Monarch® Total RNA Miniprep KitNEB is adding a new “universal” RNA purification kit to its Monarch Nucleic Acid Purification (NAP) product portfolio. This kit can be used to purify high-quality total RNA from multiple sample types, including blood, cells, tissues, and tough-to-lyse samples (bacteria, yeast, plant, etc.).
The Monarch total RNA miniprep kit enables the purification of up to 100 μg of total RNA from a wide variety of biological samples, including blood, cultured cells and mammalian tissues. Additionally, bacteria, yeast and plant samples can be processed with additional steps to enhance lysis for these tough-to-lyse samples.
The kit includes components for DNA removal (i.e., gDNA removal columns and DNase I) and Proteinase K for digestion of tissues and blood samples. The kit also provides a reagent for the storage and preservation of samples that will not be processed immediately.

Which contaminants does Acclaro detect? In DNA samples, Acclaro detects the following contaminants: proteins, phenol, Guanidine HCl and Guanidine Isothiocyanate. In protein samples (direct A280 measurements) Acclaro can detect nucleic acids and phenol. More contaminants will be offered in future software updates.
How does Acclaro help you troubleshoot? When the user taps the information icon on the measurement screen, a popup box will report the sample issue detected and offer possible causes and solutions. If the user wants to “Learn More” they will be guided through a cascade of information, from fundamentals to the specifics. This is best explained with an example. When Acclaro detects a low A260/A280 result, a popup box will inform that the ratio is outside acceptable limits for pure DNA sample and that this could be due to a poor blank or a contaminant. The user will then see information on common contaminants that can affect this ratio, and other resources starting with an animation that explains “What is a purity ratio.” If protein is the more likely contaminant, you can look at spectra of DNA/ protein samples and how increasing protein levels can affect the purity ratios of a DNA sample.

NEBNext® multiplex small RNA library prep set for Illumina®
One of the fastest growing areas of biological research is regulatory small RNA structure, processing and function. Next generation sequencing (NGS) is the method of choice for studying the variety and expression of small RNAs. The novel NEBNext® Small RNA workflow has been optimized to minimize adaptor-dimers while producing high-yield, high-diversity libraries. The NEBNext® Small RNA Library Prep Set for Illumina® includes adaptors and multiplex primers with 12 indices, to enable multi-plexing. Additional index primers for multiplexing are also available.

A common problem in NGS methods for small RNAs is contamination from adapter-dimer artifacts, because these artifacts are very close in size to the small RNA library elements. To address this problem, the NEBNext small RNA library prep kits use specially engineered RNA ligases, optimized workflows and novel technology to dramatically reduce the formation of adapter-dimer artifacts during library construction. Since the workflow uses total RNA as the starting material, it is beneficial to perform a final size selection step on the amplified libraries. NEB has validated size selection methods using Pippin Prep 3% gel cassettes for size selection of NEBNext Small RNA libraries.
Pippin Prep
The Pippin Prep facilitates library construction for the most popular NGS platforms, and is recommended by Illumina and Ion Torrent for certain workflows. The platform features the ability to collect narrow and even fragment distributions, as well as the flexibility to collect wide ranges of fragments with minimal effort. Target sizes or ranges of sizes are entered in software, and fractions are collected in buffer. Up to 5 samples per gel cassette may be run, with no possibility of cross contamination.

Nanopore sequencers give optimal results with BluePippin size selection
The Oxford Nanopore team has been speaking recently about their use of the BluePippin auto-mated size selection system for optimizing the read length obtained from nanopore sequencers. As seen with PacBio, the other long-read platform, single-molecule sequencers tend to produce reads as long as the fragments fed to them. Naturally, users interested in maximizing the read lengths of these systems want to feed them only the longest possible fragments. The simplest and most effective way to do that is with high-pass sizing, or selecting all DNA fragments longer than a certain size threshold during the sample prep process.
For the MinIon and PromethIon sequencers from Oxford Nanopore, the company recommends BluePippin sizing for various protocols. BluePippin is programmed to eliminate shorter fragments; one example of outcomes shows a whopping 255 Kb read from an E. coli experiment. There’s a similar rationale for recommending BluePippin for de novo whole genome assembly with the MinIon system.

Nitrocellulose sandwiches from Cell Signaling Technology® are pre-packaged sandwiches consisting of a precut nitrocellulose membrane and 2 sheets of 3 mm thick blotting paper. Intended to provide convenience and time savings, they are also validated for use in CST's Western Blot protocol and are the preferred membrane used by Cell Signaling Technology scientists.
Nitrocellulose sandwiches are ideal for binding proteins of all molecular weights; the 0.2 μm pore size ensures high retention of low molecular weight proteins, even below 20 kDa. Nitrocellulose sandwiches are suitable for most common transfer methods including tank, semi-dry, and vacuum blotting. This product is also suitable for colorimetric, fluorescent, and chemiluminescent detection. For chemiluminescent detection, CST recommends HRP based systems. Background: The programmed cell death 1 protein (PD-1, PDCD1, CD279) is a member of the CD28 family of immunoreceptors that regulate T cell activation and immune responses. The PD-1 protein contains an extracellular Ig V domain, a transmembrane domain, and a cytoplasmic tail that includes an immunoreceptor tyrosine-based inhibitory motif (ITIM) and an immunoreceptor tyrosine-based switch motif (ITSM). PD-1 is activated by the cell surface ligands PD-L1 and PD-L2. Upon activation, PD-1 ITIM and ITSM phosphorylation leads to the recruitment of the protein tyrosine phosphatases SHP-1 and SHP-2, which suppress TCR signaling. In addition to activated T-cells, PD-1 is expressed in activated B-cells and monocytes, although its function in these cell types has not been fully characterized (8). The PD-1 pathway plays an important role in immune tolerance; however, research studies show that cancer cells often adopt this pathway to escape immune surveillance. Consequently, blockade of PD-1 and its ligands is proving to be a sound strategy for neoplastic intervention.

Background: OX40 (TNFRSF4, CD134) is a member of the tumor necrosis factor (TNF) receptor super-family that regulates T cell activity and immune responses. The OX40 protein contains four cysteine rich domains, a transmembrane domain, and a cytoplasmic tail containing a QEE motif. OX40 is primarily expressed on activated CD4+ and CD8+ T-cells, while the OX40 ligand (OX40L, TNFSF4, CD252) is predominantly expressed on activated antigen presenting cells. The engagement of OX40 with OX40L leads to the recruitment of TNF receptor-associated factors (TRAFs) and results in the formation of a TCR-independent signaling complex. Background: Poliovirus receptor (PVR, CD155) is an immunoglobulin-like, transmembrane glycoprotein originally described as a mediator of poliovirus attachment to cells and later identified as important in adherens junction formation. Also known as nectin-like 5 (Necl-5), PVR binds nectin-3 and interacts with integrin αvβ3 and PDGFR to regulate integrin clustering and focal contact formation at the leading edge ofmigrating cells. Research studies demonstrate that PVR and nectin-3 regulate contact inhibition during cell motility and proliferation in transformed 3T3 cells. Additional research indicates that PVR (CD155, Necl-5) expression may play a role in invasiveness of lung adenocarcinoma. Background: Poliovirus receptor (PVR, CD155) is an immunoglobulin-like, transmembrane glycoprotein originally described as a mediator of poliovirus attachment to cells and later identified as important in adherens junction formation. Also known as nectin-like 5 (Necl-5), PVR binds nectin-3 and interacts with integrin αvβ3 and PDGFR to regulate integrin clustering and focal contact formation at the leading edge of migrating cells (1,2). Research studies demonstrate that PVR and nectin-3 regulate contact inhibition during cell motility and proliferation in transformed 3T3 cells (3). Additional research indicates that PVR (CD155, Necl-5) expression may play a role in invasiveness of lung adenocarcinoma (4,5). In the immune system, CD155 plays a role in natural killer (NK) cell-mediated cytotoxicity (6).

Source/Purification: Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Asn188 of human CD155 protein.

Haematology FISH probes for multiple myeloma
Multiple myeloma (MM) is a plasma cell neoplasm, characterised by the accumulation of clonal plasma cells in the bone marrow and by very complex cytogenetic and molecular genetic aberrations. The modal chromosome number in newly diagnosed symptomatic patients is usually either hyperdiploid, with multiple trisomies of chromosomes 3, 5, 7, 9, 11, 15, 19 and 21, or hypodiploid with immunoglobulin heavy chain (Ig) translocations. At disease progression, several genetic progression factors have been identified as deletions of 13q, deletions of 17p and deletion of 1p and/or amplification of 1q. Cytogenetic abnormalities are detected by conventional cytogenetics in about one third of the cases, but FISH increases the proportion of chromosomal abnormality detection to >90%.

We recommend performing all liquid handling operations in DNA/RNA UV-Cleaner Cabinet to protect samples from contamination.
Additional processing of the sample is often needed before NA extraction. Select appropriate instruments for sample preparation, depending on the sample type. For mixing and resuspen-sion choose vortex V-1 Plus or shaker MPS-1, depending on the amount of samples. For sedimentation choose centrifuge Microspin 12. Multi-motion programmable rotator Multi Bio RS-24 is ideal for mixing.
Cell lysis is a destruction of cell mem-branes with the lysis buffer. Depending on the protocol it is performed at the room temperature or higher. To ensure the most comprehensive lysis of your samples choose our Dry Blocks or Thermo-Shakers that provide one of the highest temperature uniformity and accuracy on the market.
Lysis procedure is followed by binding of the nucleic acid to magnetic particles. The magnetic rack is used to immobilize magnetic beads with the bound nucleic acid and then, protein and contaminants are removed by washing with wash buffers.Biosan Magnetic rack contains a high-performance magnet allowing instantaneous and most complete bead collection from the solution and providing the most extensive NA recovery.
