Received for publication December 29, 1952 T he in vitro responses of rings, strips or segments of arteries to various drugs and physical changes have been the subject of numerous investigations since the pioneer work of 0. B. Meyer (1905). In mos cases the source of arteries has been slaughterhouse animals. However, in the course of studies in this labora- tory on the relation between meta olism and contraction of smooth muscles it was found that spirally cut strips of rabbit thoracic aorta were admirably suited for both qualitative and quant tative investigations of the effects of drugs on arterial smooth muscle. The present paper is concerned primarily with the response of such strips to various drugs, and also with their general characteris- tics as pharmacological test objects. METHODS. For each experiment a rabbit weighing 2.5 to 3.5 kgm. was rapidly decapitated. The descending thoracic aorta was removed and placed in a Petri dish containing Krebs- bicarbonate solution (Krebs and Hetiseleit, 1932) at room temperature. Excess fat and connective tissue were trimmed off. The whole length of aorta was then cut along a close spiral. During the cutting, which was done with a small, sharp-pointed scissors, the uncut portion of the aorta, held gently between the thumb and fingers of the operator’s free hand, was gradually rotated and moved forward toward the scissors in such a manner as to permit a continuous spiral incision. The resulting strip was usually about 0.4 mm. thick, 2 mm. wide and 20 cm. long, and contained smooth muscle fibers (the circular fibers of the intact aorta) oriented at about 15 degrees relative to its long axis. From this strip shorter strips of 2 to 4 cm. length (uns retched) were cut for use in experiments. During the whole pro- cedure of preparing the strips, the tissue w as kept moistened with Krebs solution. The strips were mounted in muscle chambers of 20 ml. wor ing volume with the aid of stainless steel S-hooks. The muscle bath assembly, maintained at 37.5#{176},was of the type previously described (Furchgott, 1950). Replacement of solutio in a muscle chamber was accomplished by the over-flow principle, with enough fresh solution from the reservoir flask to dilute any previously added drug at least a thousand fold. The solution was Krebs- bicarbonate solution containing 0.01 3! glucose. Through this solution 95 per cent 02-5 per cent CO2 was bubbled, both in the muscle chambers and reservoir flask, giving a pH of 7.4. The isotonic levers were of the ink-w iting gravity-type, adjusted to give 9 fold am- plification, and counterweighted to exert 4 gm. tension on the strips. In many experiments in order to mini ize the ag in response due to the small amount of friction between writing points and kymograph paper, the levers were subjected to continuous vibration of small amplitude by means of a small vibrating motor connected to the iron stand supporting them. ‘This investigation was supported by a rant from the Life Insurance Medical Research Fund.
