CRC Mission
At CRC, we’re fighting cancer, hoping youwon’t need to. Our extensive cancer research isfunded in large part by the citizens and busi-nesses of central Missouri who, like yourselves,desire a solution to this lethal disease.
Because of the extensive cancer research that
is being performed all over the world, includ-ing right here in Columbia, we are going waybeyond poisoning, radiating and cutting outcancer cells. We are developing procedures tostop cancer before it ever starts and techniquesto treat it that avoid destroying good cells.

We conduct and sponsor research to reduce cancer deaths by earlier detection,
novel therapies, and identification of environmental and nutritional components
that reduce tumor initiation.

Dear Friends,The year 2006 brought the
Cancer Research Center successas well as inspiration. Ourprostate cancer project is stillprogressing, and we are soproud of our results. Dr. Abe,Alison, and Dr. Rob have beenworking diligently in the lab,as well as at the computer.They’ve been furiously prepar-ing grant applications tosecure funding that will ensurethe completion of the prostatecancer project. The support ofour individual donors, UnitedWays, Order of the EasternStar, Missouri State EmployeeCharitable Campaign, andCosmopolitan Club (just toname a few) have funded theresearch to this point. Wecould not have come this farwithout you. Be sure to readabout our prostate cancerresearch on pages 24–25.
A secondary mission for us atCRC is to train scientists.Internships are an importantpart of the education processfor those who pursue a futurein the field of research, and we
have had the pleasure of work-ing with some top-notch schol-ars. We put them to work inthe laboratory, and they leaveCRC as published scientists withsome very valuable experienceunder their belts. Be sure toread about the interns, theirexperiences, and our scholar-ship program on pages 18–21.
In September, we werereminded that cancer researchscholars come in all shapes andsizes. We were treated to avisit with a student, muchyounger than our typicalintern. After seeing two of hisgrandfathers battle cancer, 11-year-old Aaron Harris ofTuscumbia, Missouri, has decid-ed he wants to do somethingabout the disease and the dev-astation it causes. Read abouthis visit to the CRC laboratoryon page 39.
Cancer Research Center vol-unteers help us in so manyways, but especially in prepar-ing for the Jim KidwellMemorial Gala. In 2006 theyworked so diligently as wehonored our long-time friend,
Hartley Banks Jr. The level ofsupport at the event washeartwarming to all of us whoknew and appreciated Hartley.See highlights of the event onpages 27–38. Mark your calen-dar for Saturday, April 14,when we will again gather atthe Elks Lodge to raise fundsfor the Ray Freese MemorialFellowship.
We sometimes see the unex-pected. For example, did youknow pigs and gators can helpfight cancer? Thanks to ourgenerous friends at FarmPower Lawn and Leisure andapproximately 300 other gen-erous supporters, two pigs anda gator did just that. Readabout the first CRC Pig Roastand John Deere Gator Raffleon page 9 and learn who pur-chased the lucky ticket!

Meet Our StaffDr. Abe Eisenstark
is Research Director ofthe CRC. Previously, hewas Byler DistinguishedProfessor and Directorof the Division ofBiological Sciences,University of Missouri-Columbia. He hasserved as ProgramDirector of theMolecular Biology
Section, National Science Foundation,Washington, DC. His career includes years ofcollaborative research at other institutions,including universities at Copenhagen,Denmark; Leicester, England; Paris, France;Leiden, Netherlands; and Brookhaven NationalLaboratories. He is a native of Kansas City,MO. He received his BS, MS, and PhD inMicrobiology from the University of Illinois.He is married to the former Joan Ragsdell ofColumbia and is the father of three children.
Jack Bozarth isDirector ofDevelopment at theCancer ResearchCenter. He is responsi-ble for fundraising andpublic affairs. Jack isa retired Air Force offi-cer who was the PublicAffairs Officer for theCommissioner ofMilitary Affairs for the
State of Alaska. He is married to the formerAnn Freese of Columbia and has three chil-dren and two grandchildren. Born in KansasCity and raised in Raytown, Missouri, Jack is agraduate of Central Missouri State Universityand was awarded a MS degree by Embry-Riddle Aeronautical University. Jack is amember of the Columbia Elks Lodge No. 594,Missouri Aerie 2730 of the Fraternal Order ofEagles, and the Columbia Kiwanis Club.
Marnie Tutt isDirector of the CRC andhas been with theorganization for morethan 10 years. Sheserved as DevelopmentDirector from 1994 to1999, and was a mem-ber of CRC’s Board ofTrustees from 1999 to2001 while working atWilliam Woods
University. She has extensive experience in pub-lic relations, is a member of Columbia MetroRotary, and is active in many community organ-izations. Marnie was a 2003 recipient of the 40Under 40 in Columbia award. She earned a BFAdegree from William Woods. Marnie lives inFulton with her children Bailey and Brian.
Damon Young grad-uated from UniversityCity High School in St.Louis and earned adegree in BusinessAdministration fromthe University ofMissouri-Columbia. Hehas worked in a varietyof social services andnonprofits in case man-agement and direct
client care. Since joining the Cancer ResearchCenter in 2001, Damon has been a key memberof the support staff, maintaining the CRC website and database while gaining experience infundraising and development. Damon is mar-ried to Sharee and is the proud father of Dasha.

Meet Our StaffAlison Fea is the
Senior Technician andLaboratory Manager ofthe CRC. A New Jerseynative, Alison attendedStephens College inColumbia prior to join-ing the CRC in 2002.During her time as anundergraduate, Alisonserved as the ChemistryLaboratory Assistant
for the college’s Natural Science Departmentand served as an officer in several honor soci-eties. The summer of her junior year she waselected to Sigma Xi, the Scientific ResearchSociety, for her HIV-1 research work when shewas an infectious disease intern in Galveston,TX. Alison graduated Magna Cum Laude fromStephens in 2002, with a BS in Biology and aminor in Chemistry. She is the eldest of three;her family now resides in Kansas City, MO.
Dustin Newmanjoined the CRC in fall2001, working part timeas a lab assistant duringcollege and full timeafter graduation. Heearned his BS in Biologyfrom Columbia Collegein 2005. Dustin is a 1997graduate of HickmanHigh School. Upon grad-uation from high school,
he earned an associate degree in welding andmachining technology at Ranken College.Dustin has worked as a machinist for GeneralElectric, and also as a welder. In summer 2006,he accepted a position as a microbiologist witha chemical company in St. Louis.
Rachel Leist concep-tualizes and designsmarketing and advertis-ing materials for theCRC. She co-owns aweb and print designcompany calledMayeCreate Design andhas worked with theCRC since 2004. Sheearned a BA in Businessand Professional
Communications from Central MethodistUniversity and is enrolled in the MBA programat Stephens College in Columbia. As a minis-ter’s daughter, Rachel grew up all over. Shegraduated from high school in Dexter, MO. Herparents now live in Kearney, MO. She lives inJefferson City with Mike and their four dogs.
Dr. Robert A.Kazmierczak is theRaymond W. Freese fellow at the CRC. Hereceived his BS inBacteriology at theUniversity of Wisconsin,studying regulation ofVitamin B12 productionby Salmonella enterica(serovar Typhimurium)and interning atUpjohn Pharmaceuti-cals. He earned his MS
and PhD in Microbio-logy from the Universityof Illinois, during which time he held an NIHCell and Molecular Biology Training Grant fel-lowship to research chronic viral infections. Helater studied at Washington University in St.Louis. At the CRC, he is conducting research tocharacterize and develop therapeuticSalmonella enterica as a treatment for prostatecancer.

Thanks to our generous supporters andraffle participants, our John Deere GatorRaffle was a huge success! The 250 people inattendance feasted on an old-fashionedMissouri barbecue and a good time was hadby all. Jim Cherrington won the Gator andCRC raised more than $5,000.

After lunch the CRCstaff presented a short program on our con-tinuing efforts to fight cancer. The staffgave our distinguished visitors a tour of thelaboratory and discussed the results ofefforts to develop a Salmonella bacteriawhich will kill PC-3 (prostate cancer cell line)cells. This very promising area of researchhas great potential to treat not onlyprostate cancer but other types as well.
The Order of the Eastern Star is the largestfraternal organization in the world to whichboth men and women may belong.Worldwide, there are approximately onemillion members under the General GrandChapter.
While this is an order composed of peopleof deep spiritual convictions, it is open to allfaiths, except no faith. The personal welfareof members is vital to all of those in theEastern Star and it is considered a privilegeto help another member whenever they can.
Concrete evidence of the charitable nature
of the organization as it affects others is themillions of dollars that have been raisedwithin the membership to support national,jurisdictional, and local charities. Throughthis fraternal service to humanity, the Orderstrives to build a better life for all.
In this spirit of service to humanity, theOrder of the Eastern Star donated a checkfor more than $25,000 to the CancerResearch Center. The check was presentedat the 2006 Jim Kidwell Memorial Gala.Please see a picture of the presentation onpage 40.

CRC Community OutreachEducation Mission

The Cancer Research Center, in conjunction with its research mission, engages students inorder to enhance their scientific backgrounds. There are several support categories, depend-ing upon other academic arrangements.
1. Students can gain experience at CRC while earning undergraduate credit at University ofMissouri-Columbia or other local colleges. Students are assigned a semester project. For eachhour of credit, the student is expected to be in the laboratory approximately three hours perweek. A term paper is submitted at the end of the semester. Since it may be necessary to havea car, CRC provides $100 compensation for each of four months per semester.
For additional compensation, students are encouraged to apply for scholarship funds. UMCstudents should contact Linda Blockus, Tucker Hall. Preference is given to students who haveapplied for scholarship funds.
If an arrangement is made to have the student work additional hours in the laboratory,the student is compensated $8 per hour for the additional hours.
2. If a student desires an intern arrangement without college credit, CRC has limited schol-arship funds for this. The student should submit a resume to the CRC’s Research Director,including a list of college science courses. The stipend for this type of internship is usually$800 for the semester. Again, preference is given to students who have applied for scholar-ship funds.
3. As grants are obtained, CRC may employ technicians who have backgrounds inMicrobiology and Biochemistry.
4. CRC also welcomes graduate students to perform rotations in our laboratory. In suchcases, since it will be necessary to have a car, CRC will provide $100 compensation for each offour months per semester.

Shelly Erickson, a CRCDirector’s Scholar, graduated in2006 from the University ofMissouri-Columbia in Biologyand is now pursuing a gradu-ate degree in Microbiology atSt. Louis University in St. Louis,MO.
She graduated from FortZumwalt North High School.
Shelly has completed twosemesters of research on thecompetitive fitness of archivedSalmonella. In the summer of2005, she won a prestigiousundergraduate research intern-ship from UMC.
Shelly is senior author of ascientific paper that has beensubmitted for publication.
Rula Khreis, a CRC Director’sScholar, is a senior at theUniversity of Missouri-Columbia.She is working on a major inBiochemistry and a minor inEnglish. After graduation, Rulaplans to attend medical school;currently, she is interviewing atvarious institutions.
She was a 2003 honors gradu-
ate of Rock Bridge High School.Rula worked nine hours per
week at the CRC on ourprostate cancer project. Duringher sophomore year at UMC,she presented a term paper andreceived three hours of academ-ic credit. Rula worked under thesupervision of Dr. Abe Eisen-stark, CRC research director.
Crystal Dreyer, a CRCDirector’s Scholar, graduatedfrom the University of Missouri-Columbia in 2006 with a degreein Biology. She is now in a grad-uate program at the Universityof Chicago, studying to be aphysician assistant.
Crystal is a 2002 graduate ofFort Zumwalt High School in

Breca Starr Tracy, who willreceive her PhD in Biochemistryand Molecular Biology from theUniversity of Oklahoma in May2007, was awarded a CRCDirector’s Scholarship in 2000.Breca graduated from StephensCollege in Columbia, MO, with adegree in Biology and HealthSciences.
Breca is a 1998 graduate ofAda High School in Ada, OK.
Breca is the senior author of ascientific paper that described herCRC research. She is the firstauthor on several other articles,the most recent in the Journal ofBiochemistry. Breca also has pre-sented current research findingsat an international symposium.

Cathy Miller, spent two yearsworking at the CRC and is now anassistant professor of Microbiolo-gy at Iowa State University.
She graduated from JeffersonCity High in 1984. After earning aBS in Microbiology at theUniversity of Missouri-Columbia in1992, Cathy went on to graduateschool in the Department of
Microbiology and MolecularGenetics at the UM MedicalSchool. Her CRC research resultedin a published scientific paper.
Cathy’s achievements includea postdoctoral fellowship atHarvard Medical School, thepublication of several first-author manuscripts and thereceipt of many grants.

Swathi Bhaskaran, a CRCDirector’s Scholar, is a freshmanat the University of Missouri-Columbia majoring in ChemicalEngineering. The Hickman Highgraduate was salutatorian of theClass of 2006. Swathi has beenawarded the prestigiousSiemschens Advanced PlacementScholarship, as well as an
Engineering Dean’s Scholarshipfrom UMC.
At the CRC during the summerof 2004, Swasthi studied changesin archived Salmonella bacteria,investigating whether a specificgene accounts for survival underharsh conditions. She learnedbasic lab techniques and how toanalyze and record her findings.
Emily Schupp is a junior atthe University of Missouri-Columbia pursuing a degree inMicrobiology.
While working in the CancerResearch Center laboratory, shestudied various phage (virus)samples to determine theirrelationship to the mutationsobserved in our archived
Salmonella collection. Emily’swork was supervised by DustinNewman.

Current Research
Prostate cancer is associated with aging andoccurs in a latent or clinical form in 30–40%of men by age 40–50, increasing substan-
tially in men over 50 years of age. Prostate can-cer mortality in men is now second only to lungcancer, and as life expectancy increases so willthe number of men affected. While hormonetherapy is often effective during the initialstages of cancer development, most men devel-op hormone-resistant cancer within two yearsafter onset of the disease. This is why the firstline cancer treatment—chemotherapy utilizingtaxol—has met with only partial success. Manypatients develop taxol resistance and alternativetreatment and new approaches are needed toarrest tumor cell growth.
Recently, genetically engineered non-toxicSalmonella have been utilized as a weaponagainst prostate cancer. These strains prefer toattach and invade tumor cells to normal cells atratios of >1000:1 while not causing the toxiceffects normally seen in systemic Salmonellainfections. A number of these strains(VNP20009, AR-1) have inhibited or eliminatedprostate cancer and breast cancer tissues grownin mice. However, clinical studies using thesestrains have shown unacceptable toxicity levelsin human patients.
Development of the TherapeuticSalmonella Strain CRC2631
The Cancer Research Center has a uniqueopportunity to achieve the balance necessary toturn Salmonella into a dedicated but gentlecancer killer in human patients. Our laboratoryhas thousands of Salmonella samples that differfrom each other genetically but retain the natu-ral tumor-targeting ability described above.These strains have been allowed to age formore than 40 years and have naturally mel-lowed into non-toxic variants. We have focusedour efforts into direct genetic engineering of
our most promising candidates. These changeswere made to meet the following objectives: 1)Reduce the natural toxicity of our therapeuticSalmonella further; 2) Allow survival of theSalmonella in the cancer tissue only, not the cir-culatory system; and 3) Encourage killing ofcancer tissue by Salmonella. We are excitedabout our genetically engineered strainCRC2631 (a genetic descendant of our 1674Salmonella described last year) that has beenshown to attach, invade, and specifically destroythe energy-producing mitochondria in PC-3Mcancer cells (Slides 1, 3). Mitochondria areessential for supplying power to cancer cells;their destruction leads to death of the cancercells. CRC2631 kills PC-3M prostate cancer cellswithout breaking the cells open and spillingtheir contents into the surrounding area. Suchspilling is a very strong immune system triggerthat produces undesirable toxic side effects in

cancer patients. All the strains previously testedin human patients broke cancer cells open afterfive hours, and this may have been responsiblethe unacceptable toxicity levels during therapy(Slide 2).
We are currently raising mice that naturallydevelop prostate tumors to confirm CRC2631non-toxicity and measure how much Salmonellais required to destroy solid prostate tumors inmice. Upon completion of the mouse toxicityexperiments, we plan to move forward to clini-cal testing.
How Does Therapeutic Salmonella Target and Kill Tumors?
In addition to development of new therapeu-tic Salmonella strains, we are focusing ourefforts to discover answers to the followingquestions: 1) Why is Salmonella specificallyattracted to cancer cells? 2) How does ourSalmonella attach to and invade solid tumors?3) How does our Salmonella kill cancer cells bydegrading mitochondria? Answering thesequestions will tell us what functions are impor-tant during each stage of Salmonella-based can-cer therapy and enable us to improve our thera-peutic strains.
One way to determine which characteristicsof Salmonella are important for cancer therapyis to measure which genes are activated whenSalmonella is allowed to attach, invade, and kill
cells. Once we know which genes Salmonellaneeds to exert its anti-cancer effects, we canamplify these genetic functions while reducingor eliminating other functions that might causetoxic side effects. We are currently workingwith Drs. Michael McClelland and SteffenPorwollik at the Sidney Kimmel Cancer Centerto identify which CRC2631 Salmonella genes areturned on when co-incubated with PC-3Mprostate cancer cells. We expect this work to becompleted in the first half of 2007.
In order to track Salmonella attachment andinvasion, we have inserted a gene into ourSalmonella strain that makes it glow red orgreen under a fluorescent microscope. Thisallows us to easily quantify how quicklySalmonella can attach to any particular cancercell. We have identified that our CRC2631strain attaches to PC-3M prostate cancer cellswithin the first 20 minutes of co-incubation,demonstrating that our therapeutic Salmonellacan quickly recognize and attach to cancer cells.(Slides 4, 5) These red fluorescent strains canalso be used to identify where Salmonella pref-erentially invade and persist within solidtumors, characteristics that may determine howfast a particular Salmonella strain destroys can-cer within the body. We are collaborating withProfessor Cheryl Nickerson of Arizona StateUniversity who has experience at growing largenumbers of three-dimensional solid tumors inreactors that simulate the physiological condi-tions found in the body. Observing the way ourCRC2631 and other therapeutic Salmonella can-didates attach, invade, persist, and destroythese solid tumors will tell us how Salmonellaperforms these functions and allow us to quick-ly screen out superior cancer-killing strains.
Thanks to our generous supporters and a lotof hard work, our Salmonella strains are well ontheir way to becoming a potent yet gentletreatment for prostate cancer.

This has been a significantyear of cancer researchprogress at CRC!
We are fully aware thatour generous supporters arean important factor in thisprogress. In January 2006,Dr. Robert Kazmierczak wasappointed as the RaymondFreese Fellow and has madeimportant contributions toour research program.
With your continued confi-dence in our program, weare optimistic that 2007 willbe a year in which CRC willdevelop a novel approach toextending the lives of cancervictims. Progress may bereviewed in this annualreport. In particular, ourexciting accomplishment hasbeen to genetically engineerthe bacterial organismSalmonella typhimurium sothat it will maximally destroytumor cells without harmingnormal cells. A dozengenetic changes were madeto rid the attacking bacteriaof its toxic properties, whileenhancing its tumor destruc-tion properties.
Although we have succeed-ed in demonstrating prostatetumor cell destruction by spe-cial microscope observations,we will soon extend our stud-ies to include experimentswith mice. During 2007, stepone will be to verify that ourtherapeutic bacteria are harm-
less to mice. This will beaccomplished within the nextfew weeks. Step two will beto inject mice that have vari-ous stages of prostate cancer,and to follow the reduction oftumor size, and hopefully,complete cure.

Five undergraduatestudents will be part of ourresearch team in 2007. Thescientific progress of ourrecent trainees is included inthis annual report.

At CRC we know thatscientists come in allsizes. Encouraging
young people, even when it issomething as simple as adviceon a school science project,can touch off the career of acancer researcher or Nobelprize winner. You just neverknow. CRC is committed tothe community, and whenAaron Harris wanted to tour aworking lab and discuss hisscience project, we jumped atthe opportunity. Everyone inthe lab had a great time visit-ing with an eager young sci-entist and his family. Keep upthe good work, Aaron!
CRC has a rather strangegarden to tend. Whilemost toil in the soil to
grow blooms and veggies, wegrow cancer cells. Every daythey are checked, fed, andtended. Depending on thestrain, a week or so later weharvest the garden, set aside
some “seed” for the next crop,and use the cells to test andperfect our cancer-hungrySalmonella strain. Some gar-den in soil—we garden in bot-tles, but the rewards are stilljust a rich.

The Ray Freese MemorialFellowship in CancerResearch. This fellowship sup-ports a postdoctoral scientistwho engages in basic researchat CRC in the development of aprofessional career findingways to reduce cancer deaths.
The Pamela Dunn Legacyof Life. Due to her dedicationand contributions to cancerresearch, the Cancer ResearchCenter established the Dr.Pamela Dunn Legacy of Hope inher honor. The legacy is usedto recognize individuals, busi-nesses and foundations whodemonstrate a tangible interestin supporting the CRC.
Income from these funds pro-vide for instruments, booksand support for our scientistswho receive fellowships,
stipends and other financialhelp based on their trainingand experience.

Raymond Freese, a local builder, developer, and president ofBoone County Lumber Company, was a member of the CRCBoard of Trustees for 21 years, beginning in 1973. He workedtirelessly to secure funding and support for various CRC projectsand was especially devoted to the CRC Gala. Ray lost his sisterand several close friends to cancer and decided to do what hecould to fight this lethal disease. Unfortunately, he lost his ownbattle with cancer in 1994. After his death, the CRC Board ofTrustees set up the Raymond W. Freese Memorial Fellowship.This fellowship is awarded to promising postdoctoral scientistsfor an initial one-year appointment, with reappointment foradditional years based on performance. The stipend is compara-ble to those awarded by the Health Sciences Center at theUniversity of Missouri-Columbia. The major research project isprostate tumor therapy with genetically altered Salmonella.Applicants are expected to have (1) expertise in molecular genet-
ics of bacteria, (2) willingness to develop a mouse testing model, (3) skills in writing grant proposalsand manuscripts, (4) skills in presenting research results at national meetings, and (5) skills to developprojects independently and creatively.
