.NET Framework 2.0”) or from the Microsoft® website (Download Center) by clicking here.

5. Next, install the 3300 ELSD ChemStation Driver onto the PC where ChemStation is installed either by clicking on “3300 ELSD ChemStation Driver” on the “3300 ELSD Driver for Agilent ChemStation CD” or by left double-clicking on the “setup.exe” file included in the download from the www.elsd.com website. Follow the instructions of the installation procedure as it proceeds:

a. First, the “Welcome to the ELSD 3300 Setup Wizard” window will appear. Click on “Next” to continue on with the

installation. b. The “Select Installation Folder” window will then appear. To check for available memory, press “Disk Cost”. To

save to an alternate folder other than the one displayed, press “Browse”. Otherwise, press “Next” to continue on with the installation.

c. The “Confirm Installation” window will then appear. Press “Next” to continue on with the installation. d. The “Installing 3300 ELSD” window will then appear. A timer bar will appear showing the progress of the

installation.

e. The “Installation Complete” screen will then appear. Press “Close” to complete the installation.

6. Make the proper connections between the instruments and the PC, depending on the system configuration:

a. For a system with a 3300 ELSD and ChemStation-controlled Agilent instrumentation: The following connections should be made using standard ethernet cables: 1) from the 3300 ELSD to a hub/switch, 2) from the Agilent system to a hub/switch, and 3) from the hub/switch to the PC. NOTE: It is NOT possible to piggyback the cabling of the 3300 ELSD directly to the Agilent instrumentation; a hub or switch must be used instead. b. For a system with a 3300 ELSD and no other ChemStation-controlled instrumentation: A crossover ethernet

cable can be connected directly from the 3300 ELSD to the PC. 7. Configure the network settings on the PC:

a. On the PC, select “Network/Local Area Connection/Properties” from the Start menu to bring up the “Local

Area Connection Properties” window:

b. Select “Internet Protocol (TCP/IP)” from the list to highlight it, and then select “Properties” to bring up the

“Internet Protocol (TCP/IP) Properties” window:

c. If the TCP/IP settings have already been entered, write down the settings for “IP address”, “Subnet mask”, and “Default gateway”. These settings will be refered to when configuring the network settings on the 3300 ELSD.

d. If the blanks are empty, then the TCP/IP settings will need to be entered. The following settings could be

used as an example: “IP address: 10.1.1.100”, “Subnet mask: 255.255.255.0”, and “Default gateway: 10.1.1.100”. Write down the settings that have been entered; they will be referred to when configuring the network settings on the 3300 ELSD. Then press “OK” to save these settings and close out of the network configuration window.

8. Check the IP addresses for any Agilent instruments controlled by ChemStation (if any) in the ChemStation Configuration Editor. Write these down for reference; they will be referred to when configuring the network settings on the 3300 ELSD.

9. Configure the network settings on the 3300 ELSD:

a. Select “Configuration/Network” on the front panel of the 3300 ELSD:

b. The Network Configuration window will appear. For the “DHCP” setting, select “Disabled”. Next enter the “IP Address”, “Subnet Mask”, and “Gateway” settings. These addresses should be based on the network settings on your PC. For example, if the settings on the PC (written down in step 7c or 7d above) are: PC network settings IP Address: 10.1.1.100 Subnet Mask: 255.255.255.0 Default Gateway: 10.1.1.100 Then the network settings on the 3300 ELSD could be: 3300 ELSD network settings: IP Address: 10.1.1.101 (same as PC network setting except fourth value is incremented by one***) Subnet Mask: 255.255.255.0 (same as PC network setting) Gateway: 10.1.1.101 (same as 3300 ELSD IP address) ***EXCEPTION: Make sure the IP address for the 3300 ELSD does not conflict with any of the the IP addresses for Agilent instruments in ChemStation (written down in step 8 above). For example, if the IP address of 10.1.1.101 was already in use on one of the Agilent instruments, then the 3300 ELSD could then use IP address 10.1.1.102 instead (same concept – the first three values are the same, but the fourth value is incremented). The same address cannot be used for more than one instrument in the ChemStation system configuration.

c. Press “Accept” and the Network Configuration will begin.

d. If the error message below appears, try repeating the network configuration, and make sure that the Ethernet

cable is connected between the 3300 ELSD and the PC or hub/switch.

10. Select “Operation/Mode/Run” on the front panel of the detector to put the detector into Run mode before connecting to ChemStation. This will allow you to view the Output and Chromatogram Trace on the front panel while running ChemStation, since the display will lock once the detector is connected to ChemStation.

11. Configure the 3300 ELSD settings in ChemStation:

a. Open the ChemStation Configuration Editor window:

b. Select “Configure/Instruments” from the drop-down menu at the top of the screen to bring up the “Select

Instrument” window:

c. Press the “OK” button to bring up the “Device Configuration” screen:

d. In the “Modules” window, under “Common Instr. Ctrl”, click on “Access Point” to highlight it, and then click on the “Add” button to add it to the “Selected Modules” window:

e. Click on the “OK” button to return to the “Configuration Editor” screen with the “Common Instr. Ctrl./Access Point” module listed. f. Select “File/Save” from the drop-down menu at the top of the screen to save the new settings:

g. Select “File/Exit” from the drop-down menu at the top of the screen to exit from the “Configuration Editor” screen:

12. From the PC desktop, open “Agilent ChemStation/Instrument 1 Online” (or appropriate Instrument number).

a. If this is the first time using the 3300 ELSD Driver, the 3300 ELSD Configuration window will appear automatically, so proceed to step 12c. Otherwise, once ChemStation opens, select “View/Full Menu” to ensure all driver options will be displayed.

b. Select “Instrument/Configure CIC Access” from the drop-down menu at the top of the screen.

c. The “CIC Devices Configuration” window will then appear:

d. Click on the “Grace 3300 ELSD” icon on the left side of the window, under “Available Modules and Instruments”, to

highlight it: e. Click on the “Add” button to add the “Grace 3300 ELSD” icon to the right side, under “Configured Modules and Instruments”:

f. Click on the “Grace 3300 ELSD” icon on the right side, under “Configured Modules and Instruments” to highlight it. g. With the “Grace 3300 ELSD” icon highlighted, press the “Configure” button, and the “Configuration ELSD” window

will appear:

h. In the “Configuration ELSD” window, select the “Run Trigger” setting. Select “Start run on external trigger signal” if

the start signal will be connected to the Start input pins on the back panel of the 3300 ELSD (pins 1 (-) and 2 (+)). Select “Start run on software signal” if an Agilent pump or injector will be the source for the start signal.

i. Select the “Network Configuration” settings. Either select “IP Address” and enter the IP address in the blank

(10.1.1.102 is shown above as an example). This IP address must match the IP address that was configured on the 3300 ELSD in step 9b. Or, select “Host Name” and enter the Host Name into the blank. Press Check to test the network settings.

j. Press “OK” to save the settings in the “Configuration ELSD” screen. k. Press “OK” to save the configuration in the “CIC Devices Configuration” screen. l. The message “Changed Configuration will become active after restart of ChemStation” will appear. Press “OK” and

then close, and re-open ChemStation.

m. Once ChemStation re-opens, the 3300 driver has been configured and is ready to use. Continue on in the following sections for details on using the features of the 3300 ELSD driver.

1.3 3300 ELSD INSTRUMENT ACTUALS

1. Select “View/Instrument Actuals” to view the status window of the 3300 ELSD. 2. The “Grace ELSD Status” window will appear. The window provides information on the Temperature, Gas Flow, Gain,

and Mode settings of the 3300 ELSD, as well as indicating any errors.

3. The status window will also indicate if the 3300 ELSD is “Equilibrating”. If a ChemStation run has been started, the run

will not begin until the 3300 ELSD is done equilibrating. Equilibrating conditions are met once the temperatures for the lower drift tube, upper drift tube, and optics block zones are within 1.5oC, and the gas flow is within 0.3L/min for at least 60 seconds. Once the 3300 ELSD is done equilibrating, the status window will display “Pre-run” until the injection takes place. Once the run has started, the status window will display “Running”. The status window will display “Power Fail” if the 3300 ELSD driver is not configured properly or if communication has been lost (such as if Ethernet cable is disconnected or 3300 ELSD is powered off).

1.4 3300 ELSD CONTROL The ELSD Control function can be used to set the Operation Mode on the 3300 ELSD, or to Autozero the 3300 detector. These functions are not available if a run is in progress.

1. Select “Instrument/More Grace ELSD/Control” from the drop-down menu at the top of the ChemStation screen.

2. The “Control ELSD” window will appear. 3. Select the “Operation Mode” of the 3300 ELSD: Standby, Operational, Heating, or Cleaning.

4. Press the “Autozero” button to autozero the signal output on the detector.

5. Press “OK” to return to the main ChemStation screen with the new mode setting in effect on the 3300 ELSD.

1.5 SIGNAL WINDOWS 1. Select “View/Online Signals/Signal Window 1 (or 2)” to display the Signal Window.

2. The Signal Window will then appear, with the header “Online Plot”. 3. To select the parameters to display, press the “Change” button at the bottom of the screen. The “Edit Signal Plot”

window will then appear.

4. In the left column, under “Available Signals” select a parameter to display by highlighting it. Then press the “Add” button

to add the parameter to the right column, under “Selected Signals”. It is possible to view traces for 3300 ELSD Signal Voltage, Lower Drift Tube Temperature, Upper Drift Tube Temperature, Optics Temperature, Gas Flow, and Nebulizer Gas Pressure.

5. Enter the parameters for the x-axis and y-axis scaling as indicated.

6. Press “OK” to exit the editing window and return to the plot with the newly selected parameters displayed.

7. If multiple traces have been selected, click on the parameter in the top left corner of the screen to select the proper y-axis scaling.

1.6 3300 ELSD METHOD The ELSD Method window can be used to set the method parameters on the 3300 ELSD.

1. Select “Instrument/Set up Grace ELSD” from the drop-down menu at the top of the screen:

3. Enter the values for Temperature, Gas Flow, and Gain using the arrows or by entering the values into the blanks. 4. The Method Wizard can provide assistance in determining initial 3300 ELSD settings based on the HPLC method

conditions. Press the “Wizard” button to open the Method Wizard:

a. The first window in the Method Wizard is “Select Mobile Phase Solvents”. Select the solvents from the drop-down list and enter a % value for each. Press “Next” to continue on in the wizard.

b. The second screen in the wizard is the “Mobile Phase Flow Rate” window. Select the mobile phase flow rate using

the drop-down arrow. Press “Next” to continue on in the wizard.

c. The third screen in the wizard is the “Sample Concentration” window. Select the sample concentration range using

the drop-down arrow. Press “Next” to continue on in the wizard. d. The fourth screen in the wizard is the “Finished” window, which lists the recommended settings based on the

solvent and sample information. Press “Finish” to return to the “Grace ELSD Method” window with the new method settings listed.

5. Select the “Stop Time” setting. Select “As Pump/Injector” if the stop signal will be obtained from an Agilent pump or

injector. Enter the Stop Time manually if the stop signal will not be obtained from Agilent instruments.

6. Check the “Autozero before run” option to autozero the detector before each run. If unchecked, the detector will not perform an autozero before a run.

7. Enter timed events (if any) in the Events Table. Note: The proper wiring must be connected to the back panel output pins on the 3300 ELSD for Control Relay events. Consult the 3300 ELSD Manual for more details.

a. Enter the time value for the event in minutes.

b. Select the type of event (Change Gain or Set Control Relay) using the drop-down arrow.

c. Choose the setting for the event. Select a gain setting (1, 2, 4, 8, or 16) for a Change Gain event. Select On or Off for a Set Control Relay event.

d. Press “Insert” to add the event in the table.

e. To edit an event, click on the event to highlight it, modify the parameters in the editing window, and then press “Edit”.

f. To delete an event, click on the event to highlight it, and then press “Delete”.

1.7 LOGBOOK The Logbook feature in ChemStation displays the actions that have taken place in ChemStation, including on the 3300 ELSD.

1. Select “View/Logbook/Current Logbook” to view the Logbook.

2. The Current Logbook is then displayed. The Logbook displays information for the 3300 ELSD such as when the unit has

been powered on/off, when method settings have been changed, when the mode has been changed, etc.

1.8 HELP SCREENS Help is available for the 3300 ELSD ChemStation driver by pressing the Help button in the following screens: Grace ELSD Method, Method Wizard, Control ELSD, and Configuration ELSD. Below is a sample help screen for setting 3300 ELSD Method Parameters. We have used our best efforts to ensure the accuracy of this Manual.
