● GDF11 is most likely one of the most important blood borne substances that control aging, but by no means the only one. Otherwise we’d have “GDF11 outliers” that live hundreds of years.

Mechanisms of action of GDF11 ● Stems cells incur considerable DNA

damage in replication

● Eventually the DNA damage is so severe the stem cell becomes senescent

● GDF11 repairs the DNA damage and restores function to the stem cell. See graph on right

● Note that GDF11 has little effect on the young, though it may greatly slow aging

● Another MOA for GDF11 is thought to be moving progenitor cells into an active state

Why I decided to become patient zero on GDF11• GDF11 mimics parabiosis and was shown to improve 3 key systems in

mice cardiac, neural and muscular which makes it very compelling.

• GDF11 is endogenous which inherently makes it safer than any man made substance.

• Even the most dangerous endogenous hormones, insulin and thyroid, are not lethal in nano quantities.

• The molecular structure of GDF11 is conserved across all vertebrate species, which shows how important it is to life and how early it showed up in evolution.

• Organs transplanted from older to younger people, which have higher GDF11 levels, thrive.

• Letting aging just take it course is almost as risky as trying GDF11.

Side Effects of EXCESS GDF11

• Dyspnea (shortness of breath)

Proper dosing of GDF11• By looking at papers detailing circulating levels of GDF11, I

was able to impute this formula for starting dosage in ng:

((your weight in lbs.)/200) * (((Age – 18)/100)* 2094)/90

• Two parts to GDF11 dosing strategy:

1. Down regulation – Amy Wagers of the HSCI did say that GDF11 is a “reset thing” and this typically takes 4 to 6 weeks.

2. Maintenance dose – a tiny, typically .1 ng, dose of GDF11 once/twice per week

Down regulation dosing • Dosing is all over the map. It’s tied to age, weight, exercise and even

current or ex smoker. Unlikely we’ll find a universal dosing formula any time soon.

• To avoid side effects, we now look at daily trending of blood pressure, pulse, HRV and reaction time. If GDF11 dose is correct, blood pressure reaction time and pulse should go down. HRV should go up.

• If trending goes in the wrong direction, a week’s break is taken and then we restart at half the dose or less.

• Trending spreadsheets are a big improvement from the early days of GDF11 where patients were told to dose “just beneath the side effect level”. GDF11 dosing is far more nuanced than that…

GDF11 controversies….• For every paper that says GDF11

declines with age, there is another paper that says GDF11 increases with age

• Researchers have had difficulty replicating HSCI results

WHY???

GDF11 controversies….WHY??• GDF11 is very hard to measure

• it’s found in only nano quantities (circulating levels about 2000 ng at age 40)

• tests are easily confused by myostatin which has levels 500x that of GDF11

• 90% of GDF11 is stored in platelets, not serum.

• GDF11 probably increases in serum, but not platelets.

• Dosing window of GDF11 is very tight as we have seen.

• Too much GDF11 is surely detrimental

Future directions…● I’ll be the first to admit my study is not a

randomized double-blind, placebo-controlled  trial.

● However, it does suggest GDF11’s efficacy in many key areas, including cardiac, neural, skin elasticity, etc.

● And we do now a reasonably safe GDF11 starting dosing formula as well as trending biomarker spreadsheets to prevent overdose and increase efficacy.

● For the non do it yourselfers in the audience, we will be opening a GDF11 clinic in the Bahamas in October. And we will do a placebo controlled study at the clinic.
